New drugs are generally developed against animal models of the human disease. Before they are subjected to clinical trials it might be helpful to be able to test whether they are as effective against the disease in human tissue as they were in animals. It is proposed that this can be achieved by the use of organ maintenance culture of the human diseased tissue, the relevant biochemical parameters being measured by quantitative cytochemistry. In the present studies differences between the effect of indomethacin and of the ‘chondroprotective’ drug diclofenac sodium, on human osteoarthritic cartilage, have been measured. 相似文献

Pharmaceutical recombinant proteins are widely used in human healthcare. At present, several protein expression systems are
available to generate therapeutic proteins. These conventional systems have distinct advantages and disadvantages in protein
yielding; in terms of ease of manipulation, the time required from gene transformation to protein purification, cost of production
and scaling-up capitalization, proper folding and stability of active proteins. Depending on the research goal and priorities,
a special system may be selected for protein expression. However, considering the limited variety of organisms currently used
and their usage restrictions, there are still much more pharmaceutical proteins waiting to be economically and efficiently
produced. Distinguished biological and technical features of microalgae Dunaliella such as inexpensive medium requirement,
fast growth rate, the ease of manipulation, easy scaling up procedure, facility of milking in bioreactors and the ability
of post-translational modifications make this microorganism an attractive candidate for molecular farming. 相似文献

Bone resorption and circulating PTH-like bioactivity in an animal model of hypercalcaemia of malignancy 总被引:1,自引:0,他引:1

An in vivo model of humoral hypercalcaemia of malignancy has been used to examine the role of circulating PTH-like bioactivity in the development of bone resorption and hypercalcaemia. After inoculation of cells from a renal carcinoma cell line into nude mice, circulating PTH-like bioactivity as measured by the sensitive renal and metatarsal cytochemical bioassays for PTH was elevated in only 18% and 53% of the mice respectively. Bone resorption was elevated in all the mice investigated irrespective of the level of PTH-like bioactivity. Thus, in this model, while the circulating PTH-like moiety is more potent when acting on bone, it did not correlate with the degree of bone resorption suggesting that it may not be the sole cause of the hypercalcaemia. 相似文献
