There is an urgent need for improved tools for laboratory diagnosis of active tuberculosis (TB). Here, we describe two methods, a catch-up ELISA and a dipstick test based on the detection in urine of lipoarabinomannan (LAM). LAM is a major and specific glycolipid component of the outer mycobacterial cell wall. Preliminary experiments showed that LAM is excreted in the urine of mice injected intraperitoneally with a crude cell wall preparation of Mycobacterium tuberculosis. Both methods were highly sensitive, detecting LAM at concentrations of 1 ng/ml and 5 pg/ml, respectively. Of 15 patients with active TB, all showed intermediate to high levels of LAM in their urine (absorbance values from 0.3 to 1.2, mean 0.74). Only one sample showed an absorbance value below the chosen cut off value of 0.4. All but one of the urine samples from 26 healthy nursing workers exhibited OD value below 0.4 cut off. These methods may prove valuable for rapid and simple diagnosis of TB in particular in developing countries lacking biosafety level 3 (BSL3) facilities.  相似文献

The biological functions of the acute- phase protein haptoglobin (Hp) may be related to its ability to bind hemoglobin (Hb) or to modulate immune response. Hp is expressed at a high level in lung cells, yet its protective role(s) in the lung is not known. With the use of transgenic mice overexpressing Hp in alveolar macrophages, we demonstrated that Hp diminished Hb-induced lung injury when the lung was exposed to whole blood. In transgenic mouse lungs, Hb was more efficiently removed, and the induction of stress- responsive heme oxygenase-1 gene was significantly lower when compared with wild-type mice. At 24 h after blood treatment, the ferritin level that serves as an index for intracellular iron content was also lower in alveolar macrophages in transgenic mice than in wild-type mice. We propose that an Hp-mediated Hb catabolism process exists in alveolar macrophages. This process is likely coupled to an iron mobilization pathway and may be an efficient mechanism to reduce oxidative damage associated with hemolysis.  相似文献

Insecticides are used extensively on lettuce, Lactuca sativa L., grown in southwestern Arizona because of heavy insect pressure that can potentially reduce lettuce productivity. Multiple sprays are made per season to manage these insects in lettuce. One of the major concerns related to extensive insecticide applications in lettuce is the potential subtle impact of insecticides that may reduce lettuce photosynthesis and yield. We conducted field and greenhouse experiments to examine the impact of multiple insecticides and surfactant spray applications on lettuce photosynthesis and yield. Lettuce was planted in the field in 1998, insecticides and surfactant were applied, and lettuce gas-exchange and dry weights were determined. Treatments were arranged in a split-plot consisting of insecticides as main plot and surfactant as subplot treatments in a randomized complete block design with four replications. Photosynthetic rates of lettuce were significantly reduced by endosulfan, methomyl, acephate, and surfactant at seedling stage 4 h and 2 d after the spray application was made. However, the reduction in lettuce photosynthesis by these insecticides and surfactant was only transient, and lettuce photosynthesis recovered 5 d after the spray application was made. Photosynthetic rates were not altered by zeta-cypermethrin, emamectin benzoate, and spinosad at the seedling stage. Insecticides or surfactant (Kinetic, a nonionic surfactant) did not significantly affect lettuce photosynthesis after rosette formation. In addition, lettuce dry weight was not significantly altered. These studies suggest that lettuce photosynthesis may be susceptible to some insecticides at the seedling stage. Consequently, we found that biorational insecticides, introduced to manage insect pests in lettuce, have no influence on lettuce physiology at the seedling stage, unlike the chlorinated hydrocarbons, organophosphates, or carbamates tested in this study. In a greenhouse study, we found that lettuce photosynthesis and yield were not altered by Bacillus thuringiensis application. Our results indicate that B. thuringiensis and the newer insecticides, particularly biorationals, can be used to manage lettuce insect pests without significantly altering lettuce gas-exchange and yield.  相似文献

The simulation model LSTSIM was revised and adapted to estimate the effects of different management strategies on populations of the lone star tick, Amblyomma americanum (Linnaeus), in non-agricultural areas. The control technologies evaluated were area-wide acaricide application, acaricide self-treatment of white-tailed deer, vegetation reduction and reductions in the density of white-tailed deer. Estimated changes in tick populations using these technologies compared favorably with results from actual integrated tick management studies at five different geographic locations. Area-wide acaricide application and vegetation reduction, as well as combinations of the two technologies proved to be useful for short-term, seasonal management of ticks in small non-agricultural areas and residential sites. Self-treatment of deer with acaricide, applied topically or as a systemic, appears to be the most cost-effective technology for use in area-wide, long-term programs for tick control. The results of simulations suggest that reductions in deer density should be considered as a component of any tick management program.  相似文献

物质流、能量流、信息流是生态系统过程研究中的三大主题。然而,在流域生态学研究中,有关信息流的研究一直缺位。为了推动流域信息流研究,从生物信息流切入,提出"流域生物信息流"概念,将其定义为"生物信息依托于流域生态系统过程在不同空间和系统之间进行传递、交流、作用、反馈的路径、过程与控制",并将其研究内容拟定为主要关注于水陆间、干支流间、上下游间、不同生态斑块间的流域生物信息流及其周期性节律和趋势性变迁,以及地貌、水文、人类活动等对这些生物信息流的影响等。然后,以青藏高原上青海湖重要入湖河流--沙柳河的河流水体微生物和岸带土壤微生物为研究对象,利用环境DNA技术,对沙柳河流域的自然径流驱动的流域生物信息流进行量化研究。结果表明(1)岸带土壤到水体的流域生物信息流主要由地表表面流、地下潜流等驱动,并受环境过滤效应影响,其输移效率降雨天约为62.76%、晴天约为44.16%,其中输移能力降雨天约为68.49%、晴天约为56.82%,环境过滤效应降雨天约为8.38%、晴天约为22.28%;(2)水体上游到下游的流域生物信息流主要由河川径流驱动,并受衰减效应影响,其基础综合输移效率约为97.41%/km,其中径流输移能力约为99.42%/km,无效流域生物信息流占比约为43.46%,无效流域生物信息流的半衰距离约为14.52 km;(3)降雨通过增加地表表面流等的冲蚀搬运能力并削弱环境过滤效应,促使岸带土壤到水体的流域生物信息流输移能力和输移效率增大;(4)流域生物信息流的输入在一定程度上增加了输入地的可检出生物多样性,但这种增加对于流水生态系统来讲是非累积的。  相似文献

对生态系统服务进行价值评估有助于落实生态系统的科学管理和推动生态补偿的科学实施。然而,传统的生态系统服务价值评估方法在管理实践应用中存在明显的局限性。为了规避这些局限性,另辟一条对生态系统服务价值评估的新思路,提出了描述生态系统服务价值的新指标——"选择容量价值"。选择容量价值以两个事实为基本依据:1)如果失去了任何一项生存所必需的生态系统服务(比如氧气、水),那么人类就无法生存,亦即人类经济社会活动所产生的价值是建立在消费该生态系统服务的基础上的;2)如果没有人类活动的存在,那么生态系统就给人类提供不了任何价值,亦即生态系统服务价值的度量是建立在生态系统服务所支持的人类经济社会活动所产生的价值上的。选择容量价值是指,一项人类生存生产所必需的生态系统服务,其总量为消费该生态系统服务的消费活动所提供的选择自由度(即选择容量)与该消费活动所产生的经济社会活动总价值的乘积,用非货币单位来计量。通过一个案例——对珠江流域的水资源供应的选择容量价值的估算,揭示选择容量价值对理解和量化生态系统服务价值的新洞察。结果显示,选择容量价值能够直观而综合地反映生态系统服务总量、生态系统服务消费量、社会经济...  相似文献

Iron is crucial for a variety of cellular functions in neuronal cells. Neuronal iron uptake is reflected in a robust and consistent expression of transferrin receptors and divalent metal transporter 1 (DMT 1). Conversely, the mechanisms by which neurons neutralize and possibly excrete iron are less clear. Studies indicate that neurons express ferroportin which could reflect a mechanism for iron export. We mapped the distribution of ferroportin in the adult mouse brain using an antibody prepared from a peptide representing amino acid sequences 223–303 of mouse ferroportin. The antibody specifically detected ferroportin in brain homogenates, whereas homogenates of cultured endothelial cells were devoid of immunoreactivity. In brain sections, ferroportin was confined to neuronal cell bodies and peripheral processes of cerebral cortex, hippocampus, thalamus, brain stem, and cerebellum. In brain stem ferroportin-labeling was particularly high in neurons of cranial nerve nuclei and reticular formation. Ferroportin was hardly detectable in striatum, pallidum, or hypothalamus. Among non-neuronal cells, ferroportin was detected in oligodendrocytes and choroid plexus epithelial cells. A comparison with previous studies on the distribution of transferrin receptors in neurons shows that many neuronal pools coincide with those expressing ferroportin. The data therefore indicate that neuronal iron homeostasis consists of a delicate balance between transferrin receptor-mediated uptake of iron-transferrin and ferroportin-related iron excretion. The findings also suggest a particular high turnover of iron in neuronal regions, such as habenula, hippocampus, reticular formation and cerebellum, as several neurons in these regions exhibit a prominent co-expression of transferrin receptors and ferroportin.  相似文献

Evidence, obtained with in situ perfused rat liver, indicated that pyridoxine is taken up from the perfusate by a non-concentrative process, followed by metabolic trapping. These conclusions were reached on the basis of the fact that at low concentrations (0.125 μM), the 3H of [3H]pyridoxine accumulated against a concentration gradient, but high concentrations (333 μM) of pyridoxine or 4-deoxypyridoxine prevented this apparent concentrative uptake. Under no conditions did the tissue water : perfusate concentration ratio of [3H]pyridoxine exceed unity.The perfused liver rapidly converted the labeled pyridoxine to pyridoxine phosphate, pyridoxal phosphate and pyridoxamine phosphate and released a substantial amount of pyridoxal and some pyridoxal phosphate into the perfusate. Since muscle and erythrocytes failed to oxidize pyridoxine phosphate to pyridoxal phosphate, it is suggested that the liver plays a major role in oxidizing dietary pyridoxine and pyridoxamine as their phosphate esters to supply pyridoxal phosphate which then reaches to other organs chiefly as circulating pyridoxal.  相似文献
