Aim There remains some uncertainty concerning the causes of extinctions of Madagascar’s megafauna. One hypothesis is that they were caused by over‐hunting by humans. A second hypothesis is that their extinction was caused by both environmental change and hunting. This paper systematically addresses the second hypothesis through examination of two new pollen records from south‐eastern Madagascar alongside other published records across the island. Location South‐eastern Madagascar. Methods We reconstructed past vegetation and fire dynamics over the past 6000 years at two sites in south‐eastern Madagascar (Ste‐Luce) using fossil pollen and charcoal contained in sedimentary sequences. We investigated drivers of vegetation changes and how these, in turn, influenced faunal species in the south‐east, using published climatic, archaeological and faunal records. Further, we also used published records to provide a synthesis of environmental changes on the whole island. Results Vegetation reconstructions indicate that the mosaic vegetation in the region of Ste‐Luce was highly dynamic in response to climatic changes. The open woodland, surrounding the littoral forest, transformed into an ericoid grassland between c. 5800 and 5200 cal. yr bp , possibly in response to a moderate drought recorded during this period. The littoral forest was more stable between c. 5100 and 1000 cal. yr bp , with only some minor compositional changes c. 2800 cal. yr bp and between c. 1900 and 1000 cal. yr bp . Significant forest decline, however, is observed at c. 950 cal. yr bp , coinciding with a drought and a marine surge. A comparison of these results with a synthesis of published vegetation records across the island shows asynchronous vegetation changes in response to various droughts during the Holocene, except for the 950 cal. yr bp drought event, with evidence of widespread vegetation transformations and fires across the island. Main conclusions Pronounced climatic desiccation between 1200 and 700 cal. yr bp may have been the slow driver framing and triggering vegetation transformations and decline in megafaunal populations. In addition, hunting by drought‐impacted human inhabitants and competition with newly introduced cattle would have amplified the impacts on megafaunal populations, leading to numerous extinctions in this period. 相似文献

The treatment of soils contaminated with organic compounds, such as polycyclic aromatic hydrocarbons (PAHs), by attrition produced large amounts of highly concentrated attrition sludge (PAH – attrition concentrate – PAC). This paper studied the performance of an oxidation process using potassium permanganate (KMnO4) to degrade PAHs that were initially present in attrition concentrates. The influence of operating conditions (temperature, concentration of KMnO4 and reaction time) was studied, and these parameters were optimized using a response surface methodology (RSM). The results showed that the temperature and the reaction time had a significant and positive effect on the degradation of PAHs for the experimental domain studied (temperature between 20 and 60°C and reaction time between 1 and 7 h). The interaction between the temperature and the concentration of KMnO4 significantly influenced the degradation of the PAHs. The temperature and the concentration of KMnO4 were the main parameters that influenced the degradation of both phenanthrene (Phe) and benzo [a] pyrene (BaP). For benzo [a] anthracene (BaA), the temperature was the most influential factor. According to our results, the optimal conditions were defined as [KMnO4] = 0.4 M for 5.5 h at 60°C. These optimal conditions led to degradations of 42.9%, 40.8%, 41.0% and 46.0% of the total PAHs, Phe, BaA and BaP, respectively. 相似文献

In the last few years, the interest in sulfonamides has expanded owing to their broad spectrum of biological activities. Their flexible structure turns them into amazing candidates to replace old drugs or develop modern multi-target agents. In this study, a series of new sulfonamides ( sul1-5 ) was evaluated, in vitro, for the antibacterial, cytotoxic and genotoxic effects. The antibacterial activity was investigated against 12 clinical and 4 reference strains. Cytotoxic activity was carried out by the brine shrimp bioassay and the genotoxicity was assessed in the Ames test. An interesting antibacterial activity was showed especially against Gram negative strains. The inhibition zones varied between 15 and 30 mm, and the Minimum Inhibitory Concentrations (MIC's) values between 0.5 and 256 μg/ml. No antibacterial activity was shown with S. aureus isolates. Only Sul1 and Sul4 were active against P. aeruginosa. Compounds Sul1 and Sul2 showed a significant cytotoxicity with LC50 equal to 18.29 and 18 μg/ml respectively, and a genotoxic effect against TA100 and TA1535 Salmonella strains. Only compounds Sul3 , Sul4 and Sul5 with an interesting antibacterial activity, no cytotoxicity and no genotoxic effects, could be exploited against resistant pathogens as new drugs. 相似文献

Research on mate choice has primarily focused on preferences for quality indicators, assuming that all individuals show consensus about who is the most attractive. However, in some species, mating preferences seem largely individual-specific, suggesting that they might target genetic or behavioral compatibility. Few studies have quantified the fitness consequences of allowing versus preventing such idiosyncratic mate choice. Here, we report on an experiment that controls for variation in overall partner quality and show that zebra finch (Taeniopygia guttata) pairs that resulted from free mate choice achieved a 37% higher reproductive success than pairs that were forced to mate. Cross-fostering of freshly laid eggs showed that embryo mortality (before hatching) primarily depended on the identity of the genetic parents, whereas offspring mortality during the rearing period depended on foster-parent identity. Therefore, preventing mate choice should lead to an increase in embryo mortality if mate choice targets genetic compatibility (for embryo viability), and to an increase in offspring mortality if mate choice targets behavioral compatibility (for better rearing). We found that pairs from both treatments showed equal rates of embryo mortality, but chosen pairs were better at raising offspring. These results thus support the behavioral, but not the genetic, compatibility hypothesis. Further exploratory analyses reveal several differences in behavior and fitness components between “free-choice” and “forced” pairs. 相似文献

The protonation states of the two active‐site lysines (Lys69 and Lys235) of PBP 6 of Escherichia coli were explored to understand the active site chemistry of this enzyme. Each lysine was individually mutated to cysteine, and the resultant two mutant proteins were purified to homogeneity. Each protein was denatured, and its cysteine was chemically modified to produce an S‐aminoethylated cysteine (γ‐thialysine) residue. Following renaturation, the evaluation of the kinetics of the dd ‐carboxypeptidase activity of PBP 6 as a function of pH was found consistent with one lysine in its free‐base (Lys69) and the other in the protonated state (Lys235) for optimal catalysis. The experimental estimates for their pKa values were compared with the pKa values calculated computationally, using molecular‐dynamics simulations and a thermodynamic cycle. Study of the γ‐thialysine69 showed that lysine at position 69 influenced the basic limb of catalysis, consistent with the fact that the two lysine side chains are in proximity to each other in the active site. Based on these observations, a reaction sequence for PBP 6 is proposed, wherein protonated Lys235 serves as the electrostatic substrate anchor and Lys69 as the conduit for protons in the course of the acylation and deacylation half‐reactions. UV-visible and dichroic spectrum analysis and electron microscopy have been used to characterize a new amphotericin B (AmB) lipid formulation prepared by a solvent displacement process. The composition was dimyristoylphosphatidylcholine (DMPC) and dimyristoylphosphatidylglycerol (DMPG) in molar ratio DMPC/DMPG/AmB 7:3:5, a similar composition to that of Abelcet®. Although the latter has a “ribbon-like” structure, our process gave a thin disc-like structure. Analysis of circular dichroism (CD) and UV-visible spectra of formulations containing different percentages of AmB revealed that a minimum of AmB self-association was observed with 7:3:5 molar ratio. Varying the lipid ratio (DMPC/DMPG) while maintaining the fixed ratio of AmB yielded similar results when DMPC was in excess (DMPC/DMPG from 10:0 to 6:4). However, when the ratio was between 5:5 to 3:7, AmB self-aggregation increased. For compositions rich in DMPG (2:8 and 0:10), inversion of the CD spectrum was observed. The influence of the lipid composition on the morphology of the complex was also evident in electron microscopy. DMPC/DMPG/AmB (10:0:5) gave large unfracturable lamellae. The presence of DMPG shortened the lamellae, which often appeared as disc-like structures. AmB content, the presence of DMPG and the preparation process all contribute to generating these original structures with particular CD spectra. 相似文献

Indole-3-acetic acid (IAA) is the most common, naturally occurring phytohormone that regulates cell division, differentiation, and senescence in plants. The capacity to synthesize IAA is also widespread among plant-associated bacterial and fungal species, which may use IAA as an effector molecule to define their relationships with plants or to coordinate their physiological behavior through cell-cell communication. Fungi, including many species that do not entertain a plant-associated life style, are also able to synthesize IAA, but the physiological role of IAA in these fungi has largely remained enigmatic. Interestingly, in this context, growth of the budding yeast Saccharomyces cerevisiae is sensitive to extracellular IAA. Here, we use a combination of various genetic approaches including chemical-genetic profiling, SAturated Transposon Analysis in Yeast (SATAY), and genetic epistasis analyses to identify the mode-of-action by which IAA inhibits growth in yeast. Surprisingly, these analyses pinpointed the target of rapamycin complex 1 (TORC1), a central regulator of eukaryotic cell growth, as the major growth-limiting target of IAA. Our biochemical analyses further demonstrate that IAA inhibits TORC1 both in vivo and in vitro. Intriguingly, we also show that yeast cells are able to synthesize IAA and specifically accumulate IAA upon entry into stationary phase. Our data therefore suggest that IAA contributes to proper entry of yeast cells into a quiescent state by acting as a metabolic inhibitor of TORC1. 相似文献
