Methotrexate (MTX) exerts an anti-inflammatory effect via its metabolite adenosine, which activates adenosine receptors. The A3 adenosine receptor (A3AR) was found to be highly expressed in inflammatory tissues and peripheral blood mononuclear cells (PBMCs) of rats with adjuvant-induced arthritis (AIA). CF101 (IB-MECA), an A3AR agonist, was previously found to inhibit the clinical and pathological manifestations of AIA. The aim of the present study was to examine the effect of MTX on A3AR expression level and the efficacy of combined treatment with CF101 and MTX in AIA rats. AIA rats were treated with MTX, CF101, or both agents combined. A3AR mRNA, protein expression and exhibition were tested in paw and PBMC extracts from AIA rats utilizing immunohistochemistry staining, RT-PCR and Western blot analysis. A3AR level was tested in PBMC extracts from patients chronically treated with MTX and healthy individuals. The effect of CF101, MTX and combined treatment on A3AR expression level was also tested in PHA-stimulated PBMCs from healthy individuals and from MTX-treated patients with rheumatoid arthritis (RA). Combined treatment with CF101 and MTX resulted in an additive anti-inflammatory effect in AIA rats. MTX induced A2AAR and A3AR over-expression in paw cells from treated animals. Moreover, increased A3AR expression level was detected in PBMCs from MTX-treated RA patients compared with cells from healthy individuals. MTX also increased the protein expression level of PHA-stimulated PBMCs from healthy individuals. The increase in A3AR level was counteracted in vitro by adenosine deaminase and mimicked in vivo by dipyridamole, demonstrating that receptor over-expression was mediated by adenosine. In conclusion, the data presented here indicate that MTX induces increased A3AR expression and exhibition, thereby potentiating the inhibitory effect of CF101 and supporting combined use of these drugs to treat RA. 相似文献

On the basis of its physiological characeristics and DNA-DNA homology data, the strain was designated as Clostridium thermosulfurogenes (DSM 3896). The interactions between microsclerotia (ms) of the fungal plant pathogen Verticillium dahliae and the mycoparasite Talaromyces flavus were followed in soil and on agar medium. Germinability of ms, which had been incubated for 14 days in soil treated with 0.5% of a T. flavus ‐ wheat bran preparation, decreased from 84% to 17%, as compared with 81% and 74% in untreated soil and in soil treated with a sterilized biocontrol preparation respectively. Germinability of ms which had been buried in treated soil for 4 days decreased to 70%, all ms being parasitized by T. flavus. Upon transfer of the ms to untreated soil for 10 more days, germinability decreased further to 20%, indicating that T. flavus continued to parasitize sclerotia in the untreated soil. Scanning electron micrographs showed heavy fungal colonization and typical T. flavus conidia on the surface of the ms buried in the treated soil, but not in control soils. Transmission electron micrographs of ms incubated with T. flavus on agar revealed parsitism involving invasion of some host cells by means of small penetration pegs; the host cell walls were mainly lysed at their site of contact with the parasite hyphal tips. Further colonization of the ms cells occurred simultaneously with the degradation of the invaded host cell contents, rather than the cell walls. Mycoparasitism of V. dahliae ms by T. flavus hyphae may be involved in the biological control of verticillium wilt disease. 相似文献

The coronavirus pandemic has caused a global public health crisis with an unprecedented shutdown of major establishments and non-emergency services. Disruptions across the country in dental hospitals led to challenges in addressing patient’s dental complaints. The present study aimed to analyse the difference in the pattern of the Outpatient dental visits during the pandemic period in comparison to that of the pre-pandemic period in a tertiary care hospital. This retrospective study was carried out by retrieving the patient data from the diagnostic register of the Department of Oral Medicine and Radiology for a period of one year. The data that was retrieved from 24th September 2019 to 23rd March 2020 was categorized under the pre-pandemic period and the data from 24th March 2020 to 24th September 2020 was grouped under the pandemic period. Patient data regarding the age, gender and clinical diagnosis was recorded and categorized under three main categories: “Emergency”, “urgent” and “elective”. 7550 patients during pre-pandemic period and 4035 patients during the pandemic period visited the dental hospital during the study period. Under the “emergency dental care” category, during the pre-pandemic period, majority of the cases reported of acute dental pain (71.0%) followed by cellulitis and space infection (20.1%) and maxillofacial trauma (8.7%). During the pandemic period, acute dental pain consisted of all emergency visits (n = 307). The proportion of emergency dental visits during the pandemic were significantly higher than the pre-pandemic period. Elective dental visits were significantly higher during pre-pandemic period in comparison to the pandemic period. (P < 0.001). There was a notable change in the outpatient trend of the dental visits during the COVID-19 pandemic in comparison to the pre-pandemic period. Emergency dental services were utilized at a higher rate during the pandemic period in comparison to the pre-pandemic counterpart. 相似文献
