Course intragastric administration of ultralow doses of human gamma-interferon antibodies (ULD anti-IFN-gamma) to intact mice resulted in an increase of endogenous IFN-gamma production by the animal lymphocytes. Oral prophylactic administration of ULD anti-IFN-gamma significantly lowered the influenza virus concentration in the animal lungs at the initial stage of the aerogenous infection: in 2 (p = 0.05) and 3 (p = 0.07) days after the contamination. The therapeutic antiviral effect of ULD anti-IFN-gamma in mice with influenza was evident from a significant decrease of the influenza virus concentration in the lungs of the animals on the 4th (p = 0.05) and 5th (p = 0.07) days after the contamination. The antiviral effect of ULD anti-IFN-gamma after the prophylactic and therapeutic use is likely provided by induction of endogenous IFN-gamma.  相似文献

Sterol glycosyltransferases catalyze the synthesis of diverse glycosterols in plants. Withania somnifera is a medically important plant, known for a variety of pharmacologically important withanolides and their glycosides. In this study, a novel 27beta-hydroxy glucosyltransferase was purified to near homogeneity from cytosolic fraction of W. somnifera leaves and studied for its biochemical and kinetic properties. The purified enzyme showed activity with UDP-glucose but not with UDP-galactose as sugar donor. It exhibited broad sterol specificity by glucosylating a variety of sterols/withanolides with beta-OH group at C-17, C-21 and C-27 positions. It transferred glucose to the alkanol at C-25 position of the lactone ring, provided an alpha-OH was present at C-17 in the sterol skeleton. A comparable enzyme has not been reported earlier from plants. The enzyme is distinct from the previously purified W. somnifera 3beta-hydroxy specific sterol glucosyltransferase and does not glucosylate the sterols at C-3 position; though it also follows an ordered sequential bisubstrate reaction mechanism, in which UDP-glucose and sterol are the first and second binding substrates. The enzyme activity with withanolides suggests its role in secondary metabolism in W. somnifera. Results on peptide mass fingerprinting showed its resemblance with glycuronosyltransferase like protein. The enzyme activity in the leaves of W. somnifera was enhanced following the application of salicylic acid. In contrast, it decreased rapidly on exposure of the plants to heat shock, suggesting functional role of the enzyme in biotic and abiotic stresses.  相似文献

Mitochondrial mRNAs in kinetoplastids require extensive U-insertion/deletion editing that progresses 3′-to-5′ in small blocks, each directed by a guide RNA (gRNA), and exhibits substrate and developmental stage-specificity by unsolved mechanisms. Here, we address compositionally related factors, collectively known as the mitochondrial RNA-binding complex 1 (MRB1) or gRNA-binding complex (GRBC), that contain gRNA, have a dynamic protein composition, and transiently associate with several mitochondrial factors including RNA editing core complexes (RECC) and ribosomes. MRB1 controls editing by still unknown mechanisms. We performed the first next-generation sequencing study of native subcomplexes of MRB1, immunoselected via either RNA helicase 2 (REH2), that binds RNA and associates with unwinding activity, or MRB3010, that affects an early editing step. The particles contain either REH2 or MRB3010 but share the core GAP1 and other proteins detected by RNA photo-crosslinking. Analyses of the first editing blocks indicate an enrichment of several initiating gRNAs in the MRB3010-purified complex. Our data also indicate fast evolution of mRNA 3′ ends and strain-specific alternative 3′ editing within 3′ UTR or C-terminal protein-coding sequence that could impact mitochondrial physiology. Moreover, we found robust specific copurification of edited and pre-edited mRNAs, suggesting that these particles may bind both mRNA and gRNA editing substrates. We propose that multiple subcomplexes of MRB1 with different RNA/protein composition serve as a scaffold for specific assembly of editing substrates and RECC, thereby forming the editing holoenzyme. The MRB3010-subcomplex may promote early editing through its preferential recruitment of initiating gRNAs.  相似文献

The agonist binding site of ATP-gated P2X receptors is distinct from other ATP-binding proteins. Mutagenesis on P2X(1) receptors of conserved residues in mammalian P2X receptors has established the paradigm that three lysine residues, as well as FT and NFR motifs, play an important role in mediating ATP action. In this study we have determined whether cysteine substitution mutations of equivalent residues in P2X(2) and P2X(4) receptors have similar effects and if these mutant receptors can be regulated by charged methanethiosulfonate (MTS) compounds. All the mutants (except the P2X(2) K69C and K71C that were expressed, but non-functional) showed a significant decrease in ATP potency, with >300-fold decreases for mutants of the conserved asparagine, arginine, and lysine residues close to the end of the extracellular loop. MTS reagents had no effect at the phenylalanine of the FT motif, in contrast, cysteine mutation of the threonine was sensitive to MTS reagents and suggested a role of this residue in ATP action. The lysine-substituted receptors were sensitive to the charge of the MTS reagent consistent with the importance of positive charge at this position for coordination of the negatively charged phosphate of ATP. At the NFR motif the asparagine and arginine residues were sensitive to MTS reagents, whereas the phenylalanine was either unaffected or showed only a small decrease. These results support a common site of ATP action at P2X receptors and suggest that non-conserved residues also play a regulatory role in agonist action.  相似文献

Two-dimensional electrophoresis was used to separate proteins from crude extracts of pea (Pisum sativum L.) leaves, and thus isolated proteins were subjected to Western blot analysis with monoclonal antibodies against PY20 phosphotyrosine polypeptides. This analysis revealed 44 polypeptides phosphorylated on tyrosine residues. Phosphorylation of some of these proteins was changed under the action of epibrassinolide. Some of these polypeptides were identified by means of MALDI-TOF MS analysis. The results indicate that eight of these proteins belong to the Calvin cycle enzymes, namely, the isoforms of Rubisco large and small subunits, fructose-1,6-phosphate aldolases 1 and 2, and the precursor of α-subunit of Rubisco-binding protein. The observed changes in phosphorylation of these proteins may partly explain the effects of brassinosteroids on photosynthesis. The tyrosine phosphorylation sites were identified in silico for the fragments of polypeptides examined.  相似文献

Eifelian ammonoids are described for the first time from the Central Kyzylkum Desert (Uzbekistan, Navoiy Region, Aristantau Mountains). The ammonoid assemblage includes the typical Eifelian genera Fidelites, Subanarcestes (a new species S. aristanensis Nikolaeva sp. nov. is described), and Cabrieroceras, found in association with brachiopods, conodonts, and dacryoconarids. The ammonoid assemblage is similar to that from the Chote? beds of Barrandien and their equivalents in the Urals, Salair, Germany, and Morocco (Subanarcestes macrocephalus Zone), suggesting a connection between the Eifelian Kyzylkum Basin, which was part of the Turkestan Paleoocean, and European, North African, Siberian, and Uralian basins. The ammonoids studied are not associated with black shale and apparently existed in normal marine environments.  相似文献

Formation of the Escherichia coli division septum is catalyzed by a number of essential proteins (named Fts) that assemble into a ring-like structure at the future division site. Several of these Fts proteins are intrinsic transmembrane proteins whose functions are largely unknown. Although these proteins appear to be recruited to the division site in a hierarchical order, the molecular interactions underlying the assembly of the cell division machinery remain mostly unspecified. In the present study, we used a bacterial two-hybrid system based on interaction-mediated reconstitution of a cyclic AMP (cAMP) signaling cascade to unravel the molecular basis of septum assembly by analyzing the protein interaction network among E. coli cell division proteins. Our results indicate that the Fts proteins are connected to one another through multiple interactions. A deletion mapping analysis carried out with two of these proteins, FtsQ and FtsI, revealed that different regions of the polypeptides are involved in their associations with their partners. Furthermore, we showed that the association between two Fts hybrid proteins could be modulated by the coexpression of a third Fts partner. Altogether, these data suggest that the cell division machinery assembly is driven by the cooperative association among the different Fts proteins to form a dynamic multiprotein structure at the septum site. In addition, our study shows that the cAMP-based two-hybrid system is particularly appropriate for analyzing molecular interactions between membrane proteins.  相似文献

Expression of virulence-associated genes in Bordetella pertussis is under the control of the pleiotropic regulator BvgA. Although previous studies have identified recognition sequences for BvgA in several promoter regions, their structures have not been clearly characterized. We show that the BvgA binding sites within the bvgp(1) and cyaA promoters consist of inverted repeats and suggest that inverted-repeat motifs may represent the recognition elements for DNA-BvgA interaction.  相似文献
