Fertilization of an egg by a spermatozoon sets the stage for mammalian development. Viable sperm are a prerequisite for successful fertilization and beyond. Spermatozoa have a unique cell structure where haploid genomic DNA is located in a tiny cytoplasmic space in the head, mitochondria in the midpiece and then the tail, all enclosed by several layers of membrane. Proteins in sperm play vital roles in motility, capacitation, fertilization, egg activation and embryo development. Molecular defects in these proteins are associated with low fertility or in some cases, infertility. This review will first summarize genesis, molecular anatomy and physiology of spermatozoa, fertilization, embryogenesis and then those proteins playing important roles in various aspects of sperm physiology.  相似文献

In higher plants, ascorbate peroxidase (APX; EC 1.11.1.11), the major H2O2-scavenging enzyme, occurs in several distinct isoenzymes that are localized in cytosol and various cell organelles. Here, we have purified and characterized an APX from the soluble fraction of plastids of non-photosynthetic tobacco BY-2 cells. The plastidic APX was a monomer with a molecular weight of 34 000. The enzymatic properties of the plastidic APX, including the rapid inactivation by H2O2 in ascorbate-depleted medium, were highly comparable with those of the chloroplastic stromal APX of spinach and tea leaves. However, the other chloroplastic APX isoenzyme, the thylakoid-membrane bound APX, was not detected in the plastids of the BY-2 cells. The N-terminal amino acid sequence of the plastidic APX was completely identical with the deduced amino acid sequence of a previously identified cDNA sequence of tobacco chloroplastic APX. When a green fluorescence protein gene tagged with the chloroplast-targeting signal sequence of APX was expressed in the BY-2 cells, the fluorescence protein exclusively localized into plastids, and not into mitochondria. We conclude that plastidic APX in non-photosynthetic tissues is the same as the chloroplastic APX that occurs in leaves.  相似文献

Male urinary lipocalin family proteins, practically odorant-binding proteins but also could be pheromones by themselves, in rodents act as a shuttle for chemosignal communication and facilitate delivery of the signals for access to congeners. However, presence of this protein in urine of female rodents has not yet been reported. Therefore, the present investigation was carried out to find if lipocalin family protein is present in the urine of female house rat and, if so, to find whether its expression differs between the phases in the estrous cycle. The rat urinary protein was separated in single dimensional gel electrophoresis. A 14.5 kDa lipocalin protein appeared in the urine prominently during the estrus and metestrus phases compared to proestrus and diestrus phases. The expression of this protein in the urine was very low in ovariectomized rats. MALDI-TOF/MS analysis affirmed the 14.5 kDa protein as a lipocalin family protein. Analysis adopting bio-informatics tools further proved the protein as a lipocalin family member. Thus, this study for the first time demonstrated the presence of a lipocalin family protein in the urine of a female rodent and it was highly expressed during estrus phase. This lipocalin protein in female rat urine may facilitate a chemosignal function independently of a pheromone or in association with a specific pheromone.  相似文献

Mating of potato tuber moth, Phthorimaea operculella (Zeller) (Lepidoptera: Gelechiidae), was investigated in relation to the dispersal of males in laboratory and field trials. The effect of stimulating the flight of males to light sources in a large cage on their mating ability was estimated for three age groups, and compared with similar estimates for confined moths. Although the mating of males declined with ages of up to 15 days, simulated dispersal had no effect on subsequent mating when the males were paired with virgin females. The dispersal of male moths was also categorised by the initial flight activity of untethered moths to a light source. Scores for poor, moderate, and good flight provided a repeatable measure of initial male flight activity, but the degree of activity was not related to their subsequent mating ability. In the field, virgin female potato tuber moths were tethered at various distances from the edge of isolated potato crops and then dissected to determine their mating status. Female mating frequency averaged 75% at the crop margin, remained above 50% up to 200 m, and then declined to 19% at 360 m from the margin. Derivation of the mating probability for an individual male potato tuber moth confirmed earlier work by other researchers that has indicated a tendency for dispersal prior to mating, and that males retain their ability to mate as they disperse from a crop. The influence of dispersal and mating on gene flow between crops, and its potential effects on refuge size required to minimise the development of resistance to Bt transgenic potato crops was examined.  相似文献

EVA (http://cubic.bioc.columbia.edu/eva/) is a web server for evaluation of the accuracy of automated protein structure prediction methods. The evaluation is updated automatically each week, to cope with the large number of existing prediction servers and the constant changes in the prediction methods. EVA currently assesses servers for secondary structure prediction, contact prediction, comparative protein structure modelling and threading/fold recognition. Every day, sequences of newly available protein structures in the Protein Data Bank (PDB) are sent to the servers and their predictions are collected. The predictions are then compared to the experimental structures once a week; the results are published on the EVA web pages. Over time, EVA has accumulated prediction results for a large number of proteins, ranging from hundreds to thousands, depending on the prediction method. This large sample assures that methods are compared reliably. As a result, EVA provides useful information to developers as well as users of prediction methods.  相似文献

Coronary Artery Disease (CAD) is responsible for most of the deaths in patients with cardiovascular diseases. Diagnostic coronary angiography analysis offers an anatomical knowledge of the severity of the stenosis. The functional or physiological significance is more valuable than the anatomical significance of CAD. Clinicians assess the functional severity of the stenosis by resorting to an invasive measurement of the pressure drop and flow. Hemodynamic parameters, such as pressure wire assessment fractional flow reserve (FFR) or Doppler wire assessment coronary flow reserve (CFR) are well-proven techniques to evaluate the physiological significance of the coronary artery stenosis in the cardiac catheterization laboratory. Between the two techniques mentioned above, the FFR is seen as a very useful index. The presence of guide wire reduces the coronary flow which causes the underestimation of pressure drop across the stenosis which leads to dilemma for the clinicians in the assessment of moderate stenosis. In such condition, the fundamental fluid mechanics is useful in the development of new functional severity parameters such as pressure drop coefficient and lesion flow coefficient. Since the flow takes place in a narrowed artery, the blood behaves as a non-Newtonian fluid. Computational fluid dynamics (CFD) allows a complete coronary flow simulation to study the relationship between the pressure and flow. This paper aims at explaining (i) diagnostic modalities for the evaluation of the CAD and valuable insights regarding FFR in the evaluation of the functional severity of the CAD (ii) the role of fluid dynamics in measuring the severity of CAD.  相似文献
