A new “marker” protein for astrocytes

A monoclonal antibody (Mab J1-31) has been produced by using human brain homogenate as immunogen in mouse. Double-label immunofluorescence microscopy on cryostat sections of human, rabbit and rat brain, reveals staining of cells that are also stained with antiserum to glial fibrillary acidic protein (GFAP, a commonly used marker protein for astrocytes). However, there is no decrease in staining due to Mab J1-31 in sections incubated in antiserum to GFAP prior to incubation with the J1-31 ascites fluid. Immunoprecipitation of aqueous and detergent extracts of brain homogenate gives a single band at 30K by SDS PAGE followed by autoradiography. Immunoelectron microscopy shows that the J1-31 antigen is associated with the cytoskeleton. Thus, the Mab J1-31 recognizes a new protein present in GFAP positive cells (astrocytes) in the brain.     

Investigations on the variability of haptoglobin, transferrin and Gc polymorphisms in Assam, India

Ten different population groups of Assam - Brahmins, Kalitas, Kaibartas, Rajbanshis, Muslims, Ahoms, Chutias, Kacharis, Karbis and Sandwals - have been typed for haptoglobin and for transferrin (Tf) and Gc subtype polymorphisms. Tf and Gc allele subtype frequencies show a considerable inter-population heterogeneity. From genetic distance analysis it appears that the populations under study form some distinct clusters, which can be explained by the historical and ethnic affiliations of these populations. Especially the distribution of Gc subtype alleles reveals some Mongoloid admixture among Assamese populations, which is reflected by the presence of Gc1A8 alleles in them.

A structural characterization of gap junctions isolated from mouse liver

Mouse liver gap junctions have been isolated by using an anionic detergent, n-dodecanoyl sarcosine, in combination with non-ionic polyoxyethylene ethers (Brij 35 and Brij 58) and (W-1) detergents. Purified gap junctions are obtained in a sucrose step gradient containing 1-o-n-octyl-beta-D-glucopyranoside. This procedure is aimed at minimizing proteolysis. The junctions thus isolated have a hexagonal lattice of connexons with a lattice constant of 7.6-8.4 nm. Presence of a major Mr 26,000 gap junctional protein has been confirmed by SDS-PAGE.
