The NKLy ascitic tumor cells in the stationary phase of growth were fractionated by velocity sedimentation method. Cells from the obtained fractions were characterized by measurements of DNA contents and 3H-thymidine incorporation. The surface properties of the cells from five fractions, differing in proliferative capacity, stage of the cell cycle and ploidy were considered using cell electrophoresis, two polymer aqueous phase system and Alcian blue sorption. A correlation between electrophoretic mobility and cell partition constant for these fractions has been obtained. No correlation was found between these parameters and dye absorption. The surface charge of the cells from G0/G1 and S fractions was higher than that of other cells. The polyploid NKLy cells demonstrated a lower surface charge. The surface properties of the tumor cells differing in proliferative capacity, stage of the cell cycle and ploidy are discussed.  相似文献

The efficiency of different methods for recovery of spore toxin complex (STC) from solid-state fermentation (SSF) extracts of Bacillus thuringiensis israelensis (Bti) and Lysinibacillus sphaericus 14N1(Ls14N1) was studied. Drying at 30–50°C, lyophilisation, flocculation by ferric chloride at 0.1–0.2% and encapsulation by entrapment in calcium alginate beads, agar, or polyacrylamide gel were efficient methods for recovery of STC of Ls14N1 and Bti from SSF extracts. Also, STC was precipitated with remained the mosquitocidal activity through adjusting the pH of the SSF liquid extract before centrifugation at pH 2–5 or after addition of acetone (200–400%), ethanol (400%) or acetic acid (2.5–4%) to SSF liquid extract of Ls14N1. On the other hand, STC of Bti showed a good recovery by centrifugation after adjusting SSF liquid extract at pH 4 followed by pH 3 and pH 5 or after the addition of ethanol (300%) or acetone (200–300%). This study contains simple and multiple methods for efficient recovery of STC of Bti and Ls14N1 grown under SSF. The choice of the best method depends on the availability of the necessary equipment as well as the cost of the treatment.  相似文献

Epoxide hydrolase (EC 3.3.2.3) purified from rat liver microsomes has been immobilized by covalent linking to dextran activated by imidazolyl carbamate groups, under mild conditions. Kappm values of free and dextran bound epoxide hydrolase toward benzo(a)pyrene-4,5-oxide were 0.5 and 0.35 μM respectively, while Vappmax was lowered from 300 to 120 nmol min?1mg?1protein. The activity lost upon coupling could not be restored by digestion of the support by dextranase (1,6-α-d-glucan 6-glucanohydrolase, EC 3.2.1.11) treatment. This fact, along with the similarity of the activation energy values for both native and bound epoxide hydrolase, indicated that steric hindrance effects due to the polymer support played only a minor role in this loss of activity. Evidences of changes in the conformation of epoxide hydrolase were obtained by a comparative study of u.v. circular dichroism and tryptophan fluorescence emission spectra of the native and dextran bound enzymes. On the other hand, the enzyme conjugate showed greater resistance than the free enzyme to thermal inactivation.  相似文献

Two simple and sensitive analytical assay methods using spectrophotometry and spectrofluorimetry techniques were developed for the estimation of succinylcholine chloride (SUC) in pharmaceutical preparations. The suggested methods are based on the formation of an ion pair complex formed between the drug and eosin Y spectrophotometrically (Method I), or the suppressive effect of succinylcholine on the native fluorescence property of eosin Y (Method II). The spectrophotometric method (Method I) involves measuring the absorbance of the complex between succinylcholine and eosin Y at 550 nm in Britton Robinson buffer of pH 3. However, the spectrofluorimetric method (Method II) involves measuring the quenching effect of the studied drug on the native fluorescence property of eosin Y at the same pH at 550 nm after excitation at 480 nm. The absorbance versus concentration of the drug is rectilinear over the range of 0.5 to 15 μg/ml. The formation constant was 3.5 × 104 and the Gibb's free energy change was ?2.5 × 104 J/mol. In Method II, the relative fluorescence intensity was directly proportional to SUC concentration over the range of 0.05 to 1 μg/ml. The proposed methods allowed a successful application to the estimation of succinylcholine ampoules. An explanation of the reaction pathway was postulated.  相似文献

There are two different muscle fiber types in haplotaxids. The pseudo-circomyarian type is typical of Haplotaxis gordioides and the flattened circomyarian type of Pelodrilus leruthi. The mechanisms of growth in fiber size and in fiber number of the two fiber types in the hindmost region of adult specimens have been studied ultrastructurally. The increase in length and girth of the muscle fiber is always the result of the insertion of new myofilaments in the peripheral zones of the muscle cells. The increase in the number of fibers seems to be due to division of differentiated muscle cells.  相似文献

The migration strategy of many capital breeders is to garner body stores along the flyway at distinct stopover sites. The rate at which they can fuel is likely to be strongly influenced by a range of factors, such as physiology, food availability, time available for foraging and perceived predation. We analysed the foraging behaviour and fuel accumulation of pink‐footed geese, an Arctic capital breeder, at their mid‐flyway spring stopover site and evaluated to what extent their behaviour and fuelling were related to physiological and external factors and how it differed from other stopovers along the flyway. We found that fuel accumulation rates of geese at the mid‐flyway site were limited by habitat availability rather than by digestive constraints. However, as the time available for foraging increased over the stopover season, geese were able to keep constant fuelling rate. Putting this in perspective, geese increased their daily net energy intake along the flyway corresponding to the increase in time available for foraging. The net energy intake per hour of foraging remained the same. Geese showed differences in their reaction to predators/disturbance between the sites, taking higher risks particularly at the final stopover site. Hence, perceived predation along the flyway may force birds to postpone the final fuel accumulation to the last stopover along the flyway. Flexibility in behaviour appears to be an important trait to ensure fitness in this capital breeder. Our findings are based on a new, improved method for estimating fuel accumulation of animals foraging in heterogeneous landscapes based on data obtained from satellite telemetry and habitat specific intake rates.  相似文献
