An examination of the external morphology of a recent collection from Araracuara, Colombia, has suggested that the plant belongs to the primarily Old World family Dipterocarpaceae. A study of the wood, bark, and pith anatomy of this new taxon,Pseudomonotes tropenbosii Londoño, Alvarez & Forero, was undertaken to help confirm its systematic affinities. Comparisons ofPseudomonotes with data from the literature and reference wood slides, coupled with the use of computer-aided identification keys, support the view that its closest relationships probably are within the family Dipterocarpaceae. Detailed anatomical comparisons have revealed thatPseudomonotes' relationships are most likely to be with the subfamily Monotoideae, comprised of the African generaMonotes A. Abstract: The subtilisin-like prohormone convertase SPC3 is likely to play a role in the biosynthesis of a variety of biologically active peptides. SPC3 undergoes a series of posttranslational processing events during its biosynthesis. Multiple forms have been identified that show varying degrees of truncation at the carboxyl terminus. In this study we show that the 86-kDa form of recombinant SPC3 with an intact carboxyl terminus can undergo rapid carboxyl-terminus truncation to produce a 64-kDa form. We have defined the optimal conditions for carboxyl-terminus truncation in vitro. The carboxyl-terminus truncation reaction was less calcium sensitive, active over a broader pH range, and showed differences in inhibitor sensitivity compared with the enzymatic activities of full-length and truncated forms of SPC3 toward a fluorescent peptide substrate. Increases in enzymatic activity of 86-kDa SPC3 were also measured over a time frame consistent with conversion to the 64-kDa form. However, similar specific activities for both forms of the enzyme suggest such activity increases may not be due to carboxyl-terminus truncation. The different enzymatic properties of the major molecular forms of SPC3 highlight the importance of understanding the molecular events regulating carboxyl-terminal processing of this endoprotease. 相似文献

Phylogenetic relationships in the Desmarestiales (Phaeophyceae) were inferred among the monotypic Arthrocladia (Arthrocladiaceae) and 27 isolates from Desmarestiaceae, representing 17 taxa of Desmarestia and the monotypic Antarctic genera Himantothallus and Phaeurus. Phaeurus and Arthrocladia were used as outgroups. Parsimony analyses of nuclear ribosomal DNA internal transcribed spacer (ITS1 and ITS2) sequences, in which gaps were both included and excluded, yielded well-resolved trees with a consistent general branching pattern. A parallel analysis of nine morphological and life-history characters and three ecological characters yielded a similar tree but provided little resolution in the terminal clades. The position of the monotypic Arthrocladia villosa within the Desmarestiales is consistent with monophyly for the order, but its position as the most primitive desmarestialean is not resolvable from the molecular data set. The basal position of Phaeurus, the Antarctic Desmarestia species, and Himantothallus is consistent with the hypothesis of a Southern Hemisphere origin for the family Desmarestiaceae. The more recent Northern Hemisphere “aculeata” clade evolved from an Antarctic ancestor. A “D. aculeata-like” species was ancestral to a lineage characterized by annual sporophytes with high sulfuric acid content, which radiated into many species, widely distributed in both hemispheres. Mapping of morphological and ecological characters onto the molecular tree confirm the informativeness of sulfuric acid-containing vacuoles and unilocular sporangial types. There is good congruence between phylogenetic tree topology and temperature impints in relation to biogeographic distribution, supporting the theory that temperature tolerance is a conservative trait. 相似文献

Arbuscule-forming fungi in the order Glomales form obligate endomycorrhizal associations with plants that make them difficult to quantify, and taxonomy of the group is only beginning to be objectively understood. Fatty acid methyl ester (FAME) profiles were analyzed to assess the diversity and quantity of fatty acids in 53 isolates of 24 glomalean species. Spores and endomycorrhizal roots of sudan grass (Sorghum sudanense) and the citrus rootstock Carrizo citrange (Poncirus trifoliata x Citrus sinensis) were examined. Spores yielded reproducible FAME profiles from replicate spore collections extracted from soil pot cultures despite being grown in association with a host plant and with contaminating microorganisms present. Unweighted pair group analysis revealed relatively tight clusters of groups at the intraspecific, specific, and generic levels; however, lipid profiles at the family level were convergent. Thus, FAME profile comparisons provided a robust measure of similarity below the family level. FAME profiles in sudan grass roots containing vesicles and/or spores of Glomus intraradices were more similar to spore profiles than to profiles from nonmycorrhizal roots. The FAME profiles for Gigaspora species, which do not form vesicles or spores in roots, were less distinct from nonmycorrhizal roots. G. intraradices and G. rosea produced fatty acids in roots that were distinguishable from each other as well as from the host root. Production in citrus roots of the fatty acid 16:1(inf(omega)5) cis by two Glomus species was correlated with the development of mycorrhizal colonization as measured by clearing and staining procedures and by estimates of total incidence and vesicle intensity. FAME analysis of roots not only provided a measure of colonization development but also served as an index of carbon allocated to intraradical fungal growth and lipid storage. 相似文献

Pteria brevialata characteristically attaches to Hicksonella princeps (Cnidaria: Gorgonoidea) at depths > 10 m in Hong Kong and the South China Sea. Attachment is by a stout byssus to the basal regions of the gorgonian colonies. The shell appears typical of the Pteriidae, but closer inspection shows that the antero-ventral margin, in particular, moulds itself to the form of the cylindrical gorgonian, which also changes its growth form, so that securer attachment is achieved. The margin of the right valve is not nacred so that the flexible outer prismatic layer adpresses firmly against the more extensively nacred left valve when they close; mantle and ctenidia also being withdrawn deeply. This is probably an anti-predation device. There are no mantle fusions, even between posterior inhalant and exhalant streams. The mantle margin is, however, complexly tentaculate. Pteria brevialata is monomyarian with reduced anterior byssal retractor muscles. The anterior face of the shell is thus greatly reduced whereas the posterior is inflated and postero-dorsally 'winged'.
Other features of the anatomy are described, including the ctenidial eyespot and simple photoreceptors in the inner component of the duplicated outer mantle folds, under the periostracum. Similar structures in other pterioideans and arcoideans suggest a close relationship between their respective orders.
It is concluded that gorgonian-associated pterioids can be derived from an epibyssate, crevice-dwelling ancestor, exploiting, as solitary individuals, the currents and clean oceanic waters above the substratum. This has three important advantages: (a) removal from surface-roving predators (the gorgonian providing additional protection); (b) exploitation of a niche hitherto unoccupied by bivalves and (c) removal of the animal from the sea-bed and exclusive exploitation of potential food held in suspension. Rat basophil leukemia cell homogenates effectively catalyze the conversion of leukotriene A4 to a mixture of leukotrienes C4 and D4 in the presence of glutathione. These homogenates also catalyze the formation of adducts of halogenated nitrobenzene with glutathione, as determined spectrophotometrically. While all the classical glutathione S-transferase activity resides in the soluble fraction of the homogenates, the thiol ether leukotriene-generating activity is found in the particulate fraction. This “leukotriene C synthetase” activity has been solubilized from a crude high-speed particulate fraction by means of the nonionic detergent, Triton X-100. The solubilized enzyme is incapable of converting 2,4-dinitrochlorobenzene to a colored product in the presence of glutathione. Nor will it react with 3,4-dichloronitrobenzene. On the other hand, under optimal conditions, this enzyme preparation is capable of generating about 0.1 nmol leukotriene C mg protein?1 min?1 in a reaction which continues in linear fashion for at least 10 min. This dissociation in substrate specificity, as well as differences in the inhibition profile, distinguish the enzyme activity in the particulate fraction from rat basophil leukemia cell homogenates from the microsomal glutathione S-transferase which has been described in rat liver homogenates, suggesting that this “leukotriene C synthetase” is a new and unique enzyme. 相似文献

Summary A low-molecular-weight (1,400) factor isolated from a human plasma -globulin concentrate by acid-salt dissociation and ultrafiltration inhibits proliferation of mitogen-stimulated T cells and L1210 leukemia cells. The factor (UM05R) inhibits DNA, RNA, and protein synthesis in sensitive cells, acts in G1 of the cell cycle, and appears to suppress mitogen-responsive T cells without an accessory cell requirement. UM05R activity is enhanced by known cAMP-elevating agents and by sulfhydryl compounds. The results of the present study are consistent with the hypothesis that the plasma-derived agent inhibits lympho-proliferation as a result of elevation of intracellular cAMP. 相似文献
