The DNA segments containing the ADR1 gene and a mutant allele, ADR1-5c, have been isolated by complementation of function in Saccharomyces cerevisiae. The ADR1 gene is required for synthesis of the glucose-repressible alcohol dehydrogenase (ADHII) when S. cerevisiae cells are grown on a nonfermentable carbon source, whereas the ADR1-5c allele allows ADHII synthesis even during glucose repression. A plasmid pool consisting of yeast DNA fragments isolated from a strain carrying the ADR1-5c allele was used to transform a strain containing the adr1-1 allele, which prevents ADHII depression. Transformants were isolated which expressed ADHII during glucose repression. A plasmid isolated from one of these transformants was shown to carry the ADR1-5c allele by its ability to integrate at the chromosomal adr1-1 locus. The wild-type ADR1 gene was isolated by colony hybridization, using the cloned ADR1-5c gene as a probe. The ADR1-5c and ADR1 DNA segments were indistinguishable by restriction site mapping. A partial ADR1 phenotype could be conferred by a 1.9-kilobase region, but DNA outside of this region appeared to be necessary for normal activation of ADHII by the ADR1 gene.  相似文献

Citrus leaves exposed to subfreezing temperatures evolved ethylene at rates between 0.1 and 38.3 microliters per kilogram fresh weight per hour whereas untreated leaves evolved between 0.01 and 0.50 microliter per kilogram fresh weight per hour. Leaves not injured by freezing temperatures did not abscise, and ethylene evolution was near normal after 2 days. Freeze-injured leaves continued evolving high ethylene levels 4 or 5 days subsequent to freeze injury, and many of the freeze-killed leaves abscised. Supportive evidence suggested freeze-induced ethylene was involved in freeze-induced leaf abscission; whereas freeze-inhibited abscission was not due to a lack of ethylene but injury to other metabolic systems necessary for abscission.  相似文献

Polyglycerolteichoic acid:glucosyl transferase (TAG transferase), one of the three enzymes involved in the pathway leading to the glucosylation of teichoic acid in Bacillus subtilis 168, was investigated. During the early stages of the growth of B. subtilis, TAG transferase is predominantly a soluble enzyme found in the cytoplasm. As growth proceeds, the amount of soluble enzyme decreases and the proportion of insoluble, membrane-bound TAG transferase increases, reaching a maximal value at the close of the logarithmic phase. Data are presented which suggest that these are two forms of the same enzyme, or have some common component. The effects of chaotropic agents, such as sodium trichloroacetate and sodium perchlorate, on the cytoplasmic membrane were also studied. These data show that such compounds can effectively remove the TAG transferase from the membrane in a water-soluble form. A study of some of the physical properties of this solubilized enzyme suggests that there is little difference between the two forms of the enzyme. Experiments are described which indicate that the glucosyl transfer by both the membrane-bound and soluble enzymes is not mediated by lipids.  相似文献

  1. Monthly quantitative samples of the invertebrate fauna (except Protozoa) in a small pond were taken over a period of three years. During one year, insect emergence traps were in operation. Water temperatures were recorded during the investigation.
  2. The most abundant organisms in the pond were Phaenocora typhlops, Limnodrilus hoffmeisteri and Chaoborus crystallinus. Certain species of Micro-Crustacea and Chironomidae were also abundant but these groups have been dealt with elsewhere (see p. 66). Dendrocoelum lacteum, Polycelis nigra, Helobdella stagnalis, Lumbriculus aariegatus, Tubifex tubifex, Planorbis complanatus, and Asellus meridianus also occurred in considerable though lower numbers; other species occurred in low numbers.
  3. The life-cycles and changes in numbers of the more numerous species are considered. The life-histories of D. lacteum, P. nigra, H. stagnalis, P. complanatus, A. meridianus are in agreement with published information. P. typhlops is seasonal in occurrence, being active from May to Sept. inclusive. Times of emergence of adults of various insect species agree with information available in the literature.
  4. The life-cycle of L. hoffmeisteri in the pond is as follows: young worms hatch in spring/summer and form the bulk of the population from April to July/Aug; they mature from Aug. onwards and breeding starts in earnest from Feb./March. The life-cycle of T. tubifex is as follows: breeding starts in Feb., recruitment of young takes place from April till June, and these start to mature in Nov./Dec. It is not certain if some animals which breed in the spring/early summer survive to breed the following year.
  5. The life-cycle of C. crystallinus appears to be as follows: first instars present from May to Oct., second instars from May to Dec., third instars from June till following Jan., fourth instars all the year round, pupae from May till Aug., and eggs from May to Sept. Adult emergence takes place from late April till mid-Sept.
  6. A six-week drought in Oct/beginning Nov. in the second year of the investigation caused considerable mortality in most species, but most survived with only a few exceptions.

A study was conducted to determine a physicochemical profile of a new deep-storage reservoir and to determine the influence of impoundment and thermal stratification in the reservoir on the physicochemical limnology of the parent river. The presence of thermal stratification from May through November caused the most significant change in water conditions. Of 23 parameters studied, 12 remained unchanged, 10 improved, and 2 deteriorated (Table VI).The greatest downstream changes in water conditions from those upstream from the reservoir were a decrease in temperature, an increase of ammonia, and the presence of hydrogen sulfide during the period of thermal stratification. Ammonia did not increase to a level considered to be toxic to aquatic species. It could, however, serve as a nutrient for certain species of plants and result in a change in community structure.Water tempeature downstream from the reservoir was always within the annual temperature range of the river upstream from the reservoir; however, the summer maximum in the tailrace was decreased to a temperature that could interfere wih the normal life cycle of many species.  相似文献

Mercuric ions, as well as organomercuric ions and cadmium ions, can inhibit deoxyribonucleic acid-mediated transformation in Bacillus subtilis 168 without decreasing the viability of the total population. Differences in the inhibition of transformation by mercuric ions are identifiable on a temporal and concentration dependence basis. Sensitivity to low concentrations (9.2 x 10(-8) M) appears early in the uptake of deoxyribonucleic acid before the transformed markers have become insensitive to deoxyribonuclease. Resistance to "low concentrations" of Hg(2+) is kinetically indistinguishable from the requirement for magnesium in the transformation process. This inactivation is not reversed by the mercury-binding compound glutathione. Sensitivity to mercuric ions at a higher concentration (5.52 x 10(-7) M) occurs after the donor deoxyribonucleic acid has become insensitive to deoxyribonuclease. These complex interactions between mercuric ions and the process of transformation are discussed.  相似文献
