A putative integrative function of the striatum was evaluated through the study of the electrical activity of sensory responsive caudate neurones. Both nervous (radial nerve) and auditory stimulations were delivered in order to characterize populations of neurones affected by peripheral stimuli; the units were previously activated by iontophoretic glutamate. On these units the iontophoretic ejection of ACh and DA was tested. Experimental results demonstrated a prevalent excitatory effect of ACh, while DA appeared to exert a drastic decrease on firing rate. A comparison between peripheral stimuli and chemical substances was made. The result of such study showed a most important action of the neurotransmitters employed. The activity of caudate units following single shock activation was also explored. This investigation underlined a certain degree of facilitatory influence of ACh; DA, on the contrary, had the tendency to exert a marked inhibitory action. The results are interpreted in view of the striatal peculiar position on cortico-striato-thalamo-cortical circuit. An integrative function of basal ganglia on sensori-motor activity of the cortex is postulated and the importance of ACh and DA is emphasized. 相似文献

Phylogeography and conservation genetics of the hellbender salamander (Cryptobranchus alleganiensis)
We investigated hellbender phylogeography through phylogenetic analyses of individuals sampled from 16 locations throughout
their range in the eastern United States. Analyses were conducted on concatenated cytochrome-oxidase I (COI), cytochrome-b (Cytb) and NADH dehydrogenase subunit 4 (ND4) mtDNA sequence, totaling 2160 nucleotides. Hellbender haplotypes differed by 0.1
to 5.8% maximum likelihood (ML) corrected sequence divergence. Phylogenetic analyses reveal that hellbenders are separated
into 8 reciprocally monophyletic populations or clades differentiated by a minimum of 0.7 to 5.4% sequence divergence, each
of which constitutes a separate Management Unit (MU). High among population divergence and reciprocal monophyly suggest that
female-mediated gene flow is severely restricted or non-existent among each MU. Hellbenders are currently divided into two
subspecies, Cryptobranchus alleganiensis alleganiensis and C. a. bishopi based on morphological characters. The phylogenetic analyses presented here strongly indicate that these subspecies are paraphyletic.
Management priorities for the hellbender should be reconsidered in light of these new molecular data. Results from Bayesian
rooting indicate the root of the hellbender mtDNA tree lies on the branch leading to hellbender haplotypes from the Current,
Eleven Point and New Rivers. The rooted tree suggests that a common ancestor in the southern Ozarks and/or southern Appalachians
gave rise to northern hellbender populations, consistent with a Pleistocene refuge hypothesis. 相似文献

Three patients carrying an isodicentric (idic) Y chromosome associated with a mosaic 45,X cell line were studied using molecular
techniques. Genotype-phenotype correlations suggested an effect of the 45,X cell line on sexual differentiation. A relationship
was established between instability of the idic(Y) chromosome and localization of the breakpoint on Yq, and between azoospermia
and deletion of interval 6 on Yq.

Background aimsAdoptive immunotherapy with the use of chimeric antigen receptor (CAR)-engineered T cells specific for CD19 has shown promising results for the treatment of B-cell lymphomas and leukemia. This therapy involves the transduction of autologous T cells with a viral vector and the subsequent cell expansion. We describe a new, simplified method to produce anti-CD19-CAR T cells.MethodsT cells were isolated from peripheral blood mononuclear cell (PBMC) with anti-CD3/anti-CD28 paramagnetic beads. After 2 days, the T cells were added to culture bags pre-treated with RetroNectin and loaded with the retroviral anti-CD19 CAR vector. The cells, beads and vector were incubated for 24 h, and a second transduction was then performed. No spinoculation was used. Cells were then expanded for an additional 9 days.ResultsThe method was validated through the use of two PBMC products from a patient with B-cell chronic lymphoblastic leukemia and one PBMC product from a healthy subject. The two PBMC products from the patient with B-cell chronic lymphoblastic leukemia contained 11.4% and 12.9% T cells. The manufacturing process led to final products highly enriched in T cells with a mean CD3+ cell content of 98%, a mean expansion of 10.6-fold and a mean transduction efficiency of 68%. Similar results were obtained from the PBMCs of the first four patients with acute lymphoblastic leukemia treated at our institution.ConclusionsWe developed a simplified, semi-closed system for the initial selection, activation, transduction and expansion of T cells with the use of anti-CD3/anti-CD28 beads and bags to produce autologous anti-CD19 CAR–transduced T cells to support an ongoing clinical trial. 相似文献

The effect of substantia nigra stimulation on duodeno-jejunal intraluminal pressure and electrical activity was analysed in the cat. An inhibitory influence was noted to occur on both the intestinal pressure and the electrical response activity (ERA): the effect seemed to be in correlation with the stimulus parameters employed. After bilateral vagotomy, the inhibition remained substantially unchanged. The possibility of the existence of a sympathetic pathway mediating the effect is emphasized. 相似文献
