The discovery of human remains from the Lauricocha cave in the Central Andean highlands in the 1960’s provided the first direct evidence for human presence in the high altitude Andes. The skeletons found at this site were ascribed to the Early to Middle Holocene and represented the oldest known population of Western South America, and thus were used in several studies addressing the early population history of the continent. However, later excavations at Lauricocha led to doubts regarding the antiquity of the site. Here, we provide new dating, craniometric, and genetic evidence for this iconic site. We obtained new radiocarbon dates, generated complete mitochondrial genomes and nuclear SNP data from five individuals, and re-analyzed the human remains of Lauricocha to revise the initial morphological and craniometric analysis conducted in the 1960’s. We show that Lauricocha was indeed occupied in the Early to Middle Holocene but the temporal spread of dates we obtained from the human remains show that they do not qualify as a single contemporaneous population. However, the genetic results from five of the individuals fall within the spectrum of genetic diversity observed in pre-Columbian and modern Native Central American populations. 相似文献

Nostoc muscorum and Spirulina platensis were grown under phosphate deficiency in order to investigate the role of internal phosphate pool and activity of alkaline phosphatase on poly-β-hydroxybutyrate (PHB) accumulation. PHB accumulation in N. muscorum increased to 22.7% of dry weight (dw) after 4 day of phosphate deficiency, while the internal phosphate pool reduced to the level of 0.02 μM mg dw−1 at a maximum APase activity of 2.57 nM PNP mg dw−1 h−1. In contrary, S. platensis depicted maxima of 1.39 nM PNP mg dw−1 h−1 on day 30 of incubation, which was about 2 fold lower than the observed value of N. muscorum. PHB content in S. platensis remained low even after prolonged phosphate starvation, and a rise only up to 3.5% of dw was recorded on day 60 of phosphate deficiency. Supplementation of NADPH exogenously to S. platensis cultures grown under phosphate deficiency favoured PHB accumulation in 10, 20 and 30 days old cultures, but not in the cultures grown under phosphate deficiency for 60 days. The possible role of phosphate limitation on PHB accumulation is discussed. 相似文献

We have observed restriction of the murine sarcoma growth by therapeutic intervention of neem leaf glycoprotein (NLGP). In order to evaluate the mechanism of tumor growth restriction, here, we have analyzed tumor microenvironment (TME) from sarcoma bearing mice with NLGP therapy (NLGP-TME, in comparison to PBS-TME). Analysis of cytokine milieu within TME revealed IL-10, TGFβ, IL-6 rich type 2 characters was switched to type 1 microenvironment with dominance of IFNγ secretion within NLGP-TME. Proportion of CD8+ T cells was increased within NLGP-TME and these T cells were protected from TME-induced anergy by NLGP, as indicated by higher expression of pNFAT and inhibit related downstream signaling. Moreover, low expression of FasR+ cells within CD8+ T cell population denotes prevention from activation induced cell death. Using CFSE as a probe, better migration of T cells was noted within TME from NLGP treated mice than PBS cohort. CD8+ T cells isolated from NLGP-TME exhibited greater cytotoxicity to sarcoma cells in vitro and these cells show higher expression of cytotoxicity related molecules, perforin and granzyme B. Adoptive transfer of NLGP-TME exposed T cells, but not PBS-TME exposed cells in mice, is able to significantly inhibit the growth of sarcoma in vivo. Such tumor growth inhibition by NLGP-TME exposed T cells was not observed when mice were depleted for CD8+ T cells. Accumulated evidences strongly suggest NLGP mediated normalization of TME allows T cells to perform optimally to inhibit the tumor growth. 相似文献

This study compares the PHB synthase activity of Nostoc muscorum, a N(2)-fixing cyanobacterium under control (grown in usual BG-11 medium), nitrogen (N) and phosphorus (P) deprivation and chemoheterotrophic conditions. Specific activity of PHB synthase did not depict significant variations in the latter three types of cultures, except for the control one, where a significantly lower activity was recorded. PHB synthase activity was detected only in the soluble fractions of both the control as well as cells incubated under chemoheterotrophic conditions. A K(m) of 80.2 microM DL-beta-hydroxybutyryl-CoA and V(max) of 197.5 nmol thiobenzoate (TNB) mg protein(-1)min(-1) were observed for the enzyme. PHB synthase remained insensitive to acetyl-CoA, ATP, NADP, NADPH supplementation under in vitro condition. Addition of acetyl phosphate was found to activate the enzyme and the level of activation was dependent on the concentration of acetyl phosphate supplementation. Inhibition of PHB synthase in 2,3-butanedione supplemented cultures and reactivation following acetyl phosphate addition proved the post-translational control of acetyl phosphate over PHB synthase. 相似文献

Poly-beta-hydroxybutyrate (PHB) accumulation in the unicellular cyanobacterium, Synechocystis sp. PCC 6803, was studied under various cultural and nutritional conditions. Under controlled condition, cells harvested at the stationary phase of growth depicted maximum accumulation of PHB, i.e., 4.5% (w/w of dry cells) as compared to lag (1.8%) or logarithmic (2.9%) phases of cultures. A temperature range of 28-32 degrees C and pH between 7.5 and 8.5 were preferred for PHB accumulation. Cells cultivated under regular light-dark cycles accumulated more PHB (4.5%) than those grown under continuous illumination (2.4%). Nitrogen and phosphorus starvation stimulated PHB accumulation up to the tune of 9.5 and 11% (w/w of dry cells), respectively. Synechocystis cells pre-grown in glucose (0.1%)-supplemented BG-11 medium when subjected to P-deficiency in presence of acetate (0.4%), PHB accumulation was boosted up to 29% (w/w of dry cells), the value almost 6-fold higher with respect to photoautotrophic condition. Abrus agglutinin peptide fractions obtained from 10 kD molecular weight cut off membrane permeate (10 kMP), was shown to have selective antiproliferative activity on several tumor cell lines with induction of apoptosis through mitochondrial pathway. The present study was designed to evaluate acute general toxicity and in vivo therapeutic effectiveness of 10 kMPP in Dalton's lymphoma (DL) mice model. The acute toxicity like body weight, peripheral blood cell count, lympho-hematological and biochemical parameters remained unaffected with 1mg/kg body weight and lower of 10 kMPP. The in vivo antitumor study indicated that there were 27%, 58.5% and 84.5% reduction in DL cell survival in 100, 200 and 500 microg/kg body weight of 10 kMPP, respectively. Analysis of the growth inhibitory mechanism in DL cells revealed nuclear fragmentation and condensation with appearance of the sub G0/G1 peak is indicative of apoptosis. Further, the Western blotting showed apoptosis was mediated by reduction in ratio of Bcl-2 and Bax protein expression, and activation of caspase-3 through release of cytochrome-c in DL cells. Kaplan-Meier survival analysis showed an effective antitumor response (53 ILS%) with dose of 500 microg/kg body weight. Our result showed that the novel peptides present in Abrus agglutinin possess potent antitumor properties which need to be further explored. 相似文献

Nonproportional hazards often arise in survival analysis, as is evident in the data from the International Non-Hodgkin's Lymphoma Prognostic Factors Project. A tree-based method to handle such survival data is developed for the assessment and estimation of time-dependent regression effects under a Cox-type model. The tree method approximates the time-varying regression effects as piecewise constants and is designed to estimate change points in the regression parameters. A fast algorithm that relies on maximized score statistics is used in recursive segmentation of the time axis. Following the segmentation, a pruning algorithm with optimal properties similar to those of classification and regression trees (CART) is used to determine a sparse segmentation. Bootstrap resampling is used in correcting for overoptimism due to split point optimization. The piecewise constant model is often more suitable for clinical interpretation of the regression parameters than the more flexible spline models. The utility of the algorithm is shown on the lymphoma data, where we further develop the published International Risk Index into a time-varying risk index for non-Hodgkin's lymphoma. 相似文献
