Plant pathogens are able to influence the behaviour and fitness of their vectors in such a way that changes in plant–pathogen–vector interactions can affect their transmission. Such influence can be direct or indirect, depending on whether it is mediated by the presence of the pathogen in the vector's body or by host changes as a consequence of pathogen infection. We report the effect that the persistently aphid‐transmitted Cucurbit aphid‐borne yellows virus (CABYV, Polerovirus) can induce on the alighting, settling and probing behaviour activities of its vector, the cotton aphid Aphis gossypii. Only minor direct changes on aphid feeding behaviour were observed when viruliferous aphids fed on non‐infected plants. However, the feeding behaviour of non‐viruliferous aphids was very different on CABYV‐infected than on non‐infected plants. Non‐viruliferous aphids spent longer time feeding from the phloem in CABYV‐infected plants compared to non‐infected plants, suggesting that CABYV indirectly manipulates aphid feeding behaviour through its shared host plant in order to favour viral acquisition. Viruliferous aphids showed a clear preference for non‐infected over CABYV‐infected plants at short and long time, while such behaviour was not observed for non‐viruliferous aphids. Overall, our results indicate that CABYV induces changes in its host plant that modifies aphid feeding behaviour in a way that virus acquisition from infected plants is enhanced. Once the aphids become viruliferous they prefer to settle on healthy plants, leading to optimise the transmission and spread of this phloem‐limited virus.  相似文献

To extend the characterization of the human V beta gene repertoire, we utilized anchored or V beta-specific polymerase chain reaction to generate a large (approximately 200 clones) beta-chain library from the thymus of a single individual. Nine new V beta genes were identified, including single members for two new subfamilies (V beta 22 and 23), two new members of the V beta 5 subfamily, and one new member each for V beta 2, 6, 7, 9, and 12. Full-length sequences were also obtained for the published partial sequences of V beta 3, 5.3, 9.1, and 13.4, and additional nucleotides for V beta 7.1 and V beta 7.2. Based on consensus sequences from multiple clones, apparent allelic variants for six V beta genes (V beta 2.1, 5.3, 7.2, 8.2, 13.4, and 16) were also tentatively identified. Population and family studies for two of these (V beta 2.1 and 16) further confirmed that these V beta were alleles and not separate genes. Nonconservative substitutions in some of these alleles, as well as in previously identified alleles, are located at the hypervariable loops or the framework region. These findings indicate that V beta gene polymorphism appears to be significant in humans and might be the result of selective pressure imposed by conventional Ag (hypervariable loops) or superantigens (framework regions).  相似文献

Many studies investigate the benefits of polyandry, but repeated interactions with males can lower female reproductive success. Interacting with males might even decrease offspring performance if it reduces a female's ability to transfer maternal resources. Male presence can be detrimental for females in two ways: by forcing females to mate at a higher rate and through costs associated with resisting male mating attempts. Teasing apart the relative costs of elevated mating rates from those of greater male harassment is critical to understand the evolution of mating strategies. Furthermore, it is important to test whether a male's phenotype, notably body size, has differential effects on female reproductive success versus the performance of offspring, and whether this is due to male body size affecting the costs of harassment or the actual mating rate. In the eastern mosquitofish Gambusia holbrooki, males vary greatly in body size and continually attempt to inseminate females. We experimentally manipulated male presence (i.e., harassment), male body size and whether males could copulate. Exposure to males had strong detrimental effects on female reproductive output, growth and immune response, independent of male size or whether males could copulate. In contrast, there was a little evidence of a cross‐generational effect of male harassment or mating rate on offspring performance. Our results suggest that females housed with males pay direct costs due to reduced condition and offspring production and that these costs are not a consequence of increased mating rates. Furthermore, exposure to males does not affect offspring reproductive traits.  相似文献

The effects of two contrasting environmental conditions in nearshore waters off central Chile on the diet and morphospace of two cohorts of larval labrisomid blenny Calliclinus geniguttatus were studied using geometric morphometrics and gut content analysis. The two environmental conditions corresponded to (a) a cold period with upwelling-favourable southwesterly winds and a mixed water column of cooler water and (b) a warm period with calm winds and stratified warmer water. During the cold period, fish larvae had a more hydrodynamic head shape, longer jaws and a higher feeding incidence, suggesting a greater food supply due to upwelling events and a possible increase in encounter rates in the turbulent environment. In contrast, the larvae from the warm period had a more robust head shape with smaller jaws and a lower feeding incidence, which was related to higher water temperatures and lower wind intensities. The present study suggests that larvae have a rapid response to environmental changes on a short time scale (i.e., from weeks to months), showing a link between environmental conditions and changes in the phenotypic traits and diet of the larval stages of this cryptobenthic species.  相似文献

The levels of expression of G-protein alpha(q/11) (Galpha(q/11)) subunits and PLC-beta(1-4), -gamma, and -delta(1) isoforms were quantified by Western blot analysis in order to establish their contribution to the patterns of PLC functioning reported here. Quantitative measurements of the levels of Galpha(q/11) subunits in each region were obtained by comparison with known amounts of Escherichia coli expressed recombinant Galpha(q) subunits. Quantitative analysis indicated that Galpha(q/11) subunits are abundant polypeptides in human brain, with values ranging from about 1200 ng/mg in cerebral cortex to close to 900 ng/mg of membrane protein in caudate. In cerebral cortical membranes, the PLC-beta(1) isoform was more abundant than in caudate membranes. The highest levels of PLC-beta(2) expression were detected in caudate membranes. PLC-beta(3) was little expressed, and there were no significant differences in the relative values between both brain regions. Finally, the levels of the PLC-beta(4) isoform were significantly lower in caudate than in cortical membranes. It is concluded that although most of these data represent relative, not absolute, measures of protein levels within these regions, they contribute nonetheless to the significant differences observed in signaling capacities through the PLC system in both human brain regions.  相似文献

The growth–mortality relationship was assessed for larval cohorts of the Japanese sardine Sardinops melanostictus using two new approaches: (1) repeat measures in general linear model (RM‐GLMs) and (2) the autoregressive‐individual method (AIM). Both methods were compared to the traditional approach in which repeat‐measure ANOVA was used to compare the changes in increment width (WI) at age and otolith radii (RO) at age between individuals from an original population and survivors. In RM‐GLMs, both the WI at age and RO at age (i.e. at 5, 10, 15 and 20 days) were used as the dependent variables, and the standardized residuals of both regressions RO and age and RO and total length (LT), age class, and day of the year as independent variables. A significant increase in WI at age and RO at age from younger to older age classes was seen as indicative of growth‐dependent selection. In AIM, the RO‐at‐age relationship for each fish was fitted for the first 20 days, using autoregression, and then the growth traits (i.e. slopes) between the original cohorts and survivors were compared using ANOVA. In the traditional approach, the WI at age and RO at age for the first 20 days of an original population were compared with those of survivors sampled in later stages. GLMs and traditional approaches supported the growth rate (i.e. the faster an individual grows, the higher its probability of survival) and bigger is better (i.e. larger individuals at any given age will have lower probability of mortality than smaller individuals of the same age) mechanisms. Furthermore, AIM showed that individuals from original cohorts had lower otolith growth rates than those from survivors, giving further support for the growth–mortality hypothesis for the larval stage of this clupeid.  相似文献

Early life history traits of young‐of‐the‐year (YOY) round herring Etrumeus teres, caught in Tosa Bay (south‐western Japan), were studied using otolith microstructure analysis for the 2000–2003 year classes. Hatch dates ranged from October to March, and were restricted to either autumn or winter within each year class. YOY ranged from 50 to 123 mm total length (LT) and from 57 to 192 days in age. The relationship of LT to otolith radius was linear. Individual growth rates (GI) were backcalculated between the 70th and 150th days (the size range of most YOY caught) using the biological intercept method. GI ranged from 0·3 to 1·4 mm day?1 and decreased in most cases as season progressed irrespective of year class, although GI in winter cohorts were significantly higher than in autumn cohorts. Otolith growth rates (GO) ranged from 2·13 to 12·25 μm day?1 for autumn spawned YOY and from 3·12 to 12·41 μm day?1 for YOY spawned in winter. The GO trajectories followed three consistent patterns: (1) an increase in increment widths after first feeding through the second week of larval life, then (2) a plateau in increment spacing before increment widths increased again until reaching the maximum growth rate, followed by (3) a gradual decrease in increment widths until the end of the fifth month. The three stages occurred irrespective of spawning season, although YOY spawned in October and December had higher GO during stages (1) and (2) than YOY spawned in February and March, whereas higher GO was observed for late‐winter cohorts in stage (3). Otolith growth from YOY spawned in December and January showed an intermediate pattern between YOY hatched in the early autumn (October to December) and late winter (February to March). The GO trajectories were cross‐matched to the calendar date to estimate time series of otolith growth rates (GOTS) for each year. A parabolic trend was found with maximum GOTS in autumn and spring and minimum values in winter. This trend was significantly correlated to daily sea surface temperature variations. The differences in otolith growth trajectories suggest that the otolith microstructure of E. teres may be used as a natural tag for identifying autumn and winter spawned cohorts.  相似文献

The fluorescence decays of several analogues of the photoactive yellow protein (PYP) chromophore in aqueous solution have been measured by femtosecond fluorescence up-conversion and the corresponding time-resolved fluorescence spectra have been reconstructed. The native chromophore of PYP is a thioester derivative of p-coumaric acid in its trans deprotonated form. Fluorescence kinetics are reported for a thioester phenyl analogue and for two analogues where the thioester group has been changed to amide and carboxylate groups. The kinetics are compared to those we previously reported for the analogues bearing ketone and ester groups. The fluorescence decays of the full series are found to lie in the 1-10 ps range depending on the electron-acceptor character of the substituent, in good agreement with the excited-state relaxation kinetics extracted from transient absorption measurements. Steady-state photolysis is also examined and found to depend strongly on the nature of the substituent. While it has been shown that the ultrafast light-induced response of the chromophore in PYP is controlled by the properties of the protein nanospace, the present results demonstrate that, in solution, the relaxation dynamics and pathway of the chromophore is controlled by its electron donor-acceptor structure: structures of stronger electron donor-acceptor character lead to faster decays and less photoisomerisation.  相似文献
