The growth of wheat (Triticum aestivum L., cv. Zhnitsa) seedlings and calluses infected with spores of common bunt causal agent Tilletia caries (DC) Tul. was studied. Inoculation with the pathogenic fungus enhanced both in vivo and in vitro growth due to cell division activation and cell expansion. These growth characteristics were also retained in wheat calluses infected with T. caries and grown on the hormone-free Murashige and Skoog nutrient medium. This implies the production of hormone-like substances by the fungus itself or by the infected plant. At the phase of fungal penetration into the plants, infection resulted in IAA accumulation in seedlings. Later, the IAA content reduced to a control level and the phytohormone balance was shifted toward cytokinins. Similar changes in the IAA and cytokinin levels were observed during early fungus development on wheat callus tissues. Such a sequence of events is supposed to be required for successful fungal penetration and localization in plant tissues and for the establishment of compatible interactions between the pathogen and the host plant.  相似文献

The influence of nitric oxide (NO) on the myelinated nerve fiber and the impact of modification of SHgroups of axon and myelin membrane proteins on the amplitude and propagation velocity of action potential (AP), amount of the membrane-bound calcium (Ca mb 2+ , viscosity of the axon membrane, and saturation factor of phospholipid fatty acids (Sf) of myelin have been investigated. We established that the decrease in the number of extracellular SH-groups in membrane proteins induced by p-chloromercuribenzoate (pCMB, 10?4 M), led to a decrease in the AP amplitude and a reversible desorption of Ca mb 2+ but did not affect the axolemma viscosity and Sf. Nitric oxide (NO) caused a decrease in the AP amplitude and propagation velocity, an increase in the axolemma viscosity and a decrease in Sf of myelin; it also induced a reversible desorption of Ca mb 2+ . Pretreatment of the nerve fiber with pCMB weakened the NO-induced desorption of Pretreatment of the nerve fiber with K+-channel blocker tetraethylammonium (10?2 M) completely abolished the NO-induced change in the amount of Ca mb 2+ . We suppose that NO-mediated changes in axolemma viscosity, Sf of myelin and desorption of Ca mb 2+ affect protein-lipid interactions in axolemma and myelin, which in their turn influence the propagation of AP.  相似文献

The ability of Bacillus subtilis Cohn and Bacillus thuringiensis Berliner to induce systemic resistance in wheat plants to the casual agent of Septoria nodorum Berk., blotch has been studied. It has been shown that strains of Bacillus ssp. that possess the capacity for endophytic survival have antagonistic activity against this pathogen in vitro. A reduction of the degree of Septoria nodorum blotch development on wheat leaves under the influence of Bacillus spp. was accompanied by the suppression of catalase activity, an increase in peroxidase activity and H2O2 content, and expression of defence related genes such us PR-1, PR-6, and PR-9. It has been shown that B. subtilis 26 D induces expression levels of wheat pathogenesis-related (PR) genes which marks a SA-dependent pathway of sustainable development and that B. thuringiensis V-5689 and V-6066 induces a JA/ET-dependent pathway. These results suggest that these strain Bacillus spp. promotes the formation of wheat plant resistance to S. nodorum through systemic activation of the plant defense system. The designed bacterial consortium formed a complex biological response in wheat plants infected phytopathogen.  相似文献

Photoprotective mechanisms of cyanobacteria are characterized by several features associated with the structure of their water-soluble antenna complexes–the phycobilisomes (PBs). During energy transfer from PBs to chlorophyll of photosystem reaction centers, the “energy funnel” principle is realized, which regulates energy flux due to the specialized interaction of the PBs core with a quenching molecule capable of effectively dissipating electron excitation energy into heat. The role of the quencher is performed by ketocarotenoid within the photoactive orange carotenoid protein (OCP), which is also a sensor for light flux. At a high level of insolation, OCP is reversibly photoactivated, and this is accompanied by a sig- nificant change in its structure and spectral characteristics. Such conformational changes open the possibility for pro- tein–protein interactions between OCP and the PBs core (i.e., activation of photoprotection mechanisms) or the fluores- cence recovery protein. Even though OCP was discovered in 1981, little was known about the conformation of its active form until recently, as well as about the properties of homologs of its N and C domains. Studies carried out during recent years have made a breakthrough in understanding of the structural-functional organization of OCP and have enabled discovery of new aspects of the regulation of photoprotection processes in cyanobacteria. This review focuses on aspects of protein–pro- tein interactions between the main participants of photoprotection reactions and on certain properties of representatives of newly discovered families of OCP homologs.  相似文献

Paleoecological changes during the development of Lake Izyubrinye Solontsi from the Solontsovskie (Shanduyskie) Lakes located in the midlands of the Eastern Sikhote-Alin have been reconstructed. Lake formation is related to landslides on the paleovolcano slopes. A complex study of the peat-bog section (botanical, diatom, spore-pollen, and radiocarbon analysis) allows reconstructing paleoenvironmental changes with high resolution. A considerable variability of lake and swamp environments highly responsive to climate changes in the late Holocene has been revealed. Peat accumulation began about 400014С BP. The main peat-forming plants were Sphagnum mosses and herbs, except for the period 2330?1530 14C BP (2360?1480 cal. BP), when a swamp overgrown by larch forests and predominantly woody peat accumulated. Forest ecosystems on the lake coasts were quite stable. The role of fir and broadleaved species increased in the composition of dark coniferous forests with Korean pine during the warm phases and the role of birch, in the cold phases; secondary forests occupied the low slopes during the last 1000 years. The spectrum of mutations in the low-density lipoprotein (LDL) receptor gene was studied in a sample of hypercholesterolemia patients of Caucasoid origin from the population of Russia. The examined patients were 45 to 49 years old and had the highest level of total serum cholesterol in this age group. Seven previously nondescribed mutations have been revealed in exon 9 (R410G; M412V) and in exon 12 (Y/Y576; N/N591; L605V; L605R; A612G). Twelve previously described mutations have been identified in exons 2 (C/C27), 5 (C261F; E240X), 6 (E288K), 8 (A391T), 9 (E418G; L432R; D433E), 11 (G/G549; E558K; L/L568), and 12 (G592E). Only one of these mutations was previously described in Russia in a clinical sample of patients with familial hypercholesterolemia. The spectrum of LDL receptor gene mutations in the population sample of patients with hypercholesterolemia significantly differs from the mutation spectrum in patients with familial hypercholesterolemia (clinical samples). Sequencing of the LDL receptor gene is a highly efficient method for identifying the markers of hypercholesterolemia predisposition in a population.  相似文献

We studied the effect of hydrogen peroxide on morphological characteristics and resistance of common wheat calluses (Triticum aestivum L.) to Tilletia caries Till. The induction of the defense response and morphogenesis in calluses depended on H2O2 concentration. A correlation was revealed between the elevated concentration of hydrogen peroxide in wheat calluses and high activity of oxalate oxidase in the cell wall. Administration of H2O2 into the callus culture medium was followed by rhizogenesis, induced the formation of dense regions, and inhibited fungal growth on calluses. Hydrogen peroxide at high concentrations was less potent in inhibiting the growth of fungi. A relationship was found between oxalate oxidase activity, H2O2 concentration, and morphogenetic and defense responses of calluses induced by exogenous hydrogen peroxide. These data suggest that the induction of H2O2 generation is one of the approaches to increase callus resistance.  相似文献

In order to characterize the molecular mechanisms of lymphoma formation in HIV-infected humans, a method of two-staged substractive cloning was used, which adequately detects genes whose expression is increased in cells of one lymphoma in comparison with another. Using this method, we determined the spectrum of genes whose expression was increased in centroblastic non-Hodgkin's lymphoma in comparison with immunoblastic non-Hodgkin's lymphoma. Several gene groups were distinguished in this spectrum; their probable involvement in lymphogenesis is discussed.  相似文献

The virological, morphological, molecular biological and immunochemical study of the infective agent isolated from the patient with the symptoms of atypical pneumonia, hospitalized in the infectious department of the clinical hospital in Blagoveshchensk, was carried out. Thus the fact of the appearance of SARS virus on the territory of Russia was proved. The isolated infective agent, identified as coronavirus strain CoD, was partly characterized and deposited to the virus collection of the Center of Special Laboratory Diagnostics and Treatment of Quarantine and Exotic Infectious Diseases.  相似文献
