A historical cohort study of German nuclear power workers was set up to investigate the overall and cancer mortality risk
related to a long-term low-level exposure to ionising radiation. The cohort was part of an international collaborative study
whose pooled analyses were carried out at the International Agency for Research on Cancer (IARC), Lyon, and published recently.
Due to delays in data collection, data from the German cohort were not included in these analyses. This cohort includes 4,844
employees who worked in any of 10 nuclear power plants, between 1 January 1991 and 31 December 1997. Sixty-eight deaths among
men were observed in 31,000 person years, and none among women. Standardized mortality ratios (SMRs) were computed for all
causes of death, all cancers, cardiovascular diseases, external causes, and all other causes. Overall, a strong healthy worker
effect was observed (all-cause SMR = 0.54, 95% CI 0.42–0.67), and no increase in total cancer mortality was seen (SMR = 0.66,
95% CI 0.43–0.95). These results are in line with and complement IARC results. Figures are yet too small for stable risk estimates
and further work is therefore under way to include more power plants, and to extend the follow-up until the year 2005. 相似文献

BackgroundAsthma is characterized by type 2 T-helper cell (Th2) inflammation, goblet cell hyperplasia, airway hyperreactivity, and airway fibrosis. Monocyte chemoattractant protein-1 (MCP-1 or CCL2) and its receptor, CCR2, have been shown to play important roles in the development of Th2 inflammation. CCR2-deficient mice have been found to have altered inflammatory and physiologic responses in some models of experimental allergic asthma, but the role of CCR2 in contributing to inflammation and airway hyperreactivity appears to vary considerably between models. Furthermore, MCP-1-deficient mice have not previously been studied in models of experimental allergic asthma.MethodsTo test whether MCP-1 and CCR2 are each required for the development of experimental allergic asthma, we applied an Aspergillus antigen-induced model of Th2 cytokine-driven allergic asthma associated with airway fibrosis to mice deficient in either MCP-1 or CCR2. Previous studies with live Aspergillus conidia instilled into the lung revealed that MCP-1 and CCR2 play a role in anti-fungal responses; in contrast, we used a non-viable Aspergillus antigen preparation known to induce a robust eosinophilic inflammatory response.ResultsWe found that wild-type C57BL/6 mice developed eosinophilic airway inflammation, goblet cell hyperplasia, airway hyperreactivity, elevations in serum IgE, and airway fibrosis in response to airway challenge with Aspergillus antigen. Surprisingly, mice deficient in either MCP-1 or CCR2 had responses to Aspergillus antigen similar to those seen in wild-type mice, including production of Th2 cytokines.ConclusionWe conclude that robust Th2-mediated lung pathology can occur even in the complete absence of MCP-1 or CCR2. 相似文献

We investigated whether concentrations of carboxylates in the rhizosphere of chickpea (Cicer arietinum L.) roots were related to soil phosphorus levels. In a field experiment, cultivar Sona was grown at two P levels on eight soil types at three locations. There were large differences in extractable (0.2 mM CaCl2) rhizosphere carboxylate concentrations amongst the locations. The effect of P fertiliser was variable and carboxylate concentrations depended on soil type. To examine the effect of soil P in more detail, a glasshouse experiment was carried out, in which three cultivars (Heera, Sona and Tyson) were grown at four P levels on one soil type. The biomass of chickpea plants increased with increasing P level of the soil, and the root mass ratio decreased at the highest soil P level. However, rhizosphere concentrations of the carboxylates malonate, malate and citrate did not differ significantly between P treatments. This implied that there was no simple relation between available P and root exudation rates, in contrast to earlier results in studies using hydroponics. Cultivars differed in carboxylate concentration pattern: Sona and Tyson showed a tendency towards increased rhizosphere carboxylate concentrations at the second harvest, whereas the carboxylate concentration of Heera tended to decrease. It is hypothesised that chickpea roots always exude a basal level of carboxylates into the rhizosphere. They only increase carboxylate exudation considerably when the P availability is extremely low, which may occur in soils that strongly bind P. 相似文献

14C‐labelled straw was mixed with soils collected from seven coniferous forests located on a climatic gradient in Western Europe ranging from boreal to Mediterranean conditions. The soils were incubated in the laboratory at 4°, 10°, 16°, 23° and 30 °C with constant moisture over 550 days. The temperature coefficient (Q10) for straw carbon mineralization decreased with increasing incubation temperatures. This was a characteristic of all the soils with a difference of two Q10 units between the 4–10° and the 23? 30 °C temperature ranges. It was also found that the magnitude of the temperature response function was related to the period of soil incubation. Initial temperature responses of microbial communities were different to those shown after a long period of laboratory incubation and may have reflected shifts in microbial species composition in response to changes in the temperature regime. The rapid exhaustion of the labile fractions of the decomposing material at higher temperatures could also lead to underestimation of the temperature sensitivity of soils unless estimated for carbon pools of similar qualities. Finally, the thermal optima for the organic soil horizons (Of and Oh) were lower than 30 °C even after 550 days of incubation. It was concluded that these responses could not be attributed to microbial physiological adaptations, but rather to the rates at which recalcitrant microbial secondary products were formed at higher temperatures. The implication of these variable temperature responses of soil materials is discussed in relation to modelling potential effects of global warming. 相似文献

Obesity has been implicated as a significant risk factor for development of pancreatic cancer. In the setting of obesity, a systemic chronic inflammatory response is characterized by alterations in the production and secretion of a wide variety of growth factors. Leptin is a hormone whose level increases drastically in the serum of obese patients. High fat diet induced obesity in mice leads to an overall increased body weight, pancreatic weight, serum leptin, and pancreatic tissue leptin levels. Here we report the contribution of obesity and leptin to pancreatic cancer growth utilizing an in vivo orthotopic murine pancreatic cancer model, which resulted in increased tumor proliferation with concomitant increased tumor burden in the diet induced obese mice compared to lean mice. Human and murine pancreatic cancer cell lines were found to express the short as well as the long form of the leptin receptor and functionally responded to leptin induced activation through an increased phosphorylation of AKT473. In vitro, leptin stimulation increased cellular migration which was blocked by addition of a PI3K inhibitor. In vivo, depletion of the leptin receptor through shRNA knockdown partially abrogated increased orthotopic tumor growth in obese mice. These findings suggest that leptin contributes to pancreatic tumor growth through activation of the PI3K/AKT pathway, which promotes pancreatic tumor cell migration. 相似文献

The Hoechst dye staining method has been successfully applied to the central nervous system in mammals and its use has been demonstrated in intracerebral transplantation. The technique is rapid, simple and based on intrinsic nuclear properties. It was found to be permanent and valid whatever the animal strains or ages, allowing the distinction of rat cells from those of mouse, studied either separately or in a cross-transplantation model. It permitted the detection of grafted cells in the area of transplantation and the observation of early dispersion around the implantation site. Moreover, it can be combined with immunohistochemistry as demonstrated by a myelin marker in a relevant model. Immunodetection can thus help to directly observe grafted cells, at distance from the locus of transplantation, confirming their presence in the graft-type myelin patches.
Because of its rapid performance, this technique can be used systematically after transplantation to check for the presence of grafted cells in the host. 相似文献
