The amino terminal sequences of the 4 caseins synthesized by translation of ovine mammary mRNAs in a wheat germ cell-free system have been investigated by automated Edman degradation. The 3 “calcium-sensitive” caseins (αs1, αs2 and β) and κ-casein were synthesized as precaseins with amino terminal hydrophobic extensions of 15 and 21 amino acid residues respectively, resembling “signal peptides” of other secretory proteins. The extra pieces of the 4 caseins, which start with a methionyl residue, end with an alanyl residue which may be one of the signals recognized by the mammary membrane-bound enzyme responsible for the specific cleavage of precaseins. The amino terminal extensions of αs1, αs2 and β-caseins show a high degree of homology suggesting that they have derived from a common ancestor.  相似文献

Oxalacetate and glyoxylate are each weak inhibitors of NADP+-specific isocitrate dehydrogenase (threo-DS-isocitrate:NADP+ oxidoreductase (decarboxylating), EC 1.1.1.42)9 Together, however, they act in a concerted manner and strongly inhibit the enzyme. The rates of formation and dissociation of the enzyme inhibitor complex, and the rate of formation and the stability of the aldol condensation product of oxalacetate and glyoxylate, oxalomalate, were examined. The data obtained do not support the often suggested possibility that oxalomalate, per se, formed non-enzymatically in isocitrate dehydrogenase assay mixtures containing oxalacetate and glyoxylate, is responsible for the observed inhibition of the enzyme. Rather, the data presented in this communication suggest that oxalacetate binds to the enzyme first, and that the subsequent binding of glyoxylate leads to the formation of a catalytically inactive enzyme-inhibitor complex.  相似文献

Two studies were conducted to synchronize breeding in cattle using PGF2α and LHRH/FSHRH analogs. In the first study, 60 mature lactating Angus cows were assigned at random to 4 treatment groups: saline and saline (SS); 30 mg PGF2α tham salt + saline (PS); saline + 2 mg D-ala6-des-GlyNH210 LHRH/FSHRH ethylamide (D-ala6) (SA); 30 mg PGF2α tham salt + 2 mg D-ala6 (PA). The first letter of the two-letter code for each group always indicates a dual injection at an 11-day interval. PGF2α or saline was administered intramuscular (im) twice at an 11 day interval. D-ala6 or saline was administered 48 hr after PGF2α treatment. In the SA group, the D-ala6 was administered at first signs of estrus, and cows were then artificially inseminated (AI) 24 hr later. All cows in the PS group were inseminated 72 hr after the second PGF2α injection. In the SS group, cows were inseminated 24 hr after first signs of estrus. An additional 6 mature lactating Angus cows were added equally to the PS and PA groups to evaluate changes in serum LH. The percent calf crops was: SS = 40% (frsol|6/15); PS = 47% (715); SA = 47% (715); PA = 53% (815). In the second study, 51 mature lactating Angus cows and 39 Holstein heifers were assigned at random to 3 treatment groups: saline + saline (SS); 33.4 mg PGF2α tham salt + saline (PS); 33.4 mg PGF2α tham salt + 1 mg D-leu6-des GlyNH210 LHRH/FSHRH ethylamide (D-leu6) (PL). PGF2α tham salt or saline was administered im twice at an 11 day interval. D-leu6 or saline was administered 68 hr following the second PGF2α treatment. Cows pretreated with PGF2α were inseminated 80 hr after the second PGF2α injection. In the SS group, cows were administered saline at the time of natural estrus and were artificially inseminated 12 hr later. Calving percent to the first AI was SS = 70% (2130); PS = 53% (1630) and PL = 40% (1230). An additional 10 mature lactating Angus cows were used to evaluate changes in serum LH. Five of the cows were assigned to the PS treatment and five to the PL treatment. Sequential blood samples were collected to monitor serum LH levels. Using the Chi-square test, there were no significant differences between calving percentages of the control and PGF2α treated cows in either study. These results indicate that the PGF2α treatments were successful in timed artificial insemination of cows without detection of estrus. The LHRH/FSHRH analogs did not improve the conception even though they appear to induce a pituitary release of LH simultaneously in all cows within 1 hr of treatment.  相似文献

Summary Deficient limb buds composed of prospective stylopod and autopod are able to regulate the missing intercalary zeugopod, the origin of which was investigated by heterospecific quail/chick recombinants. The associations of quail prospective autopod and chick prospective stylopod failed to regulate. The reverse combination of chick prospective autopod grafted onto a quail prospective stylopod gave rise to a three-segmented limb. In 13 out of 16 cases the regulated zeugopod was made up of both chick and quail cells. Chick cells were located predominantly along the postaxial half of the zeugopod, while the quail cells made up most of its preaxial half. In two cases, the intercalary zeugopod consisted exclusively of chick cells originating from the tip and in one case of quail cells originating from the base.These results demonstrate that during the regulative processes, the prospective values of some of the original stylopodial and autopodial cells have been shifted along the proximo-distal axis, towards the expression of more distal as well as of more proximal structures.Heteropolar stylo-autopodial or zeugo-autopodial recombinants, in which the proximo-distal axis of the base was reversed with respect to that of the tip, were unable to regulate the pattern defects and thus revealed the importance of concordant p-d polarity for regulative processes to take place between abutted tissues.  相似文献

Summary Hyperaccumulators (here defined as species containing at least 500 μg/g dryweight of either copper or cobalt in leaves), are of interest for the fields of mineral exploration and phytochemistry. Reported hyperaccumulation of copper inAeolanthus biformifolius and the presence of two other species on copper/cobalt mineralization in Shaba (Za?re) led to a survey of these elements in 49 species of the African genusAeolanthus Mart.A. biformifolius appears to be also a hyperaccumulator of Co (2520 μg/g in leaves, 4300 μg/g in corms). Cobalt levels for most species ofAeolanthus were considerably above typical values for phanerogams and indicate the favorable potential of the genus for further study.  相似文献

The prognosis for patients who undergo surgery for stage IIB malignant melanoma is poor. Animal studies have suggested that BCG and tumour cell vaccines given together may provide effective immunotherapy. To assess the effectiveness of this treatment 15 patients with stage IIB malignant melanoma who had their tumour excised were studied. Seven were treated conservatively, and eight were vaccinated with BCG and autologous irradiated cells. Three vaccinated patients suffered widespread recurrence within three months. All four vaccinated patients who suffered a recurrence within the first year died, while none of the three controls with recurrent disease died. In view of this alarming trend the trial was stopped after a year. BCG and the tumour cells may have enhanced the tumour growth, although there was no apparent reason for this. The results of uncontrolled or unrandomised trials that have suggested that this treatment is beneficial should be treated with scepticism.  相似文献

Female Sprague-Dawley rats were decapitated at various stages of the estrous cycle, pregnancy, lactation and following ovariectomy. Anterior pituitary and ovarian tissues were collected and assayed to quantify luteinizing hormone releasing hormone (LHRH) receptors. No changes were noted in receptor affinity either between tissues or physiological stages studied. Pituitary LHRH receptor concentrations and content were greater (P less than 0.05) during diestrus II and proestrus than during estrus. Pituitary LHRH receptor concentrations and content during pregnancy were not different from those during estrus, however, a significant decrease was noted in pituitary LHRH receptor content and concentrations during lactation compared to estrus. Ovarian LHRH receptor content did not change with stage of reproduction (P less than 0.05). There was, however, a decrease (P less than 0.05) in ovarian LHRH receptor concentrations at Week 3 of pregnancy and Week 1 of lactation which was possibly due to the increase ovarian weight noted at both these physiological stages. There was no correlation (P less than 0.1) between ovarian and pituitary LHRH receptor numbers (r = 0.096). These findings suggest that the internal mechanisms which control changes in pituitary LHRH receptor numbers do not control ovarian LHRH receptor numbers.  相似文献

The thermodynamic functions of biopolymer hydration were investigated by multitemperature vapor pressure studies. Desorption measurements were performed that allowed determination of reversible isotherms in the hydration range of 0.1 to 0.3–0.5 g H2O/g dry polymer. These isotherms are accessible to thermodynamic interpretation and are relevant to the interaction of water with biopolymers in their solution conformation. The results obtained on a series of different biopolymers (lysozyme, α-chymotrypsin, apo-lactoferrin, and desoxyribonucleic acid), show the following common features of interest: (1) The differential excess enthalpies (ΔHe ) and entropies (ΔSe ) are negative, and exhibit pronounced anomalies in a well-defined low-humidity range (approx. 0.1 g H2O/g dry polymer). These initial extrema are interpretable by structural changes, induced in the native biopolymer structures by water removal below a critical degree of hydration. (2) The ΔHe and ΔSe terms exhibit statistically significant linear enthalpy–entropy compensation effects in all biopolymer–water systems investigated. The compensation temperatures \documentclass{article}\pagestyle{empty}\begin{document}$ \hat \beta = \overline {\Delta H} ^e /\overline {\Delta S} ^e $\end{document} are approximately identical for all biopolymers, ranging from 360 to 500 K. The compensation effects are attributable to phase transitions of water molecules between the bulk liquid and the inner-sphere hydration shell of native biopolymers. (3) The negative excess free energies (ΔGe ) decrease monotonically with increasing water content and are close to zero at 0.3 to 0.5 g H2O/g polymer. This result indicates that only transitions between the bulk liquid and the inner-sphere hydration shell are associated with significant net free energy effects. The outer-sphere hydration water is thermodynamically comparable to bulk water. The importance of the proportionality factor \documentclass{article}\pagestyle{empty}\begin{document}$ \hat \beta $\end{document} in the control of the free energy term is discussed.  相似文献

We have constructed a plasmid, pLTR100, which contains human interleukin 2 receptor light (IL-2R L) chain cDNA in the inverted orientation relative to the upstream SV40 promoter. The cDNA segment is flanked by the immunoglobulin gene recombination signal sequences so that the cDNA segment can invert and the human IL-2R L chain is subsequently expressed under the control of the SV40 promoter. A murine pre-B cell line, 38B9, transfected with pLTR100 began to express the human IL-2R L chain on the cell surface. The frequency of human IL-2R L chain positive cells increased almost linearly up to 50% for 60 days of culture after transfection. Southern blot analysis and sequencing of the DNA fragments at the recombination junction confirmed that the cDNA segment was inverted in a signal sequence-dependent manner by the variable-diversity-joining recombination process. Transgenic mice bearing the recombination substrate DNA similar to pLTR100 expressed the human IL-2 L chain in the spleen, thymus, and bone marrow, but not in the other tissues examined at the detectable level. Both IgM- and CD3-positive cells expressed the human IL-2R L chain, indicating that this artificial DNA can serve as a substrate for recombination both in B- and T-cells and that another DNA segment may be necessary to confer the cell-type specificity on the substrate DNA.  相似文献

A two-step enzymatic synthesis of dTDP-L-rhamnose is developed using enzymes from sonicated extracts of cultures of Escherichia coli K12 strains harboring plasmids containing different parts of the rfb gene cluster of Salmonella enterica LT2. The intermediate dTDP-6-deoxy-D-xylo-4-hexulose was isolated after a 1-h reaction, using only dTDP-D-glucose and dTDP-D-glucose 4,6-dehydratase, followed by protein precipitation and desalting by gel chromatography (yield 89%). In a two-step reaction using dTDP-D-glucose and dTDP-D-glucose 4,6-dehydratase in the first step, and with NADPH, dTDP-6-deoxy-D-xylo-4-hexulose 3,5-epimerase and NADPH:dTDP-6-deoxy-L-lyxo-4-hexulose-4-reductase in the second hour of incubation, the dTDP-D-glucose was fully converted to dTDP-L-rhamnose. The hexoses of both products were identified by mass spectroscopy. The molar yield of dTDP-L-rhamnose, after protein precipitation, anion-exchange chromatography and desalting by gel chromatography, was 62%, corresponding to more than 150 mg, starting from 250 mg of dTDP-D-glucose. When stored lyophilysed under nitrogen, these products were found to be stable for several months. Both dTDP-6-deoxy-D-xylo-4-hexulose and dTDP-L-rhamnose have light absorption maxima at 267 nm, with molar absorption coefficients close to that of dTMP. However, the absorption coefficient of dTDP-6-deoxy-D-xylo-4-hexulose at the absorption maximum of 320 nm (specific for sugars containing keto groups) was found to be approximately 20% higher than values presented earlier. Furthermore, an HPLC technique is presented for determining the net activity of dTDP-6-deoxy-D-xylo-4-hexulose 3,5-epimerase and NADPH:dTDP-6-deoxy-L-lyxo-4-hexulose-4-reductase, based on separation of dTDP-6-deoxy-D-xylo-4-hexulose and dTDP-L-rhamnose. The HPLC technique is also suitable for determination of all the nucleotide components involved in the synthesis.  相似文献
