A series of quinazolinone-based rhodanine-3-acetic acids was synthesized and tested for in vitro aldose reductase inhibitory activity. All the target compounds displayed nanomolar activity against the target enzyme. Compounds 3a, 3b, and 3e exhibited almost 3-fold higher activity as compared to the only marketed reference drug epalrestat. Structure-activity relationship studies indicated that bulky substituents at the 3-phenyl ring of the quinazolinone moiety are generally not tolerated in the active site of the enzyme. Insertion of a methoxy group on the central benzylidene ring was found to have a variable effect on ALR-2 activity depending on the nature of peripheral quinazolinone ring substituents. Removal of the acetic acid moiety led to inactive or weakly active target compounds. Docking and molecular dynamic simulations of the most active rhodanine-3-acetic acid derivatives were also carried out, to provide the basis for further structure-guided design of novel inhibitors. 相似文献
The effect of dietary selenium yeast, a source of organic selenium, on heat shock protein 70 (hsp70) responses, redox status, growth and feed utilization were evaluated either in enteropathogenic Escherichia coli-challenged (EPEC) or in heat-stressed (HS) male broiler chickens grown to 42 days of age. One day-old chicks in experiment 1 were challenged orally with EPEC (10(6) cfu/chicken on day 1 and boosted by water application on days 2, 3, and 4) and fed diets with or without selenium yeast. Body weight (BW), feed conversion ratio (FCR), and total mortality were determined at 42 days of age, and this was followed by collection of ileal tissue for the quantification of total glutathione (TGSH), reduced glutathione (GSH), oxidized glutathione (GSSG), and hsp70 in randomly selected chickens from each treatment. In experiment 2, male broiler chickens were fed diets with or without selenium yeast under a thermoneutral rearing condition. At four weeks of age, blood and hepatic tissue were collected from chickens maintained in the thermoneutral environment and from chickens subjected to HS (40 degrees C for 1 h) and analyzed for TGSH, GSH, GSSG, and hsp70. Selenium yeast improved BW, FCR, and decreased mortality in both control and EPEC-challenged chicks. Selenium yeast significantly attenuated hsp70 expression in EPEC-challenged chickens and in those subjected to HS. The EPEC challenge increased TGSH and GSSG levels and decreased GSH/GSSG ratio. However, GSSG level accumulated in chickens fed diets without selenium supplementation resulting in a lower GSH/GSSG ratio in the selenium yeast-fed group. Heat stress increased GSSG level and decreased GSH/GSSG ratio. Selenium yeast-fed groups maintained higher levels of GSSG before and after HS with a resultant lower GSH/GSSG ratio. The hsp70 response was significantly less in those chickens fed selenium yeast and challenged with either EPEC or HS than in those chickens given no supplemental selenium. The results of this study suggest that selenium yeast supplementation had imparted resistance to oxidative stress associated with enteric bacteria infection and to high temperature exposure. Summary Following a phase of predominantly siliciclastic sedimentation in the Early and Middle Jurassic, a large-scale, low-latitude
carbonate depositional system was established in the northern part of the Tabas Block, part of the central-east Iranian microplate,
during the Callovian and persisted until the latest Oxfordian/Early Kimmeridgian. Running parallel to the present eastern
block margin, a NNW/SSE-trending carbonate platform developed in an area characterized by reduced subsidence rates (Shotori
Swell). The growth of this rimmed, flat-topped barrier platform strongly influenced the Upper Jurassic facies pattern and
sedimentary history of the Tabas Block. The platform sediments, represented by the predominantly fine-grained carbonates of
the Esfandiar Limestone Formation, pass eastward into slope to basin sediments of the Qal'eh Dokhtar Limestone Formation (platform-derived
allochthonites, microbialites, and peri-platform muds). Towards the west, they interfinger with bedded limestones and marlstones
(Kamar-e-Mehdi Formation), which were deposited in an extensive shelf lagoon. In a N−S direction, the Esfandiar Platform can
be traced for about 170 km, in an E-W direction, the platform extended for at least 35–40 km. The width of the eastern slope
of the platform is estimated at 10–15 km, the width of the western shelf lagoon varied considerably (>20–80 km). During the
Late Callovian to Middle Oxfordian, the Esfandiar Platform flourished under arid climatic conditions and supplied the slope
and basinal areas with large amounts of carbonates (suspended peri-platform muds and gravitational sediments). Export pulses
of platform material, e.g. ooids and aggregate grains, into the slope and basinal system are interpreted as highstand shedding
related to relative sealevel variations. The high-productivity phase was terminated in the Late Oxfordian when the eastern
platform areas drowned and homogeneous deep water marls of the Upper Oxfordian to Kimmeridgian Korond Formation onlapped both
the Qal'eh Dokhtar Limestone Formation and the drowned Esfandiar Limestone Formation. Tectonic instability, probably caused
by faulting at the margins of the Tabas Block in connection with rotational movements of the east-central Iranian block assemblage,
was responsible for the partial drowning of the eastern platform areas. In some areas, relicts of the platform persisted to

Mesenchymal stem cells (MSCs) are multipotent cells capable of differentiating into a wide range of cell types and provide a potential to transfer therapeutic protein in vivo, making them valuable candidates for gene therapy and cell therapy. However, using MSCs in in vivo is limited due to the low rate of transfection and transduction efficacy. Therefore, developing methods to efficiently transfer genes into MSCs would provide a number of opportunities for using them in the clinic. Here, we introduce a simple and robust method for efficient transduction of human adipose-derived MSCs by modification under the culture condition of human embryonic kidney cells 293 (HEK293T) and MSCs. Moreover, as a transduction enhancer, polybrene was replaced with Lipofectamine, a cationic lipid. The combined effect of gamma radiation and both hot water and sodium carbonate on physiological decay and chemical constituents of stored mandarin fruits was investigated in this work. In this consideration, the studied fruits were gamma irradiated at doses of 0, 0.3, 0.6, 1.2 and 2.4 KGy. Then, the irradiated and non-irradiated fruits were treated either by hot water or sodium bicarbonate to examine their capacity to give a further maintenance and increasing shelf life or storage periods of mandarin. Treated and non-treated fruit samples were taken at intervals of 10 days and up to 60 days. Physiological decay, total acidity, vitamin C, total sugars and TSS were investigated during the different intervals. Gamma radiation treatments showed a promising effect for maintaining the studied fruits and retarding the development of decay. On the other hand, applying both hot water and sodium bicarbonate as a further combined treatments induced a powerful effect on delaying decay development, keeping the chemical constituents near to the normal level and in turn maximizing shelf life and storage periods of the fruits under investigation. 相似文献
As recently announced by the American Society for Reproductive Medicine (ASRM), human ovarian tissue cryopreservation is an established option for fertility preservation in prepubertal girls and young women undergoing gonadotoxic treatments for cancer as well as some autoimmune diseases. Proper ovarian tissue assessment before and after cryopreservation is essential to increase success rates. Ovarian fragments from 16 patients were divided into small pieces in form of cortex with medulla, and randomly divided into the following two groups. Pieces of Group 1 (n?=?16) were frozen immediately after operation, thawed and just after thawing their quality was analyzed. Group 2 pieces (n?=?16) after operation were cooled to 5 °C for 24 h, then frozen after 24 h pre-cooling to 5 °C, thawed and just after thawing their quality was analyzed. The effectiveness of the pre-freezing cooling of tissue was evaluated by the development and viability of follicles (Calcein-AM and Propidium Iodide) using complex object parametric analyzer and sorter machine (COPAS). Positive effect of cooling of cells to low supra-zero temperatures on their future development after re-warming has been observed. New flow cytometry- technique is suitable for the evaluation and sorting of cryopreserved whole human whole intact ovarian fragments. Long time (24 h) cooling of ovarian tissue to 5 °C before cryopreservation has a trend of a cell viability increasing.
BackgroundNon-small cell lung cancer (NSCLC) is leading cause of cancer related death and the survival rate for patients with NSCLC remain poor so early diagnosis of NSCLC represents the best opportunity for cure. Cell-free DNA (cf-DNA) is extracellular nucleic acids found in cell-free plasma/serum of humans, given the recent approval of a liquid biopsy in lung cancer, the use of circulating tumor DNA as a novel non-invasive diagnostic and prognostic biomarker is promising.ObjectivesStudying whether the concentrations of circulating Cell Free DNA in serum can be used as a diagnostic and prognostic biomarker for NSCLC patients.MethodThis study was carried out on 140 subjects included 60 patients with non small cell lung cancer,40 patients with Chronic Obstructive Pulmonary Disease (COPD) and 40 healthy controls. Quantitative analysis of serum circulating cf-DNA was done b y AlU-based quantitative real time PCR. Serum level of CEA was measured by ELISA.ResultsNSCLC patients demonstrated significantly higher values of each of ALU 215, ALU 247, and DNA integrity than both COPD patients and controls. On ROC curve analysis, the total accuracy of ALU 247, ALU 115, DNA integrity (92.1%, 83.6%, 56.4%) at cutoff points (325, 565 & 0.48) respectively. On combining both DNA integrity and CEA, improved sensitivity to 93.3% was noted. For NSCLC patients, ALU 115 & ALU 247 increased significantly with more advanced stage and highest level was noticed in metastatic patients. Regarding survival there was better overall survival among patients with low DNA integrity.ConclusionSerum cf-DNA concentrations and integrity index may be valuable tool in early diagnosis of NSCLC and prediction of prognosis of those patients. 相似文献
