The rare alleles model of mutation-selection balance (MSB) hypothesis for the maintenance of genetic variation was evaluated for two quantitative traits, ovariole number and body size. Mutational variances (VM) for these traits, estimated from mutation accumulation lines, were 4.75 and 1.97 x 10(-4) times the environmental variance (VE), respectively. The mutation accumulation lines were studied in three environments to test for genotype x environment interaction (GEI) of new mutations; significant mutational GEI was found for both traits. Mutations for ovariole number have a quadratic relationship with competitive fitness, suggesting stabilizing selection for the trait; there is no significant correlation between mutations for body size and competitive fitness. Under MSB, the ratio of segregating genetic variance, VG, to mutational variance, VM, estimates the inverse of the selection coefficient against a heterozygote for a new mutation. Estimates of VG/VM for ovariole number and body size were both approximately 1.1 x 10(4). Thus, MSB can explain the level of variation, if mutations affecting these traits are under very weak selection, which is inconsistent with the empirical observation of stabilizing selection, or if the estimate of VM is biased downward by two orders of magnitude. GEI is a possible alternative explanation.  相似文献

Multivariate predictive models are widely used tools for assessment of aquatic ecosystem health and models have been successfully developed for the prediction and assessment of aquatic macroinvertebrates, diatoms, local stream habitat features and fish. We evaluated the ability of a modelling method based on the River InVertebrate Prediction and Classification System (RIVPACS) to accurately predict freshwater fish assemblage composition and assess aquatic ecosystem health in rivers and streams of south-eastern Queensland, Australia. The predictive model was developed, validated and tested in a region of comparatively high environmental variability due to the unpredictable nature of rainfall and river discharge. The model was concluded to provide sufficiently accurate and precise predictions of species composition and was sensitive enough to distinguish test sites impacted by several common types of human disturbance (particularly impacts associated with catchment land use and associated local riparian, in-stream habitat and water quality degradation). The total number of fish species available for prediction was low in comparison to similar applications of multivariate predictive models based on other indicator groups, yet the accuracy and precision of our model was comparable to outcomes from such studies. In addition, our model developed for sites sampled on one occasion and in one season only (winter), was able to accurately predict fish assemblage composition at sites sampled during other seasons and years, provided that they were not subject to unusually extreme environmental conditions (e.g. extended periods of low flow that restricted fish movement or resulted in habitat desiccation and local fish extinctions).  相似文献

Some streams near Dorset in south-central Ontario suffer from acid precipitation via run-off and seepage from thin soils with little buffering capacity. A spring-summer survey of eight headwater streams revealed some characteristics of their insect communities which could be correlated with pH. The streams could be divided into three groups according to pH and community structure. In the most acid group (annual pH range 4.3–4.8), Ephemeroptera were absent from two streams although mature Leptophlebia were collected just after spring thaw from the most acid one (pH 4.3–4.5). One of these three streams also lacked Plecoptera but the others had two or three genera, all shredders. The second group of three streams (pH 5.0–6.3), with one exception, did support Ephemeroptera (3–4 genera) and Plecoptera (1–4 genera), most of the latter being shredders. In all six of these acid streams, Trichoptera were more diverse and more dense than Ephemeroptera and Plecoptera; again, shredders were clearly dominant, especially the limnephilid caddisfly, Frenesia difficilis (Walker). These six streams also had similar chironomid communities (densities were an order of magnitude higher than other insects). Dominance by Chironomini and abundant Tanypodinae typified the most acid streams. In contrast, the two streams in the third group (pH 5.3–6.7) had richer and more balanced communities in general with relatively fewer shredders (no Frenesia), more collectors, and fewer Chironomini and Tanypodinae. As a field experiment showed that autumn-shed leaves decomposed more slowly in acid than in non-acid streams, summer-growing shredders may benefit from the pulse of acidity at snowmelt.  相似文献

The relationship between anionic-lipid concentration and the functional properties of plasma-membrane domains was explored using the guinea-pig sperm membrane as a model, with polymyxin B (PXB) as a probe. Areas of plasmalemma specialized for fusion during the acrosome reaction had a higher affinity for the probe than adjacent nonfusigenic regions. In addition, capacitation--a process preceding acrosome:plasma-membrane fusion--markedly enlarged the area susceptible to PXB binding over the acrosomal cap. Protease treatment mimicked capacitation by increasing the acrosome-reaction incidence as well as PXB binding, at enzyme concentrations not affecting the surface coat nor altering filipin/sterol localization. Both proteolytic digestion and capacitation failed to augment PXB- or filipin-affinity in nonfusigenic zones, such as the post-acrosomal segment, including its particle-free maculae. Incubation of sperm in capacitating medium supplemented with 32P-labeled phosphate, followed by lipid extraction, thin-layer chromatography, and autoradiography, revealed a radioactive band comigrating with cardiolipin and phosphatidic acid. Vermiform protrusions elicited by PXB in the outer lamellae of cardiolipin- phosphatidylcholine liposomes resembled those seen in fusional regions of sperm membrane. We conclude that (a) differing concentrations of anionic lipids are found in adjacent domains of the sperm plasma membrane; (b) these domains mirror the functional regions of the membrane, with higher anionic-lipid concentrations localized over fusional zones; (c) the surface coat does not participate in the maintenance of such domains; (d) anionic-lipid synthesis may contribute to their formation; and (e) anionic-lipid concentrations increase as the membrane becomes fusionally competent, indicating that cellular modulation of lipid domains accompanies regulation of membrane function.  相似文献

Individual variation in alcohol consumption in human populations is determined by genetic, environmental, social and cultural factors. In contrast to humans, genetic contributions to complex behavioral phenotypes can be readily dissected in Drosophila, where both the genetic background and environment can be controlled and behaviors quantified through simple high‐throughput assays. Here, we measured voluntary consumption of ethanol in ~3000 individuals of each sex from an advanced intercross population derived from 37 lines of the Drosophila melanogaster Genetic Reference Panel. Extreme quantitative trait loci mapping identified 385 differentially segregating allelic variants located in or near 291 genes at P < 10?8. The effects of single nucleotide polymorphisms associated with voluntary ethanol consumption are sex‐specific, as found for other alcohol‐related phenotypes. To assess causality, we used RNA interference knockdown or P{MiET1} mutants and their corresponding controls and functionally validated 86% of candidate genes in at least one sex. We constructed a genetic network comprised of 23 genes along with a separate trio and a pair of connected genes. Gene ontology analyses showed enrichment of developmental genes, including development of the nervous system. Furthermore, a network of human orthologs showed enrichment for signal transduction processes, protein metabolism and developmental processes, including nervous system development. Our results show that the genetic architecture that underlies variation in voluntary ethanol consumption is sexually dimorphic and partially overlaps with genetic factors that control variation in feeding behavior and alcohol sensitivity. This integrative genetic architecture is rooted in evolutionarily conserved features that can be extrapolated to human genetic interaction networks.  相似文献

The effect of free Cd(II) ions on monomeric Cd7-metallothionein-2 (MT) from rabbit liver has been studied. Slow, concentration-dependent dimerization of this protein was observed by gel filtration chromatographic studies. The dimeric MT form, isolated by gel filtration, contains approximately two additional and more weakly bound Cd(II) ions per monomer. The incubation of MT dimers with complexing agents EDTA and 2-mercaptoethanol leads to the dissociation of dimers to monomers. The results of circular dichroism (CD) and electronic absorption studies indicate that the slow dimerization process is preceded by an initial rapid Cd-induced rearrangement of the monomeric Cd7-MT structure. The 113Cd NMR spectrum of the MT dimer revealed only four 113Cd resonances at chemical shift positions similar to those observed for the Cd4 cluster of the well-characterized monomeric 113Cd7-MT. This result suggests that on dimer formation major structural changes occur in the original three-metal cluster domain of Cd7-MT.  相似文献

The fortuitous occurrence of cases of jaagsiekte (sheep pulmonary adenomatosis) in a group of young Scottish Blackface sheep exposed to experimental cold stress is described. The incidence (11/62 cases) was high and the age of onset early (less than 20 months) by comparison with the authors' local experience of the disease. There was also some indication that males were more susceptible (9/26) than females (2/36). The results are not conclusive, but suggest the need for similar cold stress experiments in animals deliberately infected with the disease.
Zusammenfassung Es wird über das zufällige Auftreten von Jaagsiekte (Lungenadenomatose der Schafe) in einer Gruppe von Schottisch-Blackface Schafen berichtet, die in einem Kältebelastungsversuch verwendet wurden. Die Häufigkeit war hoch (11/62 Fälle) und nach den Erfahrungen der Autoren früh, denn die Tiere waren unter 20 Monate alt. Ausserdem bestanden Hinweise auf Geschlechtsunterschiede in der Empfindlichkeit, weil mehr männliche (9/26) als weibliche Tiere (2/36) erkrankten. Die Ergebnisse sind nicht endgültig. Zur weiteren Abklärung sind Versuche erforderlich, in denen vorsätzlich infizierte Tiere der gleichen Kältebelastung ausgesetzt werden.

Ces résultats ne sont cependant pas définitifs. Afin de les compléter, il faudrait placer des animaux préalablement infectés dans des conditions semblables de contrainte au froid.
