Primaquine (PQ) and Tafenoquine (TQ) are clinically important 8‐aminoquinolines (8‐AQ) used for radical cure treatment of Pvivax infection, known to target hepatic hypnozoites. 8‐AQs can trigger haemolytic anaemia in individuals with glucose‐6‐phosphate dehydrogenase deficiency (G6PDd), yet the mechanisms of haemolytic toxicity remain unknown. To address this issue, we used a humanized mouse model known to predict haemolytic toxicity responses in G6PDd human red blood cells (huRBCs). To evaluate the markers of eryptosis, huRBCs were isolated from mice 24–48 h post‐treatment and analysed for effects on phosphatidylserine (PS), intracellular reactive oxygen species (ROS) and autofluorescence. Urinalysis was performed to evaluate the occurrence of intravascular and extravascular haemolysis. Spleen and liver tissue harvested at 24 h and 5–7 days post‐treatment were stained for the presence of CD169+ macrophages, F4/80+ macrophages, Ter119+ mouse RBCs, glycophorin A+ huRBCs and murine reticulocytes (muRetics). G6PDd‐huRBCs from PQ/TQ treated mice showed increased markers for eryptosis as early as 24 h post‐treatment. This coincided with an early rise in levels of muRetics. Urinalysis revealed concurrent intravascular and extravascular haemolysis in response to PQ/TQ. Splenic CD169+ macrophages, present in all groups at day 1 post‐dosing were eliminated by days 5–7 in PQ/TQ treated mice only, while liver F4/80 macrophages and iron deposits increased. Collectively, our data suggest 8‐AQ treated G6PDd‐huRBCs have early physiological responses to treatment, including increased markers for eryptosis indicative of oxidative stress, resulting in extramedullary haematopoiesis and loss of splenic CD169+ macrophages, prompting the liver to act as the primary site of clearance.  相似文献

ABSTRACT: BACKGROUND: Electrical Impedance Tomography (EIT) is used as a fast clinical imaging technique formonitoring the health of the human organs such as lungs, heart, brain and breast. Eachpractical EIT reconstruction algorithm should be efficient enough in terms of convergencerate, and accuracy. The main objective of this study is to investigate the feasibility of preciseempirical conductivity imaging using a sinc-convolution algorithm in D-bar framework. METHODS: At the first step, synthetic and experimental data were used to compute an intermediate objectnamed scattering transform. Next, this object was used in a 2-day integral equation whichwas precisely and rapidly solved via sinc-convolution algorithm to find the square root of theconductivity for each pixel of image. For the purpose of comparison, multigrid and NOSERalgorithms were implemented under a similar setting. Quality of reconstructions of syntheticmodels was tested against GREIT approved quality measures. To validate the simulationresults, reconstructions of a phantom chest and a human lung were used. RESULTS: Evaluation of synthetic reconstructions shows that the quality of sinc-convolutionreconstructions is considerably better than that of each of its competitors in terms ofamplitude response, position error, ringing, resolution and shape-deformation. In addition, theresults confirm near-exponential and linear convergence rates for sinc-convolution andmultigrid, respectively. Moreover, the least degree of relative errors and the most degree oftruth were found in sinc-convolution reconstructions from experimental phantom data.Reconstructions of clinical lung data show that the related physiological effect is wellrecovered by sinc-convolution algorithm. CONCLUSIONS: Parametric evaluation demonstrates the efficiency of sinc-convolution to reconstruct accurateconductivity images from experimental data. Excellent results in phantom and clinicalreconstructions using sinc-convolution support parametric assessment results and suggest thesinc-convolution to be used for precise clinical EIT applications.  相似文献

The formation and subsequent resolution of Holliday junctions are critical stages in recombination. We describe a new Escherichia coli endonuclease that resolves Holliday intermediates by junction cleavage. The 14 kDa Rus protein binds DNA containing a synthetic four-way junction (X-DNA) and introduces symmetrical cuts in two strands to give nicked duplex products. Rus also processes Holliday intermediates made by RecA into products that are characteristic of junction resolution. The cleavage activity on X-DNA is remarkably similar to that of RuvC. Both proteins preferentially cut the same two strands at the same location. Increased expression of Rus suppresses the DNA repair and recombination defects of ruvA, ruvB and ruvC mutants. We conclude that all ruv strains are defective in junction cleavage, and discuss pathways for Holliday junction resolution by RuvAB, RuvC, RecG and Rus.  相似文献

Investigations determined the relative preference of prekallikrein (PK) or factor XI/XIa (FXI/FXIa) binding to endothelial cells (HUVECs). In microtiter plates, biotinylated high molecular weight kininogen (biotin-HK) or biotin-FXI binding to HUVEC monolayers or their matrix proteins, but not fibronectin-coated plastic microtiter plate wells, was specifically blocked by antibodies to each of the receptors of HK, uPAR, gC1qR, or cytokeratin 1. Fluorescein isothiocyanate (FITC)-PK specifically bound to HUVEC suspensions without added Zn2+, whereas FITC-FXI or -FXIa binding to HUVEC suspensions required 10 microM added Zn2+ to support specific binding. Plasma concentrations of FXI did not block FITC-PK binding to HUVECs in the absence or presence of 10 microM Zn2+. In the absence of HK, the level of FITC-FXI or -FXIa binding was half that seen in its presence. At physiologic concentrations, PK (450 nM) abolished FITC-FXI or -FXIa binding to HUVEC suspensions in the absence or presence of HK in the presence of 10 microM Zn2+. Released Zn2+ from 2-8 x 10(8) collagen-activated platelets/ml supported biotin-FXI binding to HUVEC monolayers, but platelet activation was not necessary to support biotin-PK binding to HUVECs. At physiologic concentrations, PK also abolished FXI binding to HUVECs in the presence of activated platelets, but FXI did not influence PK binding. PK in the presence or absence of HK preferentially bound to HUVECs over FXI or FXIa. Elevated Zn2+ concentrations are required for FXI but not PK binding, but the presence of physiologic concentrations of PK and HK also prevented FXI binding. PK preferential binding to endothelial cells contributes to their anticoagulant nature.  相似文献

In this paper, a graphene-based tunable multi-band terahertz absorber is proposed and numerically investigated. The proposed absorber can achieve perfect absorption within both sharp and ultra-broadband absorption spectra. This wide range of absorption is gathered through a unique combination of periodically cross- and square-shaped dielectrics sandwiched between two graphene sheets; the latter enables it to offer more absorption in comparison with the traditional single-layer graphene structures. The aforementioned top layer is mounted on a gold plate separated by a Topas layer with zero volume loss. Furthermore, in our proposed approach, we investigated the possibility of changing the shapes and sizes of the dielectric layers instead of the geometry of the graphene layers to alleviate the edge effects and manufacturing complications. In numerical simulations, parameters, such as graphene Fermi energy and the dimensions of the proposed dielectric layout, have been optimally tuned to reach perfect absorption. We have verified that the performance of our dielectric layout called fishnet, with two widely investigated dielectric layouts in the literature (namely, cross-shaped and frame-and-square). Our results demonstrate two absorption bands with near-unity absorbance at frequencies of 1.6–2.3 and 4.2–4.9 THz, with absorption efficiency of 98% in 1.96 and 4.62 THz, respectively. Moreover, a broadband absorption in the 7.77–9.78 THz is observed with an absorption efficiency of 99.6% that was attained in 8.44–9.11 THz. Finally, the versatility provided by the tunability of three operation bands of the absorber makes it a great candidate for integration into terahertz optoelectronic devices.

Color is one of the most prominent features of an image and used in many skin and face detection applications. Color space transformation is widely used by researchers to improve face and skin detection performance. Despite the substantial research efforts in this area, choosing a proper color space in terms of skin and face classification performance which can address issues like illumination variations, various camera characteristics and diversity in skin color tones has remained an open issue. This research proposes a new three-dimensional hybrid color space termed SKN by employing the Genetic Algorithm heuristic and Principal Component Analysis to find the optimal representation of human skin color in over seventeen existing color spaces. Genetic Algorithm heuristic is used to find the optimal color component combination setup in terms of skin detection accuracy while the Principal Component Analysis projects the optimal Genetic Algorithm solution to a less complex dimension. Pixel wise skin detection was used to evaluate the performance of the proposed color space. We have employed four classifiers including Random Forest, Naïve Bayes, Support Vector Machine and Multilayer Perceptron in order to generate the human skin color predictive model. The proposed color space was compared to some existing color spaces and shows superior results in terms of pixel-wise skin detection accuracy. Experimental results show that by using Random Forest classifier, the proposed SKN color space obtained an average F-score and True Positive Rate of 0.953 and False Positive Rate of 0.0482 which outperformed the existing color spaces in terms of pixel wise skin detection accuracy. The results also indicate that among the classifiers used in this study, Random Forest is the most suitable classifier for pixel wise skin detection applications.  相似文献
