Abstract: 5-Hydroxytryptamine3 (5-HT3 ) receptors are the only known monoamine receptors mediating fast excitatory responses in mammalian neurons. Their primary structure as well as their electrophysiological and pharmacological properties show a phylogenetic relation to nicotinic acetylcholine, GABAA , and glycine receptors. As a prototypical member of this gene superfamily, we investigated the membrane topology of functional homomeric 5-HT3 receptors by using epitope tagging of the channel subunits expressed in heterologous systems. Visualization of 5-HT3 receptors in transfected COS-7 cells, either in western blot (molecular mass 61.2 ± 0.8 kDa) or in situ, was performed with previously characterized antibodies recognizing artificial epitopes as well as with anti-fusion protein antibodies directed against a wild-type receptor intracellular domain. The extracellular location of the distal C-terminal tagged domain demonstrates the presence of a fourth transmembrane domain in 5-HT3 serotonin-gated channels. In this region, the significant homology between members of this class of neurotransmitter-gated channels suggests strongly that they have a common transmembrane organization basically different from glutamate-gated and ATP-gated channels. 相似文献

The reproductive biology and breeding cycle of a tropical cyprinid, Thynnichthys thynnoides (Bleeker), from Chenderoh Reservoir, Malaysia was studied from June 1991 to November 1992. A total of 329 and 246 mature females and males and 167 immature females were sampled during the study. Three breeding cycles were observed and the cycles coincided with high reservoir water level which resulted in the floodings of the littoral zone. Five and four gonad developmental stages were observed for females and males, respectively. Oocyte diameter distribution study indicated the species was a total spawner although clutches of ripe oocytes might be released over an extended time period. Fecundity was related to body weight and both absolute and relative fecundity increased with body size. Absolute fecundity ranged from 26962 ± 1484 to 173520 ± 127420 eggs fish–1, whereas relative fecundity ranged from 121 ± 53 to 451 ± 259 eggs g–1 body weight, respectively. 相似文献

Duplications in Xp including the DSS (dosage sensitive sex reversal) region cause male to female sex reversal. We investigated two patients from families with Xp duplications. The first case was one of two sisters with karyotype 46,XY, der(22), t(X;22)(p11.3;p11)mat and unambiguous female genitalia. The living sister was developmentally retarded, and showed multiple dysmorphic features and an acrocallosal syndrome. The second case was a boy with a maternally inherited direct duplication of Xp21.3-pter with the breakpoint close to the DSS locus. He had multiple abnormalities and micropenis, but otherwise unambiguous male genitalia. We performed quantitative Southern blot analysis with probes from Xp22.13 to p21.2 to define the duplicated region. Clinical, cytogenetic, and molecular data from both patients were compared with those of previously reported related cases. A comparison of the extragenital symptoms revealed no differences between patients with or without sex reversal. In both cases, the symptoms were non-specific. Among 22 patients with a duplication in Xp, nine had unambiguous female genitalia and a well-documented duplication of the DSS region. Two patients with duplication of DSS showed ambiguous external genitalia. From these data, we conclude that induction of testicular tissue may start in these patients, but that the type of genitalia depends on the degree of subsequent degeneration by a gene in DSS. 相似文献

Somatic embryos were obtained from callus cultures derived from leaf explants of the winged bean, Psophocarpus tetragonolobus (L.) DC. Initiation and development of the somatic embryos occurred with a two-step culture method. Callus cultures initiated on MS medium with NAA and BAP, upon transfer to a new medium with IAA and BAP, produced somatic embryos. Maximum embryogenesis of 60% was obtained on induction medium with 0.5 mg/l NAA plus 1.0 mg/l BAP followed by transfer to a secondary medium with 0.1 mg/l IAA and 2.0 mg/l BAP. Optimal embryo germination and plantlet development was achieved on MS medium with 0.2 mg/l BAP plus 0.1 mg/l IBA. Ten erythropoietin (EPO) fractions differing in sialic acid content, ranging from 9.5 to 13.8 mol mol–1 of EPO, were obtained from baby hamster kidney cell-derived recombinant human EPO by Mono Q column chromatography. The mean pI values of the EPO fractions determined by IEF-gel electrophoresis systematically shifted from 4.11 to 3.31, coinciding with the sialic acid content, without a change in the constitution of asialo N-linked oligosaccharides of each fraction. Although a linear relationship between thein vivo bioactivity and the sialic acid content of the fractionated, samples was observed until 12.1 mol mol–1 of EPO, there was no further increase in their activity over 12.4 mol mol–1 of EPO. On the other hand, an inverse relationship between thein vitro bioactivity and sialic acid content of EPO was observed. Also, we showed that thein vivo bioactivity of some fractions with low sialic acid contents was increased after treatment with 2,6-sialyltransferase, but thein vivo bioactivity of the other fractions with high sialic acid contents was either decreased or not affected.Abbreviations EPO

Three groups of rats were fed two types of synthetic diets for 52 d. The—A group was allowed free access to a vitamin A-deficient
diet and showed classical signs of vitamin A deficiency. The brain was the only organ in our experiment where no significant
weight difference was present among the three groups. In the brain, calcium concentration was significantly higher in the—A
group when compared with the PF (Pair-fed; allowed restricted amount of control diet) and +A groups (allowed free access to
control diet). In the tibia, calcium and magnesium concentrations were significantly lower in the—A group when compared with
other two groups. Excessive accumulation of calcium in brain and apparently similar unbalance in bone, mineral concentration
were observed in central nervous system (CNS) degenerative diseases. Our results suggest that abnormal metabolism of calcium
and magnesium in some tissues and excessive accumulation of calcium in brain may be responsible for the development of neurological

The rhythmic production of melatonin is governed by intrapineal oscillators in all fish species so far investigated except the rainbow trout. To determine whether the latter represents an exception among fish, we measured in vitro melatonin secretion in pineal organs of nine wild freshwater and six marine teleost species cultured at constant temperature and under different photic conditions. The results demonstrate that pineal organs of all species maintain a rhythmic secretion of melatonin under light:dark cycles and complete darkness, and strongly suggest that most fish possess endogenous intrapineal oscillators driving the rhythm of melatonin production, with the exception of the rainbow trout.Abbreviations

Among Trichophyton spp. examined for urease production, T. rubrum was negative, whereas T. mentagrophytes appeared to be the most active species. Urease was not detected in cell-free culture fluids of the tested fungi. The endocellular urease of the test fungi was essentially constitutive. Moreover, addition of urea to the growth medium of these organisms markedly inhibited their mycelial biomass and ureolytic yield. Summary Normal and neoplastic growth of epithelial cells depends on mutual interactions between epithelial and stromal cells. As a
tool for the study of the underlying molecular mechanisms, we have developed temperature-sensitive, nontransformed cell lines
derived from rat uterine epithelium and stroma by transfecting primary cultures with a temperature-sensitive mutant of the
SV40 large T antigen. The epithelial and stromal cell lines obtained shared relevant morphological characteristics with the
primary cells from which they were derived. Immunocytochemical analysis showed that the epithelial cell lines expressed the
intermediate filament cytokeratin, whereas the stromal lines expressed the intermediate filament vimentin. Alkaline phosphatase
activity was present in all cell lines examined. All cell lines were anchorage dependent and did not form foci. One epithelial
cell line expressed oxytocin mRNA, a gene product recently shown to be highly expressed in vivo in the uterine epithelium
at term. If grown on Matrigel, this cell line formed domelike structures, a further characteristic of its differentiated phenotype.
In an attempt to reconstitute an endometrium in vitro, epithelial cells were seeded on top of a layer of stromal cells. Paraffin
cross sections showed that this in vitro system consisted of a bilayer structure. Four to five cuboidal epithelial cells were
typically anchored atop one stromal cell, forming an endometriumlike tissue. The present in vitro system should provide a
useful model for further studies on endometrial functions and epithelial/stromal cell interactions at a molecular level. 相似文献

Translation of the human hepatitis C virus (HCV) RNA genome occurs by a mechanism known as "internal ribosome entry." This unusual strategy of translation is employed by naturally uncapped picornaviral genomic RNAs and several cellular mRNAs. A common feature of these RNAs is a relatively long 5' noncoding region (NCR) that folds into a complex secondary structure harboring an internal ribosome entry site (IRES). Evidence derived from the use of dicistronic expression systems, combined with an extensive mutational analysis, demonstrated the presence of an IRES within the HCV 5'NCR. The results of our continued mutational analysis to map the critical structural elements of the HCV IRES has led to the identification of a pseudoknot structure upstream of the initiator AUG. The evidence presented in this study is based upon the mutational analysis of the putative pseudoknot structure. This is further substantiated by biochemical and enzymatic probing of the wild-type and mutant 5'NCR. Further, the thermodynamic calculations, based upon a modified RNAKNOT program, are consistent with the presence of a pseudoknot structure located upstream of the initiator AUG. Maintenance of this structural element is critical for internal initiation of translation. The pseudoknot structure in the 5'NCR represents a highly conserved feature of all HCV subtypes and members of the pestivirus family, including hog cholera virus and bovine viral diarrhea virus. 相似文献
