New series of pyrazolo[3,4-d]pyrimidines (7a-e and 13a-d) and pyrazole hydrazones 17a-d were synthesized and evaluated for their antiproliferative activity against human breast adenocarcinoma MCF-7 cell line. Most of the tested compounds exploited potent to moderate growth inhibitory activity, in particular compound 7e exhibited superior potency to the reference drug cisplatin (IC(50)=7.60 and 13.29 μM, respectively). The antitumor activity of the new compounds was accompanied by significant increase in the activity of superoxide dismutase with concomitant decrease in the activities of catalase and glutathione peroxidase and reduced glutathione level. Accordingly, the overproduction of hydrogen peroxide, nitric oxide and other free radicals allowed reactive oxygen species (ROS)-mediated tumor cells death, as monitored by reduction in the synthesis of protein and nucleic acids. 相似文献

Lipases are some of the crucial enzymes during the fungal penetrating process of the insect integument. Due to the importance and lack of information on the microbial lipases of Beauveria bassiana, investigations were carried out to purify and biochemically characterize these enzymes. The results obtained on growth medium demonstrated the highest activity of lipase 6 days after inoculation while the pH of the medium was 7.1. After three purification steps, the purified enzyme was 9.91-fold with specific activity of 20816 U/mg protein, recovery of 25% and molecular weight of 25 kDa. The purified lipase had the optimal pH and temperature at 7 and 35°C and was stable for 36–72 h under those conditions. Ca2 + significantly increased the enzyme activity and NaCl decreased it at all the tested concentrations. In addition, Mn2 + had no effect on enzyme activity but Mg2 + and Zn2 + increased it only at the highest concentration used. Three out of the four inhibitors used, significantly decreased the purified lipase activity so that most inhibition and changes in the enzyme kinetic parameters were obtained by using different concentrations of EDTA. Knowledge of enzymology provides important information for the development of fungi as microbial pest control agents opening new avenues for study of the role of enzymes in virulence. 相似文献

yield production of l-DOPA were 50 ml medium/250 ml flask with 200 rpm and 36 h, respectively. Two new techniques (addition of aniline or NaCl
to the medium) have been investigated to enhance the monophenolase activity and inhibit or reduce diphenolase activity of
tyrosinase to form high yield of l-DOPA without more oxidation to melanin. Addition of aniline to tyrosine medium at 3 μl/ml medium enhanced l-DOPA production 2.9 fold, however, addition of NaCl at 20% showed the same amount of l-DOPA as the control. Peptone and ram horn hydrolysate were studied as nitrogen sources instead of tyrosine in the medium
and they showed good nitrogen sources for l-DOPA production as tyrosine. Finally, addition of aniline (3 μl/ml) to ram horn hydrolysate was economically feasible and

Objectives: Resveratrol, with its robust antioxidant activity, has frequently been suggested as potentially having activity in cancer prevention and some recent reports have indicated that it has cancer treatment potential for several types of neoplasia. It has been found to block p‐glycoprotein and to protect against several chemotherapeutic agents’ side effects. In this study, we assessed interactive characteristics of resveratrol with docetaxel and doxorubicin and further investigated molecular bases of this interaction in cells of three different solid tumour lines (MCF‐7, HeLa and HepG2). Materials and methods and results: Resveratrol per se was found to have anti‐cancer properties, but with relatively low potency in all tested cell lines (IC50 ranged from 35.1 to 83.8 μM). Doxorubicin and docetaxel showed IC50 ranging from 0.48 to 0.72 μM and from 25.9 to 77.8 nM, respectively. Resveratrol in combination with doxorubicin and docetaxel significantly increased potencies of both chemotherapeutic agents showing IC50 ranging from 0.12 to 0.34 μM and from 7.2 to 53.02 nM, respectively. The combination index showed synergistic interaction between resveratrol and doxorubicin or docetaxel on MCF‐7 cells, and additive interactions on HeLa and HepG2 cells. Real time PCR revealed that expression of Bax and Bcl‐2 was simultaneously elevated on combination of resveratrol with doxorubicin or docetaxel in all tested cell lines, whereas p53 exhibited marginal elevation in MCF‐7 and HepG2 cells. In addition, p‐glycoprotein efflux activity was significantly inhibited, with subsequent accumulation of p‐glycoprotein substrate in intracellular compartments. Expression level of mdr1 gene was downregulated after resveratrol combined with doxorubicin or docetaxel in all tested cell lines. Conclusion: Resveratrol potentiates cytotoxic properties of both cancer drugs used in the study through increasing their intracellular level due to p‐glycoprotein inhibition and downregulation of mdr1 gene. 相似文献

A comparison of the two most famous groups of calcium-regulated photoproteins, cnidarians and ctenophores, showed unexpectedly
high degree of structural similarity regardless of their low sequence identity. It was suggested these photoproteins can play
an important role in understanding the structural basis of bioluminescence activity. Based on this postulate, in this study
the cDNA of mnemiopsin from luminous ctenophore Mnemiopsis
leidyi was cloned, expressed, purified and sequenced. The purified cDNA, with 621 base pairs, coded a 206 residues protein. Sequence
of mnemiopsin showed 93.5 and 51% similarity to other ctenophore proteins and cnidarians, respectively. The cDNA encoding
apo-mnemiopsin of M. leidyi was expressed in Escherichia coli. The purified apo-protein showed a single band on SDS-PAGE (molecular weight ~27 kDa). A semi-synthetic mnemiopsin was prepared
using coelenterazine and EDTA and its luminescence activity was measured in the presence of CaCl2. The results showed an optimum pH of 9.0 and lower calcium sensitivity compared to aequorin. Comparison of amino acid residues
in substrate binding site indicated that binding pocket of ctenophores contains less aromatic residues than cnidarians. This
can lead to a decline in the number of stacking interactions between substrate and protein which can affect the stability
of coelenterazine in binding cavity. Structural comparison of photoproteins with low sequence identity and high 3D structural

Water hyacinth, Eichhornia crassipes (Mart) Solms, originating in the amazonian basin, is a warm water aquatic plant. Water hyacinth is considered one of the most productive plants on earth and, accordingly, is considered one of the top ten world''s worst weeds. Water hyacinth spread to other tropical and subtropical regions by humans. It invaded about 62 countries in Africa, Asia and North America, and propagated extremely serious ecological, economical and social problems in the region between 40 degrees north and 45 degrees south. The dense weed of water hyacinth forms dense monocultures that can threaten local native species diversity and change the physical and chemical aquatic environment, thus altering ecosystem structure and function by disrupting food chains and nutrient cycling. We have separated and identified nine active fractions from water hyacinth and showed their promising therapeutic activities. Several compounds (alkaloid, phthalate derivatives, propanoid and phenyl derivatives) were identified in the extract of water hyacinth.Key words: water hyacinth, antimicrobial, anticancer, active compounds 相似文献

The ribosome-inhibiting toxin ricin binds exposed β1→4 linked galactosyls on multiple glycolipids and glycoproteins on the cell surface of most eukaryotic cells. After endocytosis, internal cell trafficking is promiscuous, with only a small proportion of ricin proceeding down a productive (cytotoxic) trafficking route to the endoplasmic reticulum (ER). Here, the catalytic ricin A chain traverses the membrane to inactivate the cytosolic ribosomes, which can be monitored by measuring reduction in protein biosynthetic capacity or cell viability. Although some markers have been discovered for the productive pathway, many molecular details are lacking. To identify a more comprehensive set of requirements for ricin intoxication, the authors have developed an RNAi screen in Drosophila S2 cells, screening in parallel the effects of individual RNAi treatments alone and when combined with a ricin challenge. Initial screening of 806 gene knockdowns has revealed a number of candidates for both productive and nonproductive ricin trafficking, including proteins required for transport to the Golgi, plus potential toxin interactors within the ER and cytosol. 相似文献
