Adaptation to deionized water (DW) affects several cell types in the goldfish. The pars intermedia PAS-positive cells are highly stimulated. Their low response or the absence of changes in goldfish kept in 1/3 Ca-free seawater (SW) and in Ca-free SW-adapted eels, respectively, suggest that sodium and/or magnesium are interfering. To test this hypothesis, young goldfish were adapted to DW supplemented or not with sodium (50 and 140 mM) for 8 and 16 days or with magnesium (16.5 and 50 mM) for 16 and 30 days. Cytological and morphometric studies of the pituitary showed that prolactin (PRL) cell activity was reduced by sodium. Thyrotropic (TSH) cells were stimulated. The activity of melanocyte-stimulating (MSH) cells increased in DW + Na+. Stimulation of the pars intermedia PAS+ cells in DW was partly inhibited by adding sodium; the cellular and nuclear areas increased only moderately, the endoplasmic reticulum (ER) was not conspicuous and mitotic activity disappeared. In DW + Mg2+ the activity of PRL, TSH and MSH cells tended to be lower after a long-term adaptation. The response of the PAS+ cells was as high as that noted in DW; complete degranulation, enlargement of the ER and important mitotic activity. Differential responses to Na+ and Mg2+ are not due to pH differences in the solutions. External sodium is able then to reduce the response of the PAS+ cells in a Ca-free environment, while magnesium is not inhibitory. Other cell types are also affected by high levels of Na+ and Mg2+.  相似文献

Male silver eels kept in fresh water were injected with 17α-methyltestosterone (MT). They received 5, 10, 15 or 20 injections (8 μg/g every other day) or 6 or 9 injections (4 μg/g or 2.5 μg/g once a week). Compared to solvent-injected controls, the treated eels showed changes in the pituitary: prolactin, corticotropic, thyrotropic and melanotropic cells were stimulated. Gonadotropic cells had enlarged nuclei, but synthesis and storage of granules and large globules were not evident, despite the progressive testicular maturation. High doses of MT induced a liver hypertrophy, a drop in plasma osmolarity and sodium level. An increase in plasma calcium level is possibly involved in the reduced activity of PAS-positive Ca-sensitive cells of the pars intermedia. These responses, previously observed in estradiol (E2) treated eels, may suggest that MT is aromatized in the brain and/or the pituitary. However, the considerable stimulation of GTH cells which always occurs in E2-treated eels is absent in MT-treated eels. With lower doses of MT, changes in the pituitary and the testis were similar to those produced by high doses, although the Ca-sensitive cells were more active. The decrease in plasma osmolarity and sodium level was less marked, plasma calcium was slightly reduced and the liver was not modified. There is no clear evidence in favour of E2 production. These data suggest that MT may stimulate the gonad directly, in agreement with biochemical data by Dufour et al. (1983).  相似文献

The purpose of this study was to investigate gender-specific motor control strategies during eccentric exercise and delayed onset muscle soreness (DOMS) in the shoulder region. Twelve healthy males and females participated in the study. Eccentric shoulder exercises were conducted on the dominant shoulder while the other side served as control. The exerted force, range of shoulder elevation, rating of perceived exertion, pain intensity, and surface electromyography (EMG) from the trapezius muscles were recorded and analyzed. A significant decrease in exerted force during exercise was only found in males despite similar rating of perceived exertion among genders. During eccentric exercise: males showed increasing root mean square (RMS) of the EMG while a decrease occurred for females, no difference between genders in mean power frequency of the EMG were seen. During static and dynamic contractions: no differences between genders in pain intensity or RMS were observed; RMS of the exercised side were lower than that of the control side (P<0.05) at 24 h after exercise. The results indicated a more prominent muscle fatigue resistance in females compared with males and mobilization of different muscle activation strategies during eccentric exercise. A protective adaptation to DOMS, i.e. decrease in RMS values was found with no gender differences.  相似文献

The microslide gel-diffusion and macro-tube agglutination techniques to detect Brucella canis antibodies in dogs were compared. Sera from dogs experimentally infected with B. canis and a random sample of dog sera with unknown histories of exposure to this organism were examined. The results of the gel-diffusion method employing specific rough Brucella saline-extract antigens of B. canis and Brucella ovis were comparable to those obtained by the tube agglutination test. The easily prepared, stable R antigen in the freeze-dried form offers a convenient, simple, and reliable diagnostic method for the serological detection of canine brucellosis by the gel-diffusion test.  相似文献

The ability of Clostridium perfringens type A to produce an enterotoxin active in human food poisoning has been shown to be directly related to the ability of the organism to sporulate. Enterotoxin was produced only in a sporulation medium and not in a growth medium in which sporulation was repressed. Mutants with an altered ability to sporulate were isolated from an sp(+) ent(+) strain either as spontaneous mutants or after mutagenesis with acridine orange or nitrosoguanidine. All sp(0) (-) mutants were ent(-). Except for one isolate, these mutants were not disturbed in other toxic functions characteristic of the wild type and unrelated to sporulation. A total of four of seven osp(0) mutants retained the ability to produce detectable levels of enterotoxin. None of the ent(-) mutants produced gene products serologically homologous to enterotoxin. A total of three sp(-) mutants, blocked at intermediate stages of sporulation, produced enterotoxin. Of these mutants, one was blocked at stage III, one probably at late stage IV, and one probably at stage V. A total of three sp(+) revertants isolated from an sp(-) ent(-) mutant regained not only the ability to sporulate but also the ability to produce enterotoxin. The enterotoxin appears to be a sporulation-specific gene product; however, the function of the enterotoxin in sporulation is unknown.  相似文献

Résumé L'étude de l'activité spontanée de 21 rats mesurée pendant 73 jours en cage à tambour permet les conclusions suivantes: les fluctuations naturelles de l'activité des animaux sont influencées par les variations de pression atmosphérique. Les hausses de pression barométrique entrainent une augmentation de la motilité spontanée alors que les baisses barométriques tendent à produire un effet inverse; cependant les chutes très brutales de pression provoquent, elles aussi, un surcroit d'activité. Des travaux récents permettent d'interpréter cette apparente contradiction.Ces corrélations disparaissent si les animaux sont protégés de l'ionisation atmosphérique par des cages reliées à la terre.Sachant que les variations de pression atmosphérique entrainent des modifications de l'ionisation atmosphérique, on peut donc admettre que cette dernière est en fait le facteur météorologique efficace sur la motilité spontanée, d'autant plus que l'aéro-ionisation est connue comme affectant nettement l'activité du rat.Enfin, au niveau neurohumoral, la sérotonine semble être l'intermédiaire par lequel l'ionisation intervient sur le comportement.
Influence of variations of barometric pressure on the spontaneous activity in the bat
Summary Changes in the spontaneous activity of 21 rats were studied for 73 days with the help of a revolving wheel.Natural fluctuations of spontaneous activity of the animals are influenced by variations of barometric pressure. When rises of barometric pressure occur, the spontaneous activity is increased, whereas barometer on the fall tends to produce an opposite effect; however, sudden and important falls in atmospheric pressure may also induce an increased activity, an observation which is discussed in the light of recent results.These correlations between activity and barometric pressure disappear when animals are shielded from atmospheric ionization by means of grounded cages.Knowing that atmospheric ionization is depending upon atmospheric pressure variation, one may admit that ionization is the main meteorological factor really effective in the action reported on activity. In other words, changes observed in spontaneous activity of the rat are mediated by variations of air ionization, themselves influenced by changes of atmospheric pressure. These data are in agreement with previous observations, by other authors demonstrating that aeroionization is acting on rat activity.Finally, the physiological link between these meteorological factors and animal behaviour appears to be a modulation of serotonin metabolism; arguments in favour of this mediation are discussed.

Heterogeneity in the intrinsic quality and nutritional condition of individuals affects reproductive success and consequently fitness. Black brant (Branta bernicla nigricans) are long‐lived, migratory, specialist herbivores. Long migratory pathways and short summer breeding seasons constrain the time and energy available for reproduction, thus magnifying life‐history trade‐offs. These constraints, combined with long lifespans and trade‐offs between current and future reproductive value, provide a model system to examine the role of individual heterogeneity in driving life‐history strategies and individual heterogeneity in fitness. We used hierarchical Bayesian models to examine reproductive trade‐offs, modeling the relationships between within‐year measures of reproductive energy allocation and among‐year demographic rates of individual females breeding on the Yukon‐Kuskokwim Delta, Alaska, using capture–recapture and reproductive data from 1988 to 2014. We generally found that annual survival tended to be buffered against variation in reproductive investment, while breeding probability varied considerably over the range of clutch size‐laying date combinations. We provide evidence for relationships between breeding probability and clutch size, breeding probability and nest initiation date, and an interaction between clutch size and initiation date. Average lifetime clutch size also had a weak positive relationship with apparent survival probability. Our results support the use of demographic buffering strategies for black brant. These results also indirectly suggest associations among environmental conditions during growth, fitness, and energy allocation, highlighting the effects of early growth conditions on individual heterogeneity, and subsequently, lifetime reproductive investment.  相似文献

An enzyme-linked immunosorbent assay (ELISA) for the Alternaria mycotoxin tenuazonic acid (TeA) was evaluated by comparative analysis of naturally contaminated sorghum grains and sorghum-based infant food, using a stable isotope dilution LC-MS assay (SIDA; limit of detection (LOD) 1.0 μg/kg) as the reference method. LODs of the ELISA were 30 μg/kg in sorghum grains and 220 μg/kg in sorghum-based infant cereals. With SIDA, 100% of the samples (n = 28) had been positive for TeA in a concentration range of 6–584 μg/kg (mean 113 μg/kg). The ELISA consistently detected TeA in all naturally contaminated samples at cut-off levels of 30–60 μg/kg (sorghum) and 200–300 μg/kg (infant cereals), as based on corresponding to SIDA values. Although the ELISA was much less sensitive than the SIDA method, it may be useful as a screening method for sorghum and sorghum-based infant foods and can be employed to identify samples containing elevated concentrations of TeA in food, well below the proposed level of concern (500 μg/kg).  相似文献
