Barriers to migration are numerous in stream environments and can occur from anthropogenic activities (such as dams and culverts) or natural processes (such as log jams or dams constructed by beaver (Castor canadensis)). Identification of barriers can be difficult when obstructions are temporary or incomplete providing passage periodically. We examine the effect of several small irrigation diversion dams on the recent migration rates of steelhead (Oncorhynchus mykiss) in three tributaries to the Methow River, Washington. The three basins had different recent migration patterns: Beaver Creek did not have any recent migration between sites, Libby Creek had two-way migration between sites and Gold Creek had downstream migration between sites. Sites with migration were significantly different from sites without migration in distance, number of obstructions, obstruction height to depth ratio and maximum stream gradient. When comparing the sites without migration in Beaver Creek to the sites with migration in Libby and Gold creeks, the number of obstructions was the only significant variable. Multinomial logistic regression identified obstruction height to depth ratio and maximum stream gradient as the best fitting model to predict the level of migration among sites. Small irrigation diversion dams were limiting population interactions in Beaver Creek and collectively blocking steelhead migration into the stream. Variables related to stream resistance (gradient, obstruction number and obstruction height to depth ratio) were better predictors of recent migration rates than distance, and can provide important insight into migration and population demographic processes in lotic species.  相似文献

Laboratory and field investigations were made in an endemic focus of cutaneous leishmaniasis in Khartoum State, Sudan, to evaluate the effects of permethrin-impregnated curtains on the human-biting activity, nocturnal activity and resting behaviour of the vector sandfly Phlebotomus papatasi (Diptera: Psychodidae) indoors. Laboratory bioassays showed that curtains impregnated with 0.5, 1.0 or 1.5 g/m2 permethrin all gave 100% mortality within 24h of exposure of P. papatasi for 3 min. Under natural field conditions, the biting activity indoors and the resting density of P. papatasi were significantly reduced (P<0.001 and P=0.036, respectively) in rooms provided with permethrin-impregnated curtains as compared to control rooms left without curtains or fitted with unimpregnated curtains. No significant difference was found between the numbers of nocturnally active P. papatasi collected in rooms provided with impregnated curtains and rooms left without curtains or provided with unimpregnated curtains (P=0.377). Evidently P. papatasi was not repelled by these doses of permethrin on curtains, but the survival rate of sandflies collected from test rooms provided with permethrin-impregnated curtains was significantly reduced (P=0.036). We conclude that use of permethrin-impregnated curtains may provide a good control method for P. papatasi and other endophilic and/or endophagic sandfly vectors of leishmaniasis.  相似文献

Cilia and flagella are required for cell motility and sensing the external environment and can vary in both length and stability. Stable flagella maintain their length without shortening and lengthening and are proposed to “lock” at the end of growth, but molecular mechanisms for this lock are unknown. We show that CEP164C contributes to the locking mechanism at the base of the flagellum in Trypanosoma brucei. CEP164C localizes to mature basal bodies of fully assembled old flagella, but not to growing new flagella, and basal bodies only acquire CEP164C in the third cell cycle after initial assembly. Depletion of CEP164C leads to dysregulation of flagellum growth, with continued growth of the old flagellum, consistent with defects in a flagellum locking mechanism. Inhibiting cytokinesis results in CEP164C acquisition on the new flagellum once it reaches the old flagellum length. These results provide the first insight into the molecular mechanisms regulating flagella growth in cells that must maintain existing flagella while growing new flagella.  相似文献

Possible life cycle of some ancient plectambonitoids (order Strophomenida) from the Middle Ordovician of Russia is reconstructed based on the well-preserved specimens composing the ontogenetic series. Four regions may be distinguished on their shell surface: protegulum, brephic shell, neanic shell and adult shell. The posterior margin of ventral protegulum bears pedicle sheath, which is a tubular outgrowth with a 40-μm-wide aperture at the distal end. The protegulum and brephic shell have common type of microstructure that possibly is spherular; the neanic and adult shells are fibrous. The strophomenide ontogeny possibly was similar to that of recent discinides. The strophomenide life cycle possibly included the planktotrophic juvenile stage; the protegulum and brephic shell were formed in the water column. The aperture of the pedicle sheath was possibly used as an anal opening of the floating juvenile and as an attachment organ during the settlement; at early adult stages, the sheath erased, the anus closed, and the animal started to lie on the ventral valve. The origin of the order Strophomenida and its relative groups is possibly connected with the loss of the pedicle lobe; judging by some strophomenide morphological features, true pedicle was present in the strophomenide ancestors. The tubes on the ventral umbones of strophomenides and billingsellides are not homologous as pedicle sheaths of strophomenides are formed at the planktotrophic swimming stage, and the tubes surrounding the pedicles of billingsellides were formed by deltidial plates of almost adult shell after settling.  相似文献

To increase our understanding of peptide–water interactions, we are simulating the behavior of water molecules in the intermolecular channels of [Phe4Val6]antamanide dododecahydrate crystals. There is good overall agreement between the positions predicted using two alternative potential functions and those that have been observed by x-ray diffraction. Mitochondrial dynamics and mitophagy are constitutive and complex systems that ensure a healthy mitochondrial network through the segregation and subsequent degradation of damaged mitochondria. Disruption of these systems can lead to mitochondrial dysfunction and has been established as a central mechanism of ischemia/reperfusion (I/R) injury. Emerging evidence suggests that mitochondrial dynamics and mitophagy are integrated systems; however, the role of this relationship in the context of I/R injury remains unclear. To investigate this concept, we utilized primary cortical neurons isolated from the novel dual-reporter mitochondrial quality control knockin mice (C57BL/6-Gt(ROSA)26Sortm1(CAG-mCherry/GFP)Ganl/J) with conditional knockout (KO) of Drp1 to investigate changes in mitochondrial dynamics and mitophagic flux during in vitro I/R injury. Mitochondrial dynamics was quantitatively measured in an unbiased manner using a machine learning mitochondrial morphology classification system, which consisted of four different classifications: network, unbranched, swollen, and punctate. Evaluation of mitochondrial morphology and mitophagic flux in primary neurons exposed to oxygen-glucose deprivation (OGD) and reoxygenation (OGD/R) revealed extensive mitochondrial fragmentation and swelling, together with a significant upregulation in mitophagic flux. Furthermore, the primary morphology of mitochondria undergoing mitophagy was classified as punctate. Colocalization using immunofluorescence as well as western blot analysis revealed that the PINK1/Parkin pathway of mitophagy was activated following OGD/R. Conditional KO of Drp1 prevented mitochondrial fragmentation and swelling following OGD/R but did not alter mitophagic flux. These data provide novel evidence that Drp1 plays a causal role in the progression of I/R injury, but mitophagy does not require Drp1-mediated mitochondrial fission.Subject terms: Mitophagy, Mechanisms of disease  相似文献
