Open-water swimming (5, 10, and 25 km) has many unique challenges that separate it from other endurance sports, like marathon running and cycling. The characteristics of a successful open-water swimmer are unclear. The purpose of this study was to determine the physical and metabolic characteristics of a group of elite-level open-water swimmers. The open-water swimmers were participating in a 1-week training camp. Anthropometric, metabolic, and blood chemistry assessments were performed on the athletes. The swimmers had a VO(2)peak of 5.51 +/- 0.96 and 5.06 +/- 0.57 ml.kg(-1).min(-1) for males and females, respectively. Their lactate threshold (LT) occurred at a pace equal to 88.75% of peak pace for males and 93.75% for females. These elite open-water swimmers were smaller and lighter than competitive pool swimmers. They possess aerobic metabolic alterations that resulted in enhanced performance in distance swimming. Trainers and coaches should develop dry-land programs that will improve the athlete's muscular endurance. Furthermore, programs should be designed to increase the LT velocity as a percentage of peak swimming velocity.  相似文献

High-yield cleavage of tryptophanyl peptide bonds by o-iodosobenzoic acid.   总被引:3,自引:0,他引:3

A new procedure to cleave tryptophanyl peptide bonds in high yield is reported. The method involves treatment of the S-alkylated protein with o-iodosobenzoic acid. The procedure is highly selective for tryptophan and does not modify tyrosine or histidine, but may convert methionine to its sulfoxide derivative. The yields in the cleavage are 70--100%. Tryptophanyl bonds to alanine, glycine, serine, threonine, glutamine, arginine, and S-(pyridylethyl)cysteine are split in nearly quantitative yield, while those preceding isoleucine or valine are split in approximately 70% yield in the proteins examined in this work. The chemical mechanism for tryptophanyl bond cleavage has not been defined, but it is likely that oxidation of the indole ring occurs during the reaction with o-iodosobenzoic acid. Some problems with the quality of commercial preparations of the reagent are discussed.  相似文献

The complete primary structures of each of the two α chains commonly found in the adult hemoglobins of Malaysian Macaca nemestrina (pig-tailed macaques) were obtained from the intact chains and five fragments produced by two nonenzymatic and three enzymatic cleavage reactions. The two chains differ at a single site, the αI chains having a glutaminyl residue and the αII chains having a histidinyl residue in position 78. Both chains differ from their human counterpart at five positions, the extent of divergence being similar to that observed for most of the α chains from other species of Macaca that have been analyzed to date. Elucidation of the structural difference between the αI and αII chains demonstrates that the high degree of heterogeneity observed among the hemoglobin phenotypes of M. nemestrina is a consequence of underlying genetic variability and not a result of postsynthetic modification of genetically identical proteins. Comparisons with the hemoglobin phenotypes found in other species of Macaca support the contention that chromosomes bearing linked αI and αII genes, as well as those bearing single αI and αII alleles, combine in zygotes to produce the phenotypic variation observed in members of Malaysian populations of M. nemestrina.  相似文献

The potent cytotoxic capacity of eosinophils for parasites and host tissue has in part been attributed to the catalytic action of eosinophil peroxidase (EPO), which preferentially oxidizes Br- to the powerful bleaching oxidant HOBr in buffers that mimic serum halide composition (100 mM Cl-, 20-100 microM Br-, less than 1 microM I-). However, serum also contains 20-120 microM SCN-, a pseudohalide whose peroxidative product, HOSCN, is a weak, primarily sulfhydryl-reactive oxidant. Because of its relative abundance and high oxidation potential, we hypothesized that SCN-, not Br- or I-, is the major substrate for EPO in physiologic fluids. We find that in Earle's buffer (100 mM Cl-) supplemented with 100 microM Br- and varying concentrations of SCN-, HOBr production by activated eosinophils and purified EPO, assayed by conversion of fluorescein to dibromofluorescein, was 50% inhibited (ID50) by only 1 microM SCN-. SCN- also blocked (ID50 10 microM) EPO oxidation of I- to HOI, assayed as iodofluorescein, despite the presence of 100 microM (i.e. grossly supraphysiologic) I-. Thionitrobenzoic acid oxidation kinetics indicate that SCN- is the initial species oxidized by EPO in equimolar mixtures of SCN- and Br- and in human serum. EPO also catalyzed the covalent incorporation of [14C]SCN- into proteins in buffers regardless of Br- concentration and in human serum. Comparing the cytotoxicity of HOSCN and HOBr for host cells, we find that even subphysiologic concentrations of SCN- (3.3-10 microM) nearly completely abrogate the potent Br(-)-dependent toxicity of EPO for 51Cr-labeled aortic endothelial cells and isolated working rat hearts, recently developed models of eosinophilic endocarditis. Thus, HOSCN, hitherto best known as a bacteriostatic agent in saliva and milk, is likely also the major oxidant produced by EPO in physiologic fluids, and the presence of SCN- averts damage to EPO-coated host tissues that might otherwise accrue as a result of HOBr generation. In view of these findings, the potential role of HOSCN in eosinophil killing of parasitic pathogens deserves close examination.  相似文献

The complete amino acid sequence of rabbit muscle phosphoglucomutase has been determined by isolating the 11 peptide fragments produced by the cyanogen bromide cleavage reaction and subjecting these to automated sequencing procedures. Products produced by treatment of some of these fragments with hydroxylamine, iodosobenzoic acid, mild acid, cyanogen bromide in formic and heptafluorobutyric acids, Staphylococcus aureus V8 protease, and trypsin (with or without blocking at lysine residues) were used to complete the sequence for each of the cyanogen bromide fragments. The cyanogen bromide fragments were ordered by isolating the four tryptic peptides produced by a limited tryptic digest of the native enzyme in the presence of its substrates and its bivalent metal ion activator, Mg2+, degrading these by means of trypsin, after blocking digestion at lysine residues, and isolating and identifying all fragments thus produced that contained 10 or more residues. The 561-residue sequence thus obtained is one of the longest that has been determined by chemical means. There is excellent agreement between this sequence and published compositions after appropriate normalization. The absorbance of the enzyme is about 7.0 at 278 nm for a 1% solution; this value is 9% lower than that previously used.  相似文献

Understanding how environmental factors affect ecological parameters is important to understanding and predicting impacts of environmental change. Given evidence and anticipated impacts of climate variability, this is especially true with respect to sea ice and its role in animal life history in northern regions. We examined relationships between the extent of consolidated spring ice cover (pack and landfast), nest initiation and clutch size in common eiders (Somateria mollissima) in northern Labrador, a sub-Arctic region on the east coast of Canada. Our initial prediction was that eiders would delay nesting and have smaller clutches in years with more extensive spring ice cover. Between 1998 and 2003, we surveyed coastal islands for breeding eiders and collected information on nest age and clutch size. For those years, we estimated ice cover based on Radarsat-1 images supplied by the Canadian Ice Service during the spring period (approximately June 7–12). We found that spring ice cover was a significant positive predictor of nest initiation date, and the regression equation indicated that if the average extent of ice cover around nesting islands increased by 18 ha, average nesting date was delayed by approximately 1 day. Nest initiation date was a significant negative predictor of clutch size, and the regression equation indicated that a 20 day delay in nesting reduced average clutch size by approximately 1 egg. However, ice cover itself was not a significant predictor of clutch size. Our findings suggest that eiders breed when ice is present, but ice extent may negatively influence aspects of their breeding ecology.  相似文献
