The aquatic gastropod mollusc, Pyrgophorus coronatus, may perform an important role in the transmission of an emergent ocular pathology among fishes in Lake Apoyo, Nicaragua. This disease emerged after an introduction of tilapia (Oreochromis niloticus) and the subsequent loss of Chara sp. beds in the lake. We compared the mollusc population densities in three habitats (sandy/muddy substrates, rocks, and Chara vegetation) at varying depths (1.5, 10, 20, and 30 m) in two volcanic crater lakes in Nicaragua: Lake Apoyo and Lake Xiloa. where lower numbers of affected fishes were found and tilapia has not been introduced. Duplicate samples at 1.5 m depth were taken in each habitat monthly for a year, and triplicate samples for bathymetric analysis of snail populations were performed during August, 2005. Samples of fixed surface area were filtered in a 0.4 cm size screen and live snails were counted from each sample. The preferred snail habitat in both lakes, Chara beds, was vastly reduced in Lake Apoyo via consumption by introduced Nile tilapia (Oreochromis niloticus). Structureless sandy substrates (mean +/- standard error 3.1+/-1.3 ind/m2) had lower population densities than other habitats in Lake Xiloa (rocks 590.9+/-185.3 ind/m2; vegetation 3 686.5+/-698.2 ind/m2; ANOVA 1, p<0.01 in both cases) but this difference was attenuated in Lake Apoyo (sand 384.4+/-111.1 ind/m2; rocks 1 480.4+/-384.8 ind/m2: 0.0 1

Quorum-sensing (QS) signals (N-acyl homoserine lactones [AHLs]) were extracted and detected from five commercially produced vacuum-packed meat samples. Ninety-six AHL-producing bacteria were isolated, and 92 were identified as Enterobacteriaceae. Hafnia alvei was the most commonly identified AHL-producing bacterium. Thin-layer chromatographic profiles of supernatants from six H. alvei isolates and of extracts from spoiling meat revealed that the major AHL species had an Rf value and shape similar to N-3-oxo-hexanoyl homoserine lactone (OHHL). Liquid chromatography-mass spectrometry (MS) (high-resolution MS) analysis confirmed the presence of OHHL in pure cultures of H. alvei. Vacuum-packed meat spoiled at the same rate when inoculated with the H. alvei wild type compared to a corresponding AHL-lacking mutant. Addition of specific QS inhibitors to the AHL-producing H. alvei inoculated in meat or to naturally contaminated meat did not influence the spoilage of vacuum-packed meat. An extracellular protein of approximately 20 kDa produced by the H. alvei wild-type was not produced by the AHL-negative mutant but was restored in the mutant when complemented by OHHL, thus indicating that AHLs do have a regulatory role in H. alvei. Coinoculation of H. alvei wild-type with an AHL-deficient Serratia proteamaculans B5a, in which protease secretion is QS regulated, caused spoilage of liquid milk. By contrast, coinoculation of AHL-negative strains of H. alvei and S. proteamaculans B5a did not cause spoilage. In conclusion, AHL and AHL-producing bacteria are present in vacuum-packed meat during storage and spoilage, but AHL does not appear to influence the spoilage of this particular type of conserved meat. Our data indicate that AHL-producing H. alvei may induce food quality-relevant phenotypes in other bacterial species in the same environment. H. alvei may thus influence spoilage of food products in which Enterobacteriaceae participate in the spoilage process. 相似文献

1. How climatic changes affect migratory birds remains difficult to predict because birds use multiple sites in a highly interdependent manner. A better understanding of how conditions along the flyway affect migration and ultimately fitness is of paramount interest. 2. Therefore, we developed a stochastic dynamic model to generate spatially and temporally explicit predictions of stop-over site use. For each site, we varied energy expenditure, onset of spring, intake rate and day-to-day stochasticity independently. We parameterized the model for the migration of pink-footed goose Anser brachyrhynchus from its wintering grounds in Western Europe to its breeding grounds on Arctic Svalbard. 3. Model results suggested that the birds follow a risk-averse strategy by avoiding sites with comparatively high energy expenditure or stochasticity levels in favour of sites with highly predictable food supply and low expenditure. Furthermore, the onset of spring on the stop-over sites had the most pronounced effect on staging times while intake rates had surprisingly little effect. 4. Subsequently, using empirical data, we tested whether observed changes in the onset of spring along the flyway explain the observed changes in migration schedules of pink-footed geese from 1990 to 2004. In order to better understand the molecular and cellular determinants of tumor cell intravasation, our laboratory has generated a pair of congenic human HT-1080 fibrosarcoma variants (i.e. HT-hi/diss and HT-lo/diss) differing 50-100-fold in their ability to intravasate and disseminate. To investigate the molecular differences underlying the distinct dissemination capacities of these HT-1080 variants, we performed a comparative analysis of the cell surface proteomes of HT-hi/diss and HT-lo/diss. Cell membrane proteins were enriched by biotinylation and avidin precipitation and analyzed by tandem mass spectrometry employing multidimensional protein identification technology. By this approach, 47 cell surface-associated molecules were identified as differentially expressed between the HT-1080 intravasation variants. From these candidates, four targets (i.e. TIMP-2, NCAM-1, JAM-C, and tissue factor (TF)) were selected for further biochemical validation and in vivo functional verification. Western blot analysis of the cell surface enriched fractions confirmed the proteomic array data, demonstrating that, in vitro, TIMP-2 protein was increased in the HT-lo/diss variant, whereas NCAM-1, JAM-C, and TF levels were increased in the HT-hi/diss variant. Corresponding in vivo differences in levels of TIMP-2, JAM-C, and TF were demonstrated in primary tumors grown in the chick embryo. Finally, functional inhibition of one selected protein (i.e. TF) by small interfering RNA silencing or ligation with a function-blocking antibody significantly reduced HT-hi/diss intravasation, thus clearly implicating TF in the early steps of tumor cell dissemination. Overall, our cell surface proteomic analysis provides a powerful tool for identification of specific cell membrane molecules that contribute functionally to intravasation and metastasis in vivo. 相似文献

Groups of female and immature sperm whales live at low latitudes and show a stereotypical diving and foraging behavior with dives lasting about 45 min to depths of between 400 and 1200 m. In comparison, physically mature male sperm whales migrate to high latitudes where little is known about their foraging behavior and ecology. Here we use acoustic recording tags to study the diving and acoustic behavior of male sperm whales foraging off northern Norway. Sixty-five hours of tag data provide detailed information about the movements and sound repertoire of four male sperm whales performing 83 dives lasting between 6 and 60 min. Dives ranged in depth between 14 and 1860 m, with a median depth of 175 m, and 92% of the surfacings lasted less than 15 min. The four whales clicked for an average 91% (SD = 10) of the dive duration, where the first usual click was produced at depths ranging between 4 and 218 m and the last usual click at depths ranging between 1 and 1114 m. Echolocation buzzes, which are used as an indication of prey capture attempts, were emitted at depths between 17 and 1860 m, during both the descent and ascent phase of deep dives. The foraging behavior varied markedly with depth, with the timing and duration of prey capture attempts during shallow dives suggesting that the whales target more sparsely distributed prey. In contrast, deep dives involve frequent prey capture attempts and seem to target more dense food layers. The evidence of exploitation of different food layers, including epipelagic prey, is consistent with the hypothesis that male sperm whales may migrate to high latitudes to access a productive, multi-layered foraging habitat. 相似文献

Distribution of freshwater snails in the river Niger basin in Mali with special reference to the intermediate hosts of schistosomes 总被引:1,自引:1,他引:0
Snail surveys were carried out in various parts of Mali. All areas surveyed are part of the Niger basin being either affluents
or irrigation schemes fed by this river. The snail species present varied greatly between areas. The following potential hosts
of schistosomes were recorded: Biomphalaria pfeifferi, Bulinus truncatus, B. globosus, B. umbilicatus, B. forskalii and B. senegalensis.
In the large irrigation schemes, i.e. ‘Office du Niger’ and Baguinéda, only B. pfeifferi and B. truncatus appear to be intermediate hosts. Snail distribution appeared to some extent to be focal and high snail densities appeared
to be associated with human water contact activities, which apparently create favourable biotopes for the snails. This is
probably due to an alteration of the vegetation and an increase of the trophic status of the site by contamination with food
remnants and other debris. The larger irrigation canals or lakes in these schemes play an important role in the transmission
of human schistosomes and transmission appears to be very focal in these habitats. Infected snails are almost exclusively
found in well-defined human water contact sites (WCS). Local infection rates with schistosomes were often high (i.e. up to
27% in B. pfeifferi).
In urban areas (i.e. Bamako), transmission patterns are more variable. In Bamako schistosome-infected B. truncatus were found in the Niger river. A number of smaller semi-permanent or permanent streams are very important transmission sites,
and schistosome infections were recorded from B. pfeifferi, B. truncatus and B. globosus. Schistosome infection rates in B. pfeifferi were often high (up to 30%).
In a new lake at Sélingué, B. truncatus was found to be widely distributed only about a year and a half after the dam was constructed, and in some sites schistosome

The carbohydrate content (starch, glucose and sucrose) in tomato plants grown in air Containing 0.04, 0.10, 0.15, 0.22, 0.32, or 0.50 vol. per cent CO2 was studied at 2 hours’ intervals over a period of 24 hours. The highest starch content was found at 0.22 vol. per cent CO2, while the highest content of soluble sugars were reached at a concentration of 0.10 vol. per cent CO2. Stochastic models of ion channels have been based largely on Markov theory where individual states and transition rates must be specified, and sojourn-time densities for each state are constrained to be exponential. This study presents an approach based on random-sum methods and alternating-renewal theory, allowing individual states to be grouped into classes provided the successive sojourn times in a given class are independent and identically distributed. Under these conditions Markov models form a special case. The utility of the approach is illustrated by considering the effects of limited time resolution (modelled by using a discrete detection limit, xi) on the properties of observable events, with emphasis on the observed open-time (xi-open-time). The cumulants and Laplace transform for a xi-open-time are derived for a range of Markov and non-Markov models; several useful approximations to the xi-open-time density function are presented. Numerical studies show that the effects of limited time resolution can be extreme, and also highlight the relative importance of the various model parameters. The theory could form a basis for future inferential studies in which parameter estimation takes account of limited time resolution in single channel records. Appendixes include relevant results concerning random sums and a discussion of the role of exponential distributions in Markov models. 相似文献
