It has been demonstrated that plant roots can take up small amounts of low-molecular weight (LMW) compounds from the surrounding soil. Root uptake of LMW compounds have been investigated by applying isotopically labelled sugars or amino acids but not labelled organic matter. We tested whether wheat roots took up LMW compounds released from dual-labelled (13C and 15N) green manure by analysing for excess 13C in roots. To estimate the fraction of green manure C that potentially was available for root uptake, excess 13C and 15N in the primary decomposers was estimated by analysing soil dwelling Collembola that primarily feed on fungi or microfauna. The experimental setup consisted of soil microcosm with wheat and dual-labelled green manure additions. Plant growth, plant N and recoveries of 13C and 15N in soil, roots, shoots and Collembola were measured at 27, 56 and 84 days. We found a small (<1%) but significant uptake of green manure derived 13C in roots at the first but not the two last samplings. About 50% of green manure C was not recovered from the soil-plant system at 27 days and additional 8% was not recovered at 84 days. Up to 23% of C in collembolans derived from the green manure at 56 days (the 27 days sampling was lost). Using a linear mixing model we estimated that roots or root effluxes provided the main C source for collembolans (54−79%). We conclude that there is no solid support for claiming that roots assimilated green manure derived C due to very small or no recoveries of excess 13C in wheat roots. During the incubation the pool of green manure derived C available for root uptake decreased due to decomposition. However, the isotopic composition in Collembola indicated that there was a considerable fraction of green manure derived C in the decomposer system at 56 days thus supporting the premise that LMW compounds containing C from the green manure was released throughout the incubation. Background

Atrial electrical remodeling has been shown to influence the outcome the outcome following cardioversion of atrial fibrillation (AF) in experimental studies. The aim of the present study was to find out whether a non-invasively measured atrial fibrillatory cycle length, alone or in combination with other non-invasive parameters, could predict sinus rhythm maintenance after cardioversion of AF.

Methods

Dominant atrial cycle length (DACL), a previously validated non-invasive index of atrial refractoriness, was measured from lead V1 and a unipolar oesophageal lead prior to cardioversion in 37 patients with persistent AF undergoing their first cardioversion.

Results

32 patients were successfully cardioverted to sinus rhythm. The mean DACL in the 22 patients who suffered recurrence of AF within 6 weeks was 152 ± 15 ms (V1) and 147 ± 14 ms (oesophagus) compared to 155 ± 17 ms (V1) and 151 ± 18 ms (oesophagus) in those maintaining sinus rhythm (NS). Left atrial diameter was 48 ± 4 mm and 44 ± 7 mm respectively (NS). The optimal parameter predicting maintenance of sinus rhythm after 6 weeks appeared to be the ratio of the lowest dominant atrial cycle length (oesophageal lead or V1) to left atrial diameter. This ratio was significantly higher in patients remaining in sinus rhythm (3.4 ± 0.6 vs. 3.1 ± 0.4 ms/mm respectively, p = 0.04).

Conclusion

In this study neither an index of atrial refractory period nor left atrial diameter alone were predictors of AF recurrence within the 6 weeks of follow-up. The ratio of the two (combining electrophysiological and anatomical measurements) only slightly improve the identification of patients at high risk of recurrence of persistent AF. Consequently, other ways to asses electrical remodeling and / or other variables besides electrical remodeling are involved in determining the outcome following cardioversion.  相似文献

Increased inactivation of nitric oxide (NO) by superoxide has been implicated in nitrate tolerance. Here, we set out to compare the inhibitory effect of superoxide on endothelium-dependent, acetylcholine (ACh)-mediated vascular relaxation with that on the endothelium-independent effects of glyceryl trinitrate (GTN) and another NO donor drug, S-nitrosoglutathione (GSNO). Rings of thoracic aorta from adult male Wistar rats (350-450 g) were precontracted with phenylephrine (approximately EC(90)) prior to cumulative additions (10 nM/L-10 microM/L) of GTN, GSNO, or ACh. Rings were then treated with the superoxide generator pyrogallol (300 micromol/L) alone or following pretreatment with the Cu/Zn superoxide dismutase inhibitor diethyldithiocarbamate (DETCA; 100 micromol/L), and cumulative additions of the vasodilators were repeated. All experiments were conducted in the presence of catalase (3000 U/ml) to prevent accumulation of hydrogen peroxide. Relaxation to ACh was abolished by pyrogallol-derived superoxide. Relaxation to GSNO was significantly inhibited by superoxide (P < 0.05, n = 8) and was more pronounced at lower GSNO concentrations. However, GTN was relatively resistant to inhibition by superoxide with modest inhibition only occurring in rings pretreated with DETCA prior to pyrogallol (P < 0.05; n = 8). In contrast to GSNO, the inhibitory effect was more pronounced with high concentrations of GTN, suggesting that the mechanism underlying superoxide-mediated inhibition is different for the two NO donor drugs. Further experiments showed that vascular responses to ACh were not inhibited (P > 0.05, n = 6) in aortic rings made tolerant to GTN (10 micromol/L, 2-h incubation) and that treatment of vessels with the antioxidant vitamin C (1 mmol/L) successfully prevented the development of tolerance. Taken together, these results suggest that superoxide is not a major factor in tolerance in vitro and imply that the protective actions of vitamin C are unrelated to its antioxidant activity in this setting.  相似文献

PI3Kα/mTOR ATP-competitive inhibitors are considered as one of the promising molecularly targeted cancer therapeutics. Based on lead compound A from the literature, two similar series of 2-substituted-4-morpholino-pyrido[3,2-d]pyrimidine and pyrido[2,3-d]pyrimidine analogs were designed and synthesized as PI3Kα/mTOR dual inhibitors. Interestingly, most of the series gave excellent inhibition for both enzymes with IC50 values ranging from single to double digit nM. Unlike many PI3Kα/mTOR dual inhibitors, our compounds displayed selectivity for PI3Kα. Based on its potent enzyme inhibitory activity, selectivity for PI3Kα and good therapeutic index in 2D cell culture viability assays, compound 4h was chosen to be evaluated in 3D culture for its IC50 against MCF7 breast cancer cells as well as for docking studies with both enzymes.  相似文献

By using denaturing gradient gel electrophoresis (DGGE) and simultaneously measuring the enzymatic activity of chitinase, we could link genetic diversity of the indigenous microbial communities with chitinase activity in compost samples. A garden/park waste compost and a source separated organic household waste compost, showed different genetic diversity as measured by PCR-DGGE of total DNA extracted from the composts. The household waste compost had the highest chitinase activity. To increase chitinase activity, the two composts were amended with chitin. This addition induced a change in both the bacterial and fungal genetic diversity when compared to the non-amended compost samples. Likewise, both composts reacted to the addition of chitin with an increase in chitinase activity. Thus, a relationship between genetic diversity and chitinase activity was established for the composts in question. The N-mineralization in the household waste compost was apparently increased by the addition of chitin, while such an effect was not observed in the garden/park waste compost.  相似文献

Privileged structure-based libraries have been shown to provide high affinity lead compounds for a variety of important biological targets. The present study describes the synthesis and screening of a 2-aminothiazole based compound library to determine their utility as antimicrobials, focusing on MRSA. Several of the compounds in this series demonstrated improved antimicrobial activity as compared to ceftriaxone (CTX), a β-lactam antibiotic. The most potent compound (21) had MICs in the range of 2–4 μg/ml across a panel of Staphylococcus aureus strains. In addition, trifluoromethoxy substituted aminothiazoles and aminobenzothiazoles were found to be potent antimicrobials with MICs of 2–16 μg/ml.  相似文献

The acrosome reaction (AR) in bull spermatozoa was induced by the Ca2(+)-ionophore A23187, by dilauroylphosphatidylcholine or by arachidonic acid in the presence of Ca2+ in the incubation medium. The occurrence of AR was determined by following the release of acrosin from the cells. Nordihydroguaiaretic acid (NDGA), an inhibitor of both lipoxygenase and prostaglandin-synthetase, caused 35%, 43% and 69% inhibition of AR at concentrations of 1, 10 or 100 microM, respectively. Eicosatetraynoic acid (ETYA), an analogue of arachidonic acid, caused 17%, 61% and 77% inhibition of AR at concentrations of 20, 40 or 80 micrograms/ml, respectively. When AR was induced by arachidonic acid, ETYA, causes 36% and 58% inhibition at concentrations of 2 or 20 micrograms/ml, respectively. Under identical conditions, 100 microM indomethacin, a specific inhibitor of prostaglandin-synthetase, showed no inhibition but rather 35% stimulation at acrosin release rate. The fact that AR is inhibited by NDGA and not by indomethacin indicates that the lipoxygenase, rather than prostaglandin-synthetase, is involved in the mechanism of AR. Since the inhibition by NDGA is seen in the presence of the Ca-ionophore, we suggest that lipoxygenase activity is not involved in enhancing calcium transport into the cell, but rather at other steps in AR mechanism. A thin-layer chromatography revealed the presence of 15-HETE, the classical product of 15-lipoxygenase activity, which was identified by HPLC. Under AR conditions, there is an elevation of lipoxygenase products and the addition of NDGA caused a reduction in their levels. The inhibition of acrosin release by NDGA can be eliminated by adding 15-HETE or 15-HPETE to the incubation medium. In conclusion, we suggest here for the first time, a physiological role for 15-lipoxygenase in the mechanism of AR in mammalian spermatozoa.  相似文献

Nigrospora oryzae is reported to be partially deficient for biotin. The external requirement for the vitamin is shown to be conditioned by the nitrogen source. The deficiency for biotin is apparent when urea or nitrate is used to supply the nitrogen. The organism is found to be more dependent on an external supply of biotin in media where autoclaved urea is the source of nitrogen than where nitrate or sterilefiltered urea is that source. Aspartic acid is found to induce a complete sparing action for biotin requirement should the nitrogen source be nitrate. Malt and yeast extracts are reported to supply probable unidentified growth promoting substances other than biotin.  相似文献
