Sodium butyrate inhibits angiogenesis of human intestinal microvascular endothelial cells through COX-2 inhibition 总被引:4,自引:0,他引:4
We examined the effect of sodium butyrate on in vitro angiogenesis and cyclooxygenase (COX) expression using primary cultures of human intestinal microvascular endothelial cells (HIMEC). Butyrate inhibited VEGF-induced cellular proliferation, transmigration and tube formation of HIMEC. Butyrate also inhibited COX-2 expression as well as prostaglandin (PG)E2 and PGI2 production, and administration of PGI2 analog partially reversed the effect of butyrate on HIMEC angiogenesis. These results indicate that sodium butyrate inhibits HIMEC angiogenesis through down-regulation of COX-2 expression and PG production, and suggest that anti-angiogenic mechanisms may also be involved in the inhibitory effect of sodium butyrate on tumor growth. 相似文献

Genetic polymorphisms in genes encoding glutathione S-transferases M1 (GSTM1; a member of class mu), T1 (GSTT1; a member of class theta) and O2 (GSTO2; a member of class omega) have been defined previously. Studies have revealed that there were significant differences between populations for allelic frequencies of GSTT1, GSTM1 and GSTO2 N412D polymorphisms. To get more insight into the genetic structure of Iranian populations the present study was done on Iranian Georgians living in Frydoonshahr (Isfahan province) and two Persian populations who living in Shiraz (Fars province) and Frydoonshahr. Study subjects consisted of 401 unrelated healthy individuals. From these 121 were Georgians. The remaining subjects were Persians from either Frydoonshahr (n = 34) or Shiraz (n = 246). The genetic polymorphism of GSTT1, GSTM1 and GSTO2 N412D was detected by PCR-based method. The frequency of GSTT1 null genotype in Georgian and Persians of Frydoonshahr and Shiraz was 15.7, 35.2 and 24.8%, respectively. There was significant difference between these populations for the distributions of the GSTT1 genotypes (χ2 = 7.00, df = 2, P = 0.030). No significant difference was observed between these populations for polymorphisms of GSTM1 (χ2 = 1.682, df = 2, P = 0.431) and GSTO N142D (χ2 = 4.622, df = 4, P = 0.328). The prevalence of GSTT1 null genotype in Iranian Georgians showed significant difference with Persians and other Asian countries, but it seems to be similar with the frequency which was reported from European populations. 相似文献

Mice genetically deficient in the chemokine receptor CXCR4 or its ligand stromal cell-derived factor (SDF)-1/CXCL12 die perinatally with marked defects in vascularization of the gastrointestinal tract. The aim of this study was to define the expression and angiogenic functions of microvascular CXCR4 and SDF-1/CXCL12 in the human intestinal tract. Studies of human colonic mucosa in vivo and primary cultures of human intestinal microvascular endothelial cells (HIMEC) in vitro showed that the intestinal microvasculature expresses CXCR4 and its cognate ligand SDF-1/CXCL12. Moreover, SDF-1/CXCL12 stimulation of HIMEC triggers CXCR4-linked G proteins, phosphorylates ERK1/2, and activates proliferative and chemotactic responses. Pharmacological studies indicate SDF-1/CXCL12 evokes HIMEC chemotaxis via activation of ERK1/2 and phosphoinositide 3-kinase signaling pathways. Consistent with chemotaxis and proliferation, endothelial tube formation was inhibited by neutralizing CXCR4 or SDF-1/CXCL12 antibodies, as well as the ERK1/2 inhibitor PD-98059. Taken together, these data demonstrate an important mechanistic role for CXCR4 and SDF-1/CXCL12 in regulating angiogenesis within the human intestinal mucosa. 相似文献

AIMS: To assist in the development of safe piggery effluent re-use guidelines by determining the level of selected pathogens and indicator organisms in the effluent ponds of 13 south-east Queensland piggeries. METHODS AND RESULTS: The numbers of thermotolerant coliforms, Campylobacter jejuni/coli, Erysipelothrix rhusiopathiae, Escherichia coli, Salmonella and rotavirus were determined in 29 samples derived from the 13 piggeries. The study demonstrated that the 13 final effluent ponds contained an average of 1.2 x 10(5) colony-forming units (CFU) 100 ml(-1) of thermotolerant coliforms and 1.03 x 10(5) CFU 100 ml(-1) of E. coli. The Campylobacter level varied from none detectable (two of 13 piggeries) to a maximum of 930 most probable number (MPN) 100 ml(-1) (two of 13 piggeries). Salmonella was detected in the final ponds of only four of the 13 piggeries and then only at a low level (highest level being 51 MPN 100 ml(-1)). No rotavirus and no Erysip. rhusiopathiae were detected. The average log10 reductions across the ponding systems to the final irrigation pond were 1.77 for thermotolerant coliforms, 1.71 for E. coli and 1.04 for Campylobacter. CONCLUSIONS: This study has provided a baseline knowledge on the levels of indicator organisms and selected pathogens in piggery effluent. SIGNIFICANCE AND IMPACT OF THE STUDY: The knowledge gained in this study will assist in the development of guidelines to ensure the safe and sustainable re-use of piggery effluent. 相似文献

Wild sheep (Ovis orientalis) as the true ancestor of domestic sheep (Ovis aries) is currently listed as vulnerable (VU) by IUCN. Effective conservation of this species requires collecting all available ecological knowledge into a single framework that could be used for management decision making. Use of Bayesian Belief Networks for such purposes have been advocated over recent decades as this approach can integrate different sources of knowledge and perform comprehensive analyses. We built a decision support tool using BBN to assist in habitat management of wild sheep populations throughout the species’ geographical range. The behaviour of the model was tested using scenario and sensitivity analysis. Habitat security, food and water suitability, and thermal cover were recognised as the main habitat variables determining wild sheep habitat suitability. Integrating the complex interactions between variables, the model can be applied for both diagnostic and predictive analyses answering “what if” and “how” questions. This approach can assist wildlife conservationists for building similar models for other poorly-studied threatened species. 相似文献

Purpose
Diminishing fossil resources and environmental concerns associated with their vast utilization have been in focus by energy policy makers and researchers. Among the different scenarios put forth to commercialize biofuels, various biorefinery concepts have aroused global interests because of their ability in converting biomass into a spectrum of marketable products and bioenergies. This study was aimed at developing different novel castor-based biorefinery scenarios for generating biodiesel and other co-products, i.e., ethanol and biogas. In these scenarios, glycerin, heat, and electricity were also considered as byproducts. Developed scenarios were also compared with a fossil reference system delivering the same amount of energy through the combustion of neat diesel.Materials and methods
Life cycle assessment (LCA) was used to investigate the environmental consequences of castor biodiesel production and consumption with a biorefinery approach. All the input and output flows from the cultivation stage to the combustion in diesel engines as well as changes in soil organic carbon (SOC) were taken into account. Impact 2002+ method was used to quantify the environmental consequences.Results and discussion
The LCA results demonstrated that in comparison with the fossil reference system, only one scenario (i.e., Sc-3 with co-production of significant amounts of biodiesel and biomethane) had 16% lower GHG emissions without even considering the improving effect of SOC. Moreover, resource damage category of this scenario was 50% lower than that of neat diesel combustion. The results proved that from a life cycle perspective, energy should be given priority in biorefineries because it is essential for a biorefinery to have a positive energy balance in order to be considered as a sustainable source of energy. Despite a positive effect on energy and GHG balances, these biorefineries had negative environmental impacts on the other damage categories like Human Health and Ecosystem Quality.Conclusions
Although biorefineries offer unique features as promising solutions for mitigating climate change and reducing dependence on fossil fuels, the selection of biomass processing options and management decisions can affect the final results in terms of environmental evaluations and energy balance. Moreover, if biorefineries are focused on transportation fuel production, a great deal of effort should still be made to have better environmental performance in Human Health and Ecosystem Quality damage categories. This study highly recommends that future studies focus towards biomass processing options and process optimization to guarantee the future of the most sustainable biofuels.39.

The English grain aphid, Sitobion avenae (Fabricius), is a severe pest of wheat plants in temperate countries. Therefore, we carried out primary screening to assess
the resistance or susceptibility of 23 commonly grown wheat varieties to this aphid at greenhouse and laboratory conditions
in Iran. Also, population attributes of this aphid were evaluated on six wheat varieties, namely Saysonz, Arta, Moghan3, Zagros,
Sardari and Shirodi. The aphids were colonized on the Hirmand wheat variety in a growth chamber at 20 ± 1°C, 60 ± 5% R.H.
and a photoperiod of 14:10 (L:D). The tested varieties were grouped into three major classes including A (e.g., Shirodi, Falat
and Moghan2), B (e.g., Sardari, Zagros and Tagan) and C (e.g., Arta, Saysonz, Moghan3 and Pishtaz). Also, the results of the
life history traits showed that the developmental time of nymphal stage ranged from 7.5 days on Zagros to 10.8 days on Saysonz.

) of S. avenae varied from 0.133 (day−1) on Saysonz to 0.210 (day−1) on Shirodi. Jackknife estimates of other population parameters on these varieties were evaluated. As a result, our findings

Lipopolysaccharide (endotoxin) tolerance is well described in monocytes and macrophages, but is less well characterized in endothelial cells. Because intestinal microvascular endothelial cells exhibit a strong immune response to LPS challenge and play a critical regulatory role in gut inflammation, we sought to characterize the activation response of these cells to repeated LPS exposure. Primary cultures of human intestinal microvascular endothelial cells (HIMEC) were stimulated with LPS over 6-60 h and activation was assessed using U937 leukocyte adhesion, expression of E-selectin, ICAM-1, VCAM-1, IL-6, IL-8, manganese superoxide dismutase, HLA-DR, and CD86. Effect of repeat LPS stimulation on HIMEC NF-kappaB and mitogen-activated protein kinase (MAPK) activation, generation of superoxide anion, and Toll-like receptor 4 expression was characterized. LPS pretreatment of HIMEC for 24-48 h significantly decreased leukocyte adhesion after subsequent LPS stimulation. LPS pretreatment inhibited expression of E-selectin, VCAM-1, IL-6, and CD86, while ICAM-1, IL-8, and HLA-DR were not altered. Manganese superoxide dismutase expression increased with repeated LPS stimulation, with a reduction in intracellular superoxide. NF-kappaB activation was transiently inhibited by LPS pretreatment for 6 h, but not at later time points. In contrast, p44/42 MAPK, p38 MAPK, and c-Jun N-terminal kinase activation demonstrated inhibition by LPS pretreatment 24 or 48 h prior. Toll-like receptor 4 expression on HIMEC was not altered by LPS. HIMEC exhibit endotoxin tolerance after repeat LPS exposure in vitro, characterized by diminished activation and intracellular superoxide anion concentration, and reduced leukocyte adhesion. HIMEC possess specific mechanisms of immunoregulatory hyporesponsiveness to repeated LPS exposure. 相似文献
