The first line drugs for the treatment of leishmaniasis are antimonial derivatives. Poor clinical response may be credited to factors linked to the host, the drug, or the parasite. We determined the sensitivity of Leishmania sp. promastigotes and amastigotes by counting parasites exposed to increasing concentrations of meglumine antimoniate (Glucantime). Leishmania braziliensis promastigotes were significantly more sensitive than those belonging to other species. The sensitivity of L. braziliensis isolates from patients with unfavorable clinical outcome, such as therapeutic failure or relapse, was significantly lower than those from patients who had clinical cure. Poor clinical response to therapy (therapeutic failure or relapse) was also associated with inadequate antimonial therapy. We also found a significant and positive correlation between promastigotes and intracellular amastigotes with regard to their in vitro susceptibilities to meglumine antimoniate. Our data provide evidence for an association between the sensitivity of promastigotes to antimonials in vitro and clinical response to therapy in American tegumentary leishmaniasis. The high sensitivity of the local L. braziliensis to meglumine antimoniate in vitro provides an explanation for the good clinical response of cutaneous leishmaniasis in the municipality of Rio de Janeiro, Brazil, even when low-dose regimens are employed. 相似文献
The pelagic larval duration ( D PL) for 10 temperate cryptobenthic species belonging to three families: Gobiidae, Gobiesocidae and Blenniidae was investigated. Overall, the Gobiesocidae presented short D PLs varying between 11 and 18 days, the Gobiidae's D PL ranged between 14 and 39 days, and Parablennius pilicornis (Blenniidae) had an average of 33 days (range 31–37 days). The high-throughput phenotypic analysis of Arabidopsis thaliana collections requires methodological progress and automation. Methods to impose stable and reproducible soil water deficits are presented and were used to analyse plant responses to water stress. Several potential complications and methodological difficulties were identified, including the spatial and temporal variability of micrometeorological conditions within a growth chamber, the difference in soil water depletion rates between accessions and the differences in developmental stage of accessions the same time after sowing. Solutions were found. Nine accessions were grown in four experiments in a rigorously controlled growth-chamber equipped with an automated system to control soil water content and take pictures of individual plants. One accession, An1, was unaffected by water deficit in terms of leaf number, leaf area, root growth and transpiration rate per unit leaf area. Although numerous studies have been carried out on the impacts of oil spills on coral physiology, most have relied on laboratory assays. This scarcity is partly explained by the difficulty of reproducing realistic conditions in a laboratory setting or of performing experiments with toxic compounds in the field. Mesocosm systems provide the opportunity to carry out such studies with safe handling of contaminants while reproducing natural conditions required by living organisms. The mesocosm design is crucial and can lead to the development of innovative technologies to mitigate environmental impacts. Therefore, this study aimed to develop a mesocosm system for studies simulating oil spills with several key advantages, including true replication and the use of gravity to control flow‐through that reduces reliance on pumps that can clog thereby decreasing errors and costs. This adaptable system can be configured to (a) have continuous flow‐through; (b) operate as an open or closed system; (c) be fed by gravity; (d) have separate mesocosm sections that can be used for individual and simultaneous experiments; and (e) simulate the migration of oil from ocean oil spills to the nearby reefs. The mesocosm performance was assessed with two experiments using the hydrocoral Millepora alcicornis and different configurations to simulate two magnitudes of oil spills. With few exceptions, physical and chemical parameters remained stable within replicates and within treatments throughout the experiments. Physical and chemical parameters that expressed change during the experiment were still within the range of natural conditions observed in Brazilian marine environments. The photosynthetic potential (Fv/Fm) of the algae associated with M. alcicornis decreased in response to an 1% crude‐oil contamination, suggesting a successful delivery of the toxic contaminant to the targeted replicates. We investigated the effect of the exogenous polyamines spermine, spermidine and putrescine on modulation by ATP, K+, Na+, NH4+ and Mg2+ and on inhibition by ouabain of posterior gill microsomal Na+,K+-ATPase activity in the blue crab, Callinectes ornatus, acclimated to a dilute medium (21‰ salinity). This is the first kinetic demonstration of competition between spermine and
spermidine for the cation sites of a crustacean Na+,K+-ATPase. Polyamine inhibition is enhanced at low cation concentrations: spermidine almost completely inhibited total ATPase
activity, while spermine inhibition attained 58%; putrescine had a negligible effect on Na+,K+-ATPase activity. Spermine and spermidine affected both V and K for ATP hydrolysis but did not affect ouabain-insensitive ATPase activity. ATP hydrolysis in the absence of spermine and
spermidine obeyed Michaelis–Menten behavior, in contrast to the cooperative kinetics seen for both polyamines. Modulation

This minireview looks at the latest trends in the use of nanoparticles (NPs) in electrochemical biosensing systems. It includes electrochemical characterization of NPs for use as labels in affinity biosensors and other applications. DNA analysis involving NPs is one of the most important topics of current research in bionanotechnology. The advantages of the use of NPs in designing novel electrochemical sensors for DNA analysis are reviewed. Electrochemical NPs can also be used in designing immunoassays, offering the possibility of easy, low cost and simultaneous detection of several proteins. Research into NP applications in electrochemical analysis is in its infancy. The building sector is one of the most relevant sectors in terms of environmental impact. Different functional units (FUs) can be used in life cycle assessment (LCA) studies for a variety of purposes. This paper aimed to present different FUs used in the LCA of buildings and evaluate the influence of FU choice and setting in comparative studies.
Methods
As an example, we compared the “cradle to grave” environmental performance of four typical Brazilian residential buildings with different construction typologies, i.e., multi-dwelling and single dwelling, each with high and basic standards. We chose three types of FU for comparison: a dwelling with defined lifetime and occupancy parameters, an area of 1 m2 of dwelling over a year period, and the accommodation of an occupant person of the dwelling over a day.
Results and discussion
The FU choice was found to bias the results considerably. As expected, the largest global warming indicator (GWi) values per dwelling unit and occupant were identified for the high standard dwellings. However, when measured per square meter, lower standard dwellings presented the largest GWi values. This was caused by the greater concentration of people per square meter in smaller area dwellings, resulting in larger water and energy consumption per square meter. The sensitivity analysis of FU variables such as lifetime and occupancy showed the GWi contribution of the infrastructure more relevant compared with the operation in high and basic standard dwellings. The definition of lifetime and occupancy parameters is key to avoid bias and to reduce uncertainty of the results when performing a comparison of dwelling environmental performances.
Conclusions
This paper highlights the need for adequate choice and setting of FU to support intended decision-making in LCA studies of the building sector. The use of at least two FUs presented a broader picture of building performance, helping to guide effective environmental optimization efforts from different approaches and levels of analysis. Information regarding space, time, and service dimensions should be either included in the FU setting or provided in the building LCA study to allow adjustment of the results for subsequent comparison.
AbstractSirtuin 2 is a key enzyme in gene expression regulation that is often associated with tumor proliferation control and therefore is a relevant anticancer drug target. Anilinobenzamide derivatives have been discussed as selective sirtuin 2 inhibitors and can be developed further. In the present study, hologram and three-dimensional quantitative structure–activity relationship (HQSAR and 3D-QSAR) analyses were employed for determining structural contributions of a compound series containing human sirtuin-2-selective inhibitors that were then correlated with structural data from the literature. The final QSAR models were robust and predictive according to statistical validation (q2 and r2pred values higher than 0.85 and 0.75, respectively) and could be employed further to generate fragment contribution and contour maps. 3D-QSAR models together with information about the chemical properties of sirtuin 2 inhibitors can be useful for designing novel bioactive ligands.Communicated by Ramaswamy H.
