This study utilized recently developed microbead technology to remove natural killer (NK) cells from peripheral blood mononuclear cell (PBMC) preparations to determine the effect of acute exercise on T-lymphocyte function, independent of changes in lymphocyte subpopulations. Twelve well-trained male runners completed a 60-min exercise trial at 95% ventilatory threshold and a no-exercise control trial. Six blood samples were taken at each session: before exercise, midexercise, immediately after exercise, and 30, 60, and 90 min after exercise. Isolated PBMC and NK cell-depleted PBMC were stimulated with the mitogen phytohemagglutinin. Cellular proliferation was assessed by using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide dye uptake. In the PBMC cultures, there was a significantly lower mitogen response to phytohemagglutinin in exercise compared with the control condition immediately postexercise. There were no significant differences between the control and exercise conditions in NK cell-depleted PBMC cultures or in the responses adjusted for the percentage of CD3 cells. The role of food supply in the dispersal behaviour of juvenile Tawny Owls Strix aluco 总被引:1,自引:0,他引:1
We investigated the effects of food supply on decisions made by dispersing juvenile Tawny Owls Strix aluco in Kielder Forest, Northumberland, in 1996 and 1997. Field Voles Microtus agrestis were the main food of the owls and clear-cuts the main habitat for voles. A vole sign index was used to estimate vole abundance. In areas near to roosting owls, mean vole densities were 83 and 115 ha−1 in 1996 and 1997, respectively. The prediction that birds would perform area-restricted searches when prey was more abundant was not confirmed. Moreover, we found no evidence that juveniles avoided conspecifics. Owls appeared to have an imperfect knowledge of the environment as they responded to variability in Field Vole densities by altering the time spent in different areas rather than by moving to areas with successively greater vole densities. Vole abundance explained 25.7% of the variation in the time spent in different areas. Movements did not decrease with time after dispersal, although the detection of such movements was prone to error. This study supports recent work suggesting that although dispersal may be initiated by a variety of proximate and ultimate factors, individual decisions made during dispersal may depend partly on environmental conditions encountered during the process itself. 相似文献

The microsomal share of liver homogenate 75Se after injection of a tracer dose of 75SeO32? was three times greater in rats fed a selenium-deficient diet than in rats fed a selenium-adequate diet. Basal levels of microsomal cytochromes P-450 and b5 were unaffected by selenium deficiency. However, induction of these cytochromes by phenobarbital was markedly inpaired in selenium-deficient rats, whereas liver weight increase and NADPH cytochrome reductase induction were not impaired. Artemisinin-based combination therapy is exerting novel selective pressure upon populations of Plasmodium falciparum across Africa. Levels of resistance to non-artemisinin partner drugs differ among parasite populations, and so the artemisinins are not uniformly protected from developing resistance, already present in South East Asia. Here, we consider strategies for prolonging the period of high level efficacy of combination therapy for two particular endemicities common in Africa. Under high intensity transmission, two alternating first-line combinations, ideally with antagonistic selective effects on the parasite genome, are advocated for paediatric malaria cases. This leaves second-line and other therapies for adult cases, and for intermittent preventive therapy. The drug portfolio would be selected to protect the 'premier' combination regimen from selection for resistance, while maximising impact on severe disease and mortality in children. In endemic areas subject to low, seasonal transmission of Plasmodium falciparum, such a strategy may deliver little benefit, as children represent a minority of cases. Nevertheless, the deployment of other drug-based interventions in low transmission and highly seasonal areas, such as mass drug administration aimed to interrupt malaria transmission, or intermittent preventive therapy, does provide an opportunity to diversify drug pressure. We thus propose an integrated approach to drug deployment, which minimises direct selective pressure on parasite populations from any one drug component. This approach is suitable for qualitatively and quantitatively different burdens of malaria, and should be supported by a programme of routine surveillance for emerging resistance. 相似文献

Differential rates of nucleotide substitution among different gene segments
and between distinct evolutionary lineages is well documented among
mitochondrial genes and is likely a consequence of locus-specific selective
constraints that delimit mutational divergence over evolutionary time. We
compared sequence variation of 18 homologous loci (15 coding genes and 3
parts of the control region) among 10 mammalian mitochondrial DNA genomes
which allowed us to describe different mitochondrial evolutionary patterns
and to produce an estimation of the relative order of gene divergence. The
relative rates of divergence of mitochondrial DNA genes in the family
Felidae were estimated by comparing their divergence from homologous
counterpart genes included in nuclear mitochondrial DNA (Numt, pronounced
"new might"), a genomic fossil that represents an ancient transfer of 7.9
kb of mitochondrial DNA to the nuclear genome of an ancestral species of
the domestic cat (Felis catus). Phylogenetic analyses of mitochondrial
(mtDNA) sequences with multiple outgroup species were conducted to date the
ancestral node common to the Numt and the cytoplasmic (Cymt) mtDNA genes
and to calibrate the rate of sequence divergence of mitochondrial genes
relative to nuclear homologous counterparts. By setting the fastest
substitution rate as strictly mutational, an empirical "selective
retardation index" is computed to quantify the sum of all constraints,
selective and otherwise, that limit sequence divergence of mitochondrial
gene sequences over time.
