Brassinolide (2α,3α,22α, 23α-tetrahydroxy-24α-methyl-B-homo-7-oxa-5α-cholestan-6-one), a novel plant growth-promoting steroid isolated from rape pollen, and its hitherto unknown 22β, 23β-isomer were synthesized from a C-24 epimeric 60:40 mixture of 22-dehydrocampesterol (24α-methyl) and brassicasterol (24β-methyl) from oysters. The method of synthesis favored the formation of the 22β, 23β-isomer by better than 4:1. Comparative plant growth-promoting capabilities of brassinolide, both natural and synthetic, and its three side chain -glycolic isomers in the bean second internode bioassay showed that the natural and synthetic brassinolides were equally active and caused splitting of the internode at the 0.1 μg level. The least active was the 22β,23β-isomer of brassinolide. The isomers with the 22α, 23α and 24β, and the 22β, 23β and 24β configurations were highly active and were required at about 10 times the concentration of brassinolide to cause the same physiological response. In the bean first internode bioassay, an auxin-induced growth test system which employs isolated bean plant segments, the isomer with 22β, 23β and 24β configuration caused a greater response than brassinolide. Similarities between DDT and pyrethroid insecticides have led to widespread concern that cross-resistance between them might limit the usefulness of the latter. Both types of insecticide have similarities in chemical structure, both have a negative temperature coefficient (ie. they are more active at lower temperatures), both act as neurotoxins on sodium channels, and both produce the twin effects of knockdown and kill. As discussed by Tom Miller (see pages S8-S12) there is firm evidence for Pyrethroid resistance in some species of medical and veterinary importance - especially in the horn fly, Haemotobia irritans. But in the case of anopheline mosquitoes, the evidence for pyrethroid resistance is much less strong. As Colin Malcolm explains here, a critical analysis of available data indicates that true physiological resistance of anophelines to pyrethroids is much less widespread than previous commentaries suggest. Moreover, the risk of cross-resistance between pyrethroids and DDT may have been over-emphasized, since different resistance mechanisms appear to be involved. 相似文献

Malaria during pregnancy is associated with poor birth outcomes, particularly low birth weight. Recently, monocyte infiltration into the placental intervillous space has been identified as a key risk factor for low birth weight. However, the malaria-induced chemokines involved in recruiting and activating placental monocytes have not been identified. In this study, we determined which chemokines are elevated during placental malaria infection and the association between chemokine expression and placental monocyte infiltration. Placental malaria infection was associated with elevations in mRNA expression of three beta chemokines, macrophage-inflammatory protein 1 (MIP-1) alpha (CCL3), monocyte chemoattractant protein 1 (MCP-1; CCL2), and I-309 (CCL1), and one alpha chemokine, IL-8 (CXCL8); all correlated with monocyte density in the placental intervillous space. Placental plasma concentrations of MIP-1 alpha and IL-8 were increased in women with placental malaria and were associated with placental monocyte infiltration. By immunohistochemistry, we localized placental chemokine production in malaria-infected placentas: some but not all hemozoin-laden maternal macrophages produced MIP-1 beta and MCP-1, and fetal stromal cells produced MCP-1. In sum, local placental production of chemokines is increased in malaria, and may be an important trigger for monocyte accumulation in the placenta. 相似文献

Zooxanthella symbioses are arguably the most important ecological interaction on coral reefs because they energetically subsidize the entire community, and enhance the calcification process that provides structure for all other organisms. While we have developed a detailed understanding of the diversity among and within the Symbiodinium clades, we currently lack a mechanistic explanation for which factors favoured zooxanthella invasion of the intracellular habitat in heterotrophic hosts, and for what molecular mechanisms permit residence within the cell. We propose two hypotheses that explain important evolutionary and ecological features of zooxanthella symbioses. The magnesium inhibition hypothesis (MIH) states that increases in the Mg/Ca ratio in sea water that occurred over the last 100 million years created a situation where Mg(2+) inhibited Ca(2+) transport to zooxanthellae. The MIH predicts, among other things, that the intracellular niche was invaded as a response to this abiotic stressor. The arrested phagosome hypothesis (APH) states that Symbiodinium spp. mimic host cell endosomal digestive machinery via the symbiosome to appear like digesting prey through perpetual release of zooxanthella-derived compounds. The APH represents a subtle but important distinction from previous hypotheses regarding interactions between symbiont and host at the cellular level. The APH predicts that symbionts tune rates of material release to match expectations of host cellular machinery. An outcome of the APH is that intra-host residence time becomes a vital parameter to consider. In maize (Zea mays L.) and pine (Pinus taeda L.) seedlings, cellulose microfibril impressions are present on freeze-fractured plasma membranes. It has been proposed that impressions of newly synthesized microfibrils are a record of the movement of terminal synthesizing complexes through the plasma membrane (Mueller and Brown, 1980, J. Cell Biol. 84, 315–326). The association of terminal complexes with the ends of microfibril impressions or with the ends of microfibrils torn through the membrane indicates the orientation of microfibril tips. Unidirectionally-oriented microfibril tips (all pointing in the same direction) are associated with the organized deposition of parallel arrays of microfibrils. Multidirectionally-oriented microfibril tips were observed in a cell in which microfibril deposition was unusually disorganized. Microfibril patterns around pit fields are asymmetric and resemble flow patterns. Unidirectionally-oriented tears are associated with these microfibrils. Although microfibril orientations are deflected around pit fields, the main axis of microfibril orientation is maintained across the surface of the cell. The hypothesis is proposed that the interaction of a flowing plasma membrane with microfibril synthesizing complexes in the plane of the membrane may result in unidirectional deposition and asymmetric microfibril impressions around pit fields.Some of this work has been published in preliminary form (Brown 1979) 相似文献
