The inactivation of naturally occurring bacterial indicators and bacteriophages by thermal treatment of a dewatered sludge and raw sewage was studied. The sludge was heated at 80°C, and the sewage was heated at 60°C. In both cases phages were significantly more resistant to thermal inactivation than bacterial indicators, with the exception of spores of sulfite-reducing clostridia. Somatic coliphages and phages infecting Bacteroides fragilis were significantly more resistant than F-specific RNA phages. Similar trends were observed in sludge and sewage. The effects of thermal treatment on various phages belonging to the three groups mentioned above and on various enteroviruses added to sewage were also studied. The results revealed that the variability in the resistance of phages agreed with the data obtained with the naturally occurring populations and that the phages that were studied were more resistant to heat treatment than the enteroviruses that were studied. The phages survived significantly better than Salmonella choleraesuis, and the extents of inactivation indicated that naturally occurring bacteriophages can be used to monitor the inactivation of Escherichia coli and Salmonella.  相似文献

In order to provide a better understanding of the dynamics of phytoplankton in the coastal regions of high latitudes, a study was carried out to estimate the dynamics of carbon biomass of autotrophic and heterotrophic algal groups over the austral spring-summer 1997/1998 period. At a fixed station located in the central basin (Paso Ancho) of the Straits of Magellan (53°S), surface water samples were collected at least once a week from September 1997 (early spring) to March 1998 (late summer). Quantitative analysis of biomass of phytoplankton was estimated from geometric volumes, using non-linear equations, and converted to biomass. The pattern of chlorophyll a showed a strong temporal variability, with maximum values (mean 2.8 mg m−3) at the austral spring phytoplankton increase or bloom (October/November) and minimum values during early spring (September: <0.5 mg m−3) and summer (January/March: 0.5–1.0 mg m−3). During the spring bloom, diatoms made up to 90% of the total phytoplankton carbon (0.01–189 μg l−1), followed by a maximum of thecate dinoflagellates (0.08–34 μg l−1), and sporadic high biomass of phytoflagellates during summer. Heterotrophic algal groups such as Gymnodinium and Gyrodinium spp. dominated (70%, in the 5- to 25-μm size range) shortly before the main diatom bloom, and small peaks were observed within spring and early summer periods (0–0.4 μg l−1). Phytoflagellates dominated earlier (spring) with higher carbon biomass (8 μg l−1) and post-bloom periods (summer) when carbon biomass ranged between 1 and 4 μg l−1. The type strain Bacillus thuringiensis var. bolivia (serotype H63), isolated from the Bolivian high valleys, has been characterized at different levels. Its parasporal crystal has an unusual shape and it is composed of a protein of 155 kDa which shows two bands of 75 and 80 kDa after activation. Analysis by PCR shows the presence of cry1 genes, and amplification with specific primers gave products for cry1 E, cry1 D, cry4 A and cry4 B with sizes different to those expected. Immunoblotting tests showed positive reaction for Cry1 E, Cry3 A, Cry4 A and Cry11 A crystal proteins. The plasmid pattern revealed two large and two small plasmids. Toxicity tests were performed against 14 insects and a slight toxicity was found against Plutella xylotella and Trichoplusia ni.  相似文献

To determine the effects of 6-weeks of heavy-resistance training on physical fitness and serum hormone status in adolescents (range 14-16 years old) 19 male handball players were divided into two different groups: a handball training group (NST, n = 10), and a handball and heavy-resistance strength training group (ST, n = 9). A third group of 4 handball goalkeepers of similar age served as a control group (C, n = 4). After the 6-week training period, the ST group showed an improvement in maximal dynamic strength of the leg extensors (12.2%; P < 0.01) and the upper extremity muscles (23%; P < 0.01), while no changes were observed in the NST and C groups. Similar differences were observed in the maximal isometric unilateral leg extension forces. The height of the vertical jump increased in the NST group from 29.5 (SD 4) cm to 31.4 (SD 5) cm (P < 0.05) while no changes were observed in the ST and C groups. A significant increase was observed in the ST group in the velocity of the throwing test [from 71.7 (SD 7) km x h(-1) to 74.0 (SD 7) km x h(-1); P < 0.001] during the 6-week period while no changes were observed in the NST and C groups. During a submaximal endurance test running at 11 km x h(-1), a significant decrease in blood lactate concentration occurred in the NST group [from 3.3 (SD 0.9) mmol x l(-1) to 2.4 (SD 0.8) mmol x l(-1); P < 0.01] during the experiment, while no change was observed in the ST or C groups. Finally, a significant increase (P < 0.01) was noted in the testosterone:cortisol ratio in the C group, while the increase in the NST group approached statistical significance (P < 0.08) and no changes in this ratio occurred in the ST group. The present findings suggested that the addition of 6-weeks of heavy resistance training to the handball training resulted in gains in maximal strength and throwing velocity but it compromised gains in leg explosive force production and endurance running. The tendency for a compromised testosterone:cortisol ratio observed in the ST group could have been associated with a state of overreaching or overtraining.  相似文献

Nitrite has been found in previous research an inhibitor on anoxic phosphorus uptake in enhanced biological phosphorus removal systems (EBPR). However, the inhibiting nitrite concentration reported varied in a large range. This study investigates the nitrite inhibition on anoxic phosphorus uptake by using four different mixed cultures performing EBPR with pH considered an important factor. The results showed that the protonated species of nitrite, HNO(2) (or free nitrous acid, FNA), rather than nitrite, is likely the actual inhibitor on the anoxic phosphorus uptake, as revealed by the much stronger correlation of the phosphorus uptake rate with the FNA than with the nitrite concentration. All the four EBPR sludges showed decreased anoxic phosphorus uptake rates with increased FNA concentrations in the studied range of 0.002-0.02 mg HNO(2)-N/L. The phosphorus uptake by all four cultures was completely inhibited at 0.02 mg HNO(2)-N/L. Granular sludge appeared to be more tolerant to HNO(2) than flocular sludge likely due to its stronger resistance to the transfer of nitrite into the bacterial aggregates. Furthermore, denitrification by the phosphorus-accumulating organisms (PAOs) was also found to be inhibited by HNO(2). The denitrification rate decreased by approximately 40% when the FNA concentration was increased from 0.002 to 0.02 mg HNO(2)-N/L.  相似文献

Extracellular Yersinia spp. disarm the immune system by injecting the effector Yersinia outer proteins (Yops) into the target cell. Yop secretion is triggered by contact with eukaryotic cells or by Ca2+ chelation. Two proteins, YopN and LcrG, are known to be involved in Yop-secretion control. Here we describe TyeA, a third protein involved in the control of Yop release. Like YopN, TyeA is localized at the bacterial surface. A tyeA knock-out mutant secreted Yops in the presence of Ca2+ and in the absence of eukaryotic cells. Unlike a yopN null mutant, the tyeA mutant was defective for translocation of YopE and YopH, but not YopM, YopO and YopP, into eukaryotic cells. This is the first observation suggesting that Yop effectors can be divided into two sets for delivery into eukaryotic cells. TyeA was found to interact with the translocator YopD and with residues 242-293 of YopN. In contrast with a yopN null mutant, a yopNDelta248-272 mutant was also unable to translocate YopE and YopH. Our results suggest that TyeA forms part of the translocation-control apparatus together with YopD and YopN, and that the interaction of these proteins is required for selective translocation of Yops inside eukaryotic cells.  相似文献

We have carried out a Fourier transform infrared spectroscopic study of mitochondrial aspartate aminotransferase in the spectral region where phosphate monoesters give rise to absorption. Infrared spectra in the above-mentioned region are dominated by protein absorption. Yet, below 1020 cm-1 protein interferences are minor, permitting the detection of the band arising from the symmetric stretching of dianionic phosphate monoesters [T. Shimanouchi, M. Tsuboi, and Y. Kyogoku (1964) Adv. Chem. Phys. 8, 435-498]. The integrated intensity of this band in several enzyme forms (pyridoxal phosphate, pyridoxamine phosphate, and sodium borohydride-reduced, pyridoxyl phosphate form) does not change with pH in the range 5-9. This behavior contrasts that of free pyridoxal phosphate (PLP) and pyridoxamine phosphate (PMP) in solution, where the dependence of the same infrared band intensity with pH can be correlated to the known pK values for the 5'-phosphate ester in solution. The integrated intensity value of this infrared band for the PLP enzyme form before and after reduction with sodium borohydride is close to that given by free PLP at pH 8-9. These results are taken as evidence that in the active site of mitochondrial aspartate aminotransferase the 5'-phosphate group of PLP remains mostly dianionic even at a pH near 5. Thus, it is suggested that the chemical shift changes associated with pH titrations of various PLP forms reported in a previous 31P NMR study of this enzyme [M. E. Mattingly, J. R. Mattingly, and M. Martinez-Carrion (1982) J. Biol. Chem. 257, 8872] are due to the fact that the phosphorus chemical shift senses the O-P-O bond distortions induced by the ionization of a nearby residue. Since no chemical shift changes were observed in pH titrations of the PMP forms (lacking an ionizable internal aldimine) of this isozyme, the Schiff base between PLP and Lys-258 at the active site is the most likely candidate for the ionizing group influencing the phosphorus chemical shift in this enzyme.  相似文献

The temporal variability of size-fractioned autotrophic biomassat three depth levels (1, 8 and 25 m) was studied during thewinter-spring transition at two oceanographic stations in ConcepciónBay. Size spectra were obtained on eight occasions by two differentmethods: (i) determining the biomass of seven autotrophic sizefractions by in vivo fluorescence; and (ii) measuring the filamentlength of chain-forming diatoms through direct microscopy. Aclear vertical gradient of biomass was found in all profiles,with maximum values in the surface layer (1 and 8 m levels).Values of chlorophyll were on average 6.2 (range 1.08–25.67)times higher at 1 m than at 25 m, and 7.4 (range 1.15–26.83)times more at 8 m than at 25 m. On a temporal basis, total biomassincreased from low average values in winter (2.5 mg chl-a m–3)to high values in late spring (11.6 mg chl-a m–3). Duringthe whole sampling period (June 8-November 19), the nano- andnet-plankton (1.8–40 µm and 40–335 µmsize fractions respectively) were more abundant near the surface(1 and 8 m depth) than close to the bottom (25 m depth); however,the picoplankton fraction (<1.8 (µm) showed an inverserelationship, with a slight trend to increase near the bottomtoward spring. The highest absolute biomass was concentratedin the net-plankton fraction during the whole period and therelative importance of the picoplankton decreased from winter(6.50 and 15.5% for shallow and bottom levels) to spring (1.5and 10.3% for shallow and bottom levels). This relative effectis caused by the higher absolute values of biomass observedin the net-plankton fraction toward spring. These changing patternsshould have an impact in the size-composition and abundanceof higher trophic levels, mainly through grazing, in particularby modifying food availability to microfJagellates, ciliatesand filter-feeding zooplankton.  相似文献

A practical procedure for the preparation of chiral 2-S-substituted-2H-pyran-3(6H)-ones is described. According to the reaction conditions, 1,5-anhydro-2,3,4-tri-O-acetyl-D-threo-pent-1-enitol (1) reacted with thiophenol under Lewis acid catalysis to afford the polysubstitution product 1,5-anhydro-2,3,4-tri-S-phenyl-2,3,4-trithio-D,L-threo-pent-1-enitol (2) or phenyl 2,4-di-O-acetyl-3-deoxy-1-thio-alpha- and beta-D-glycero-pent-2-enopyranoside (3 and 4, respectively). The iodine-promoted addition of thiophenol or alpha-toluenethiol to 1 gave (2S)-2-phenylthio-2H-pyran-3(6H)-one (5) or its 2-benzylthio analogue 6, but these products showed low enantiomeric excesses (ee approximately 40-60%). However, dihydropyranone 5 with high optical purity (ee>94%) was successfully obtained by treatment of 4 with iodine in acetonitrile. On the other hand, it was established that the benzylthio group of 5 exerts high stereocontrol in reduction and cycloaddition reactions performed on the alpha,beta-unsaturated carbonyl system.  相似文献

The nuclear stage at which oocytes are cryopreserved influences further development ability and cryopreservation affects ultrastructure of both cumulus cells and the oocyte. In this work, we analyze the effects of vitrification at different nuclear and cytoplasmic maturation stages on the oocyte ultrastructure and developmental ability. Culture in TCM199 + PVA with roscovitine 25 M during 24h led to meiotic arrest (MA) in cumulus-oocyte complexes (COCs), while permissive in vitro maturation (IVM) was performed in TCM199, 10% FCS, FSH-LH and 17beta-estradiol for 24 h. Oocytes were vitrified using the open pulled straw method (OPS) with minor modifications. Fresh and vitrified/warmed COCs were fixed as immature, after IVM, after meiotic arrest (MA) and after MA + IVM. Vitrification combined with MA followed by IVM produced the highest rates of degeneration, regardless of the vitrification time. As a consequence, lower proportions of embryos cleaved in these groups, although differences were eliminated at the five-eight cell stage. Development rates up to day 8 were similar in all experimental groups, being significantly lower than those in fresh controls. Only oocytes vitrified after IVM were able to give blastociysts. The morphological alterations observed can be responsible for compromised development. More research is needed to explain the low survival rates of the bovine oocyte after vitrification and warming.  相似文献
