The Lymantria obfuscata Walker (Lyob) multiple (M) nucleopolyhedrovirus (NPV) (LyobMNPV) has been isolated and successfully applied for the management of the Indian gypsy moth, L. obfuscata in Jammu and Kashmir (J&K), India. The present work aimed to investigate the variability of LyobMNPV isolates from six localities of J&K through molecular [amplification of the polyhedrin (polh), late expression factor-8 (lef-8) and late expression factor-9 (lef-9) genes] and biological (bioassays) characterization. To identify the position of LyobMNPV in the phylogenetic tree of baculoviruses, partial sequences of the polh, lef-8 and lef-9 genes were determined by using the DNA sequences within their coding regions by optimizing the polymerase chain reaction with degenerate primers. The sequence alignment revealed that LyobMNPV isolates exhibited seven, five and eleven single nucleotide polymorphic sites in the case of polh, lef-8 and lef-9, respectively. The phylogenetic analyses supported placing LyobMNPV with the Lymantria dispar L. MNPV (LdMNPV) isolates from different countries, and showed that it was more closely related to LdMNPV than to Lymantria xylina Swinhoe NPV and Lymantria monacha L. NPV. The contaminated diet plug bioassays using 2nd instar larvae indicated that the median lethal dose (LD50) and median survival time (ST50) of different isolates of LyobMNPV against L. obfuscata were lower than those of LdMNPV against L. dispar. LyobMNPV was more closely related to LdMNPV but its LD50 and ST50 were lower than those of LdMNPV. The study provides novel information on the position of LyobMNPV in the phylogenetic tree of baculoviruses and about biological and genetic variation of Lymantria species’ NPV isolates from different parts of the world.  相似文献

Heavy metals are among the major pollutants from anthropogenic inputs that reach mangrove ecosystem by urban and agricultural runoff, industrial effluents, boating, mining and other processes. To minimize the detrimental effects of heavy metal exposure and their accumulations, plants in general have evolved biological detoxification mechanisms, which include avoidance or exclusion, excretion and accumulation. To protect the cellular components from oxidative damage by heavy metal contamination, biological systems have developed enzymatic and non-enzymatic antioxidant mechanisms. Another detoxification mechanisms produced in plants are osmoprotectants, which are the compatible solutes which maintain a favourable water potential gradient and protect cellular structures from toxic ions. Besides these mechanisms, another heavy metal detoxification system in plants involves the chelation of metals by metal binding molecules like metallothioneins (MTs) and phytochelatins (PCs). To limit the heavy metal toxicity from mangrove ecosystem, it was found that phytoremediation is a most useful technology where in plants are used to remove pollutants from the environment and it is considered as a comparatively new, low-cost and highly promising technology for the remediation of heavy metal. Rhizofiltration, phytovolatilization, phytoextraction and phytostabilization are the important phytoremediation techniques. Among these phytoextraction and phytostabilization are found highly important in the case of mangroves and are promising means of phytoremediation.  相似文献

Many of the morphological features of animals are considered to be adaptations to the habitat that the animals utilize. The habitats utilized by birds vary, perhaps more than for any other group of vertebrates. Here, we study possible adaptations in the morphology of the skeletal elements of the hind limbs to the habitat of birds. Measurements of the lengths of the femur, tibiotarsus and tarsometatarsus of 323 bird species from 74 families are used together with body mass data, taken from the literature. The species are separated into six habitat groups on the basis of literature data on leg use. A discriminant analysis of the groups based on leg morphology shows that swimming birds, wading birds and ground living species are more easily identified than other birds. Furthermore, functional predictions are made for each group based on ecological and mechanical considerations. The groups were tested for deviation from the norm for all birds for three indices of size- and leg-length-independent measures of the bones and for a size-independent-index of leg length. Several of the groups deviate significantly from the norm for one or more of the indices used, suggesting habitat-related adaptations in the leg morphology of birds. The results indicate that stability is an important factor affecting the leg morphology of primarily long-legged birds. The femur seems to be more important than previously thought because several of the groups have high femur indices, suggesting a positive selection pressure on this bone. Clathrin depletion by ribonucleic acid interference (RNAi) impairs mitotic spindle stability and cytokinesis. Depletion of several clathrin-associated proteins affects centrosome integrity, suggesting a further cell cycle function for clathrin. In this paper, we report that RNAi depletion of CHC17 (clathrin heavy chain 17) clathrin, but not the CHC22 clathrin isoform, induced centrosome amplification and multipolar spindles. To stage clathrin function within the cell cycle, a cell line expressing SNAP-tagged clathrin light chains was generated. Acute clathrin inactivation by chemical dimerization of the SNAP-tag during S phase caused reduction of both clathrin and ch-TOG (colonic, hepatic tumor overexpressed gene) at metaphase centrosomes, which became fragmented. This was phenocopied by treatment with Aurora A kinase inhibitor, suggesting a centrosomal role for the Aurora A-dependent complex of clathrin, ch-TOG, and TACC3 (transforming acidic coiled-coil protein 3). Clathrin inactivation in S phase also reduced total cellular levels of ch-TOG by metaphase. Live-cell imaging showed dynamic clathrin recruitment during centrosome maturation. Therefore, we propose that clathrin promotes centrosome maturation by stabilizing the microtubule-binding protein ch-TOG, defining a novel role for the clathrin-ch-TOG-TACC3 complex.  相似文献

目的建立人结肠癌多药耐受性动物模型并初步探索其耐药机制。方法结合体内外诱导方法建立人结肠癌多药耐受性动物模型,利用VCR和CTX的肿瘤抑制实验评价其MDR特性;利用real-time PCR和West-ern blotting等方法分析其P-gp/MDR1和MRP1基因和蛋白的表达。结果肿瘤抑制实验结果显示,MDR和敏感型结肠癌模型的肿瘤生长速度差异不显著,MDR结肠癌动物模型对于VCR和CTX的耐药性均有较大程度的提高;表达分析结果显示,人结肠癌MDR动物模型的P-gp/MDR1表达水平有较大提高,而MRP1表达没有显著变化。结论人结肠癌多药耐受性动物模型具有较好的多药耐受性,其多药耐受性表型主要是由于P-gp/MDR1过量表达所导致。  相似文献

目的 分析物种差异对NAFLD模型复制的影响,探讨不同鼠种NAFLD形成及其机制.方法 长爪沙鼠、SD大鼠、ICR小鼠各20只,按种属随机分为对照组及模型组,对照组给予普通饲料,模型组给予高脂饲料.16周后,观察肝脏HE及Mallory三色染色病理变化,计算肝指数,检测血清血脂(CHO、TG、LDL-c、HDL-c)、肝功能(GOP、GPT)及肝组织中抗氧化酶(SOD、GSH-PX、CAT)活性及羟脯氨酸(Hyp)、丙二醛(MDA)、游离脂肪酸(FFA)水平.结果 与对照组比较,各模型组:沙鼠Hyp、CHO、TG、LDL-c、HDL-c、肝指数、GOP、GPT、MDA、FFA均升高,SOD、GSH-PX、CAT活性降低(P<0.05,P<0.01),肝脏出现纤维化;大鼠CHO、肝指数、GOP、GPT、FFA、SOD活性升高,MDA含量、GSH-PX、CAT活性降低(P <0.05,P<0.01),有局灶性脂肪肝炎;小鼠CHO、LDL-c、HDL-c、肝指数、CAT活性升高,MDA含量降低(P <0.05,P<0.01),肝脏病理正常.结论 三种动物在脂质代谢、肝功能、氧化应激等方面有显著的差异,并形成了不同的NAFLD模型:沙鼠形成伴高TG、CHO血症的肝纤维化模型、大鼠形成伴高CHO血症的局灶性脂肪肝炎模型、小鼠形成高胆固醇血症模型但肝脏未发生明显的病理改变.  相似文献

目的 探究生防细菌DS-R5施入丹参植株后根际和根表土壤细菌群落组成及多样性变化。方法 向丹参植株根部施入生防细菌DS-R5,以未施用细菌为对照组,分别采集根际和根表土壤样品提取总DNA,扩增样品总DNA的V3-V4区,采用Illumina MiSeq测序平台对PCR扩增产物进行双端测序分析,利用生物信息学分析解析丹参植株根际土壤和根表土壤细菌群落结构组成及多样性。结果 菌株DS-R5处理后增加了根际土壤细菌群落的多样性和丰度,降低了根表土壤细菌群落的多样性和丰度;高通量测序得到的根际和根表土壤的有效序列数量和OTU数量相比对照组均有所下降,根际土壤处理样品中微生物种类最丰富,根表土壤处理样品中微生物种类最少,根际土壤处理样品与根际土壤对照物种种类更接近;在门水平上,根际土壤处理样品相比对照变形菌门丰度下降,酸杆菌门丰度升高,根表土壤处理样品相比对照变形菌门和酸杆菌门丰度均升高,放线菌门丰度降低;在属水平上,根际土壤处理样品中鞘氨醇单胞菌属、芽胞杆菌属、慢生根瘤菌属相比根际土壤对照占比均有升高,根表土壤处理样品相比对照黄杆菌属和伯克菌属丰度下降,而土壤中的优势菌属根瘤菌属和芽胞杆菌属丰度升高。结论 丹参植株施用生防细菌DS-R5后,改变了根际土壤和根表土壤中微生物群落结构和多样性。  相似文献

通过对9个微卫星座位的扩增,研究了Z:ZCLA长爪沙鼠的遗传多态性。结果表明,Z:ZCLA长爪沙鼠在其中1个位点上只有一个等位基因,在其它位点上均有2~4个等位基因,平均等位基因数2.6个。平均杂合度0.4684,平均多态信息量0.4166,平均有效等位基因数2.1756。全群基因纯合度从0.1111~0.5555,平均0.3389,提示目前本群遗传多样性水平处于中度多态。  相似文献

研究森林可燃物燃烧释放的细颗粒物(PM2.5)的排放因子对于揭示森林火灾对大气和生态系统的影响至关重要,而水溶性离子是细颗粒物的重要化学成分,对颗粒物的形成具有重要意义。利用自主设计的生物质燃烧系统,模拟内蒙古大兴安岭5种典型乔木(蒙古栎、白桦、兴安落叶松、黑桦、山杨)的3种组成部分(树干、树枝、树皮)及其地表死可燃物(凋落物层、半腐殖质层、腐殖质层)以及3种典型灌木(平榛、二色胡枝子、兴安杜鹃)树枝燃烧,采用ISC1100离子色谱分析仪测定2种燃烧状态(阴燃和明燃)下PM2.5中水溶性离子(Na+、NH4+、K+、Mg2+、Ca2+、F-、Cl-、NO3-、NO2-、SO42-)的排放因子。结果表明:乔木所有组成部分及其地表死可燃物和灌木树枝燃烧所排放PM2.5中检测到的水溶性离子,阴燃以K+、Cl-和Na+为主要组分,明燃以K+、Cl-和SO42-为主要组分。不同燃烧状态下相同乔木树种及其地表死可燃物和相同灌木排放PM2.5中检测到的水溶性离子总量均存在显著差异。灌木树枝在阴燃期间PM2.5中水溶性无机离子的排放因子比明燃更高。乔木释放的PM2.5中阳离子与阴离子的比率为1.26,地表死可燃物为1.12,灌木为2.0,表明颗粒物呈碱性。内蒙古大兴安岭的森林大火不会通过释放水溶性离子导致生态系统的酸化。  相似文献

以宁夏盐池县荒漠草原人工柠条(Caragana intermedia)林为研究对象,分别选取柠条林冠下东侧(SE),冠下西侧(SW)及带间(Gap)为研究样地,从群落水平探讨柠条对冠下草本植物群落结构、物种多样性及功能群分布的影响。结果表明:(1)在3种微生境中均鉴定出12种植物,SW以蒙古冰草为优势种,SE以蒙古冰草和中亚白草为优势种,Gap则以蒙古冰草和牛枝子为优势种。(2)与Gap相比,SW和SE植物群落的平均高度分别增加了41.06%和81.75%,地上生物量分别增加了40.88%和38.73%。SW和SE中,禾本科植物地上生物量分别占地上总生物量的67.10%和58.40%,显著高于Gap (P<0.05)。(3)柠条冠层效应使得草本植物的物种丰富度指数增加,但Shannon-Winner指数、Simpson指数和Pielou指数显著差异(P>0.05),变化范围分别为1.620-1.756、0.701-0.730和0.775-0.878。(4)冗余分析表明:土壤温度、空气相对湿度及土壤有机碳是影响草本植物物种多样性及生物量的主要因子,解释量分别为42.70%,11.70%和8.80%。研究表明,柠条对冠下草本植物群落尤其是禾本科植物具有一定的保育作用,该效应的产生主要是由于柠条冠下微气候及土壤环境因子的改善为草本植物的生长发育提供了有利条件。柠条对草本植物的保育作用对荒漠草原生态环境的保护与恢复具有重要意义。  相似文献
