All six available lysine residues in bacteriorhodopsin were amidinated with dimethyl-3,3'-dithiobispropionimidate, which is a crosslinking agent. The photocycle was studied by measuring light absorption and electric signals. The data show an essential change in the photocycle: instead of single components, the rise of the signal due to the M intermediate can be decomposed into two components, and the decay into three. The life-times and the intensities of these components and in general the proton pumping activity of bacteriorhodopsin depend only negligibly upon pH. Changes upon removing the crosslinks are not significantly different from those in the crosslinked samples. The lysine residues therefore may not be considered of primary importance in proton translocation. 相似文献

We consider a template-instructed MDV-1 RNA replication catalyzed by Q beta replicase. The relaxation of the effective binding of the growing chain to the enzyme is simulated, considering a Markov process where refolding events in the chain are concomitant with chain elongation events. Neomorph and leaf differentiation as alternative morphogenetic pathways in soybean tissue culture. 总被引:1,自引:0,他引:1
Somatic embryogenesis and organogenesis were induced on immature cotyledons of different soybean cultivars. The anatomical investigation of morphogenesis proved neomorph differentiation instead of somatic embryos, and leaf formation instead of shoot development. While normal embryos were induced in 0-3.1% of the explants, neomorphs developed at a much higher rate i.e. in 10.5-78.9% depending on the genotype. Likewise organogenesis preferably followed the pathway of leaflet development (3.1-26.3%) than that of shoot tip formation (0-2.6%). Low plant regeneration frequency of soybean can partly be explained with these two alternative abortive pathways of morphogenesis probably induced with higher frequency than the normal pathways by the generally used in vitro methods. 相似文献

IAA and ethylene is in the photoperiodic flower induction in Pharbitis nil, we measured changes in their levels during inductive and non-inductive photoperiods, and the effects of ethylene biosynthesis

Adventitious root formation in vitro in 1-mm stem slices cut from microshoots of apple cv. Jork 9 was studied using light
and electron microscopy. When indole-3-butyric acid (IBA) had been added to the medium, starch grains accumulated during the
first 24 h of culture in cells of the cambial region and in cells in the vicinity of vascular tissue and in the primary rays.
This accumulation occurred only in the basal part of explants. After that, the nuclei in these cells were activated, and the
density of the cytoplasm and the number of cell organelles increased, whereas starch was broken down. Cambium cells started
to divide transversely and at 96 h, after several divisions, a continuous ring of isodiametric cytoplasmic cells had appeared
around the xylem near the basal cutting surface. The cells in this ring were rich in cell structures, and did not contain
large starch grains and a central vacuole. Root meristemoids regenerated from the portions of the ring that were localized
in the primary rays. From the other cells in the ring, callus developed. The meristemoids did not grow into the direction
of the epidermis as in shoots, but along the vascular bundles. After emergence from the cutting surface, the meristemoids
were transformed into small, dome-like primordia. They developed a typical root apex with root cap, root ground meristem and
tracheid connection with shoot vascular tissue.

Three different fluorochrome and specific counterstain combination (DAPI/AMD, DA/DAPI and CMA/DA) treatments were applied to the chromosomes of four Microtidae (Rodentia) species. The results complete the data obtained in our previous paper (Burgos, M., Jiménez, R., & Dìaz de la Guardia, R., Genome 30:540–546, 1988) and prove that the changes in the constitutive heterochromatin in the evolution of the karyotypes of these species are not only due to gain or loss of heterochromatin, but are qualitative with respect to their nucleotide composition, repeated base pair organization or DNA-protein complex modification.
