Glycoprotein O (gO) is conserved among betaherpesviruses, but little is known about the maturation process of gO in human herpesvirus 6 (HHV-6). We found that HHV-6 gO maturation was accompanied by cleavage of its carboxyl terminus and required coexpression of gH and gL, which promoted the export of gO out of the endoplasmic reticulum (ER). Glutamate is the main excitatory neurotransmitter of the central nervous system (CNS), released both from neurons and glial cells. Acting via ionotropic (NMDA, AMPA, kainate) and metabotropic glutamate receptors, it is critically involved in essential regulatory functions. Disturbances of glutamatergic neurotransmission can be detected in cognitive and neurodegenerative disorders. This paper summarizes the present knowledge on the modulation of glutamate-mediated responses in the CNS. Emphasis will be put on NMDA receptor channels, which are essential executive and integrative elements of the glutamatergic system. This receptor is crucial for proper functioning of neuronal circuits; its hypofunction or overactivation can result in neuronal disturbances and neurotoxicity. Somewhat surprisingly, NMDA receptors are not widely targeted by pharmacotherapy in clinics; their robust activation or inhibition seems to be desirable only in exceptional cases. However, their fine-tuning might provide a promising manipulation to optimize the activity of the glutamatergic system and to restore proper CNS function. This orchestration utilizes several neuromodulators. Besides the classical ones such as dopamine, novel candidates emerged in the last two decades. The purinergic system is a promising possibility to optimize the activity of the glutamatergic system. It exerts not only direct and indirect influences on NMDA receptors but, by modulating glutamatergic transmission, also plays an important role in glia-neuron communication. These purinergic functions will be illustrated mostly by depicting the modulatory role of the purinergic system on glutamatergic transmission in the prefrontal cortex, a CNS area important for attention, memory and learning. 相似文献

Aims Supplying optimal quantities of mineral nutrients to growing crop plants is one way to improve crop yields. Nutrients need to be used rationally in order to avoid a negative ecological impact and undesirable effects on the sustainability of agricultural production systems. Excessive application of nutrients also affects the farmer's economy. In order to calculate the amount of fertilizer to be applied to crops, it is necessary to develop recommendation programmers that adjust nutrient rates to crop requirements.Methods Experiments in two successive seasons were conducted to investigate the effect of K fertilization and foliar application of Zn and P on yield and fiber properties of cotton cv. Giza 86. Potassium (0.0 and 47.4 kg of K ha-1) was soil applied, while chelated zinc (0.0 and 57.6 g of Zn ha-1, applied twice at 70 and 85 days after sowing 'DAS') and phosphorus (0.0, 576, 1?152 and 1?728 g of P ha-1, applied twice at 80 and 95 DAS) were applied to the foliage.Important findings Dry matter yield, total chlorophyll concentration, K, Zn and P uptake per plant, number of opened bolls per plant, boll weight, seed index, lint index, seed cotton yield per plant, seed cotton and lint yield ha-1 and earliness of harvest increased with the application of K, Zn and P. Treatments generally had no significant effect on lint percentage and fiber properties, with exceptions, for micronaire reading and flat bundle strength, and uniformity ratio, where the mean values of these characters were significantly increased over the untreated control by applying K, and for the micronaire reading in the first season, when applying P at 1?728 g ha-1, and uniformity ratio in the second season, when applying P at 1?152 and 1?728 g ha-1, where the mean values of these characters were significantly increased over the untreated control by applying P. Under the conditions of this study, applying K fertilization at 47.4 kg ha-1 combined with spraying cotton plants with zinc at 57.6 g ha-1 and also with P at 1?728 g ha-1 improved growth and yield of Egyptian cotton. 相似文献

入侵植物与重金属胁迫的相互作用研究进展
全球变化改变了植物群落的分布格局,包括入侵植物,而人为污染可能降低本地植物对入侵植物的抗性。因此,本文总结了近几十年本地植物、入侵植物和植物-土壤生态系统中重金属生物地球化学行为的研究,以加深我们对入侵植物与环境胁迫因子相互作用的认识。我们的研究结合已有文献报道表明:(i)入侵物种对环境胁迫的影响具有异质性, (ii)影响的大小是多变的, (iii)即使在同一影响类型内,影响类型也具有多向性。然而,入侵植物暴露在重金属环境中表现出更强的自我保护机制,对重金属的生物可利用性和毒性有正向或负向的影响。另一方面,由于入侵植物普遍具有较高的耐受性,加之本地植物暴露于有毒重金属污染时具有“逃逸行为”,重金属胁迫环境更有利于植物的成功入侵。但是,对于入侵植物的重金属等元素组成是否与污染地区的本地植物不同,目前尚无共识。因此,在全球范围内对外来入侵植物与本土植物的植物体内、凋落物和土壤污染物含量进行定量比较是今后研究的一个重要方向。 相似文献

This study determined the full-length sequence of CYP2E1, one of six cytochrome P450 genes previously examined in camel tissues by western blotting and semi-quantitative PCR. The Camelus dromedarius CYP2E1 has an open reading frame of 1,473 bp, and the cDNA encodes a protein of 490 amino acid residues with a molecular weight of 54.8 kDa. The deduced amino acid sequence showed the highest identity with Bos taurus (88%), Sus scrofa (87%), and Homo sapiens (83%). In a phylogenetic analysis, the C. dromedarius CYP2E1 isoform was located beside cattle and pigs. The deduced amino acid sequence of camel CYP2E1 showed the conserved proline-rich amino terminus and the heme-binding signature localized near the carboxy terminus of the protein. 相似文献

A renewable three-dimensional chemically modified carbon ceramic electrode (CCE) containing nickel powder and K4[Mo(CN)8] was constructed by sol-gel technique. The electrochemical properties and stability of modified electrode was evaluated by cyclic voltammetry in pH range 4-10. The redox couple of [Mo(CN)8] (4-/3-) was shown both as a solute in electrolyte solution and as a component of a carbon based conducting composite electrode. The apparent electron transfer rate constant (ks) and transfer coefficient (alpha) were determined by cyclic voltammetry and they were about 17.1 and 0.57 s(-1), respectively. The catalytic activity of the modified CCE toward insulin oxidation was investigated at pH range of 3-8 by cyclic votammetry. The modified electrode showed excellent electrocatalytic activity toward insulin electroxidation at physiological pH value. The modified electrode was used for insulin detection chronoamperometrically at pH 7. Under optimized condition in amperometry method, the concentration calibration range, detection limit and sensitivity were 0.5-500 nM, 0.45 nM and 6140 nA/microM, respectively. Flow injection amperometric determination of insulin at pH 7.4, at this modified electrode yielded a calibration curve with the following characteristics, linear dynamic range 100-500 pM; sensitivity 8.1 nA/nM and detection limit 40 pM (based on S/N = 3). The inherent stability at wide pH range, high sensitivity, low detection limit, low cost and ease of preparation are of advantageous of this insulin sensor. AbstractThe free energy calculation method emerges as a viable technique for ‘in-silico’ calorimetry. Efficient sampling techniques and the good choice of a reaction path connecting the reactant and the product state enable accurate computations of the free energy differences. We argue that in many cases the thermodynamic integration technique has the lowest variance when the transformation between the reactant and the product state proceeds along the natural path of the studied chemical reaction. We provide examples of free energy calculations for the fragmentation of the charged clusters and the swapping reaction of oligomer formation in proteins that follow a tentative reaction mechanism. 相似文献

The hypoxic niche of tumor leads to a tremendous increase in the extracellular adenosine concentration through alteration of adenosine metabolism in the tumor microenvironment (TME). This consequently affects cancer progression, local immune responses, and apoptosis of tumor cells. Regulatory effect of adenosine on apoptosis in TME depends on the cancer cell type, pharmacological characteristics of adenosine receptor subtypes, and the adenosine concentration in the tumor niche. Exploiting specific pharmacological adenosine receptor agonist and antagonist inducing apoptosis in cancer cells can be considered as a proper procedure to control cancer progression. Monocrystalline natural diamond, L-amino acid oxidase (L-AAOD), D-amino acid oxidase (D-AAOD), and paraffin oil were used for the design of the modified diamond paste. The technique used for the direct voltammetric assay was differential pulse voltammetry (DPV) with applied potential pulse amplitude of 25 mV vs. Ag/AgCl. The foraminiferal contents of three surface sections [Gebel Tarboul, Wadi Tarfa (WT) and Bir Dakhl (BD) sections] in southern Galala area, North-Eastern Desert, Egypt, have been studied in detail and used in biostratigraphy and chronostratigraphy of the upper Cretaceous–Paleogene successions. Sixty-nine planktonic foraminiferal species belonging to 23 genera have yielded 17 biozones. They are given in ascending order: Globotruncana aegyptiaca Interval Zone (IZ), Gansserina gansseri IZ, Contusotruncana contusa–Racemiguembelina fructicosa IZ, Abathomphalus mayaroensis IZ, P1c, P2, P3, P3a, P3b, P4, P4a, P4b, P4c, P5, E1, E2, E3, E4 and E5. Moreover, local and widespread regional hiatus was detected in the pelagic to hemipelagic sequence during Cretaceous–Paleogene boundary in southern Galala Sub-basin. This hiatus corresponds to the Early Maastrichtian–Early Eocene in Gebel Tarboul, the interval between Late Maastrichtian–Early Paleocene throughout WT sequences and Late Maastrichtian–Late Paleocene in BD section. 相似文献
