Enhancing the nutritional quality and disease resistance of crops without sacrificing productivity is a key issue for developing varieties that are valuable to farmers and for simultaneously improving food security and sustainability. Expression of the Arabidopsis thaliana species‐specific AtQQS (Qua‐Quine Starch) orphan gene or its interactor, NF‐YC4 (Nuclear Factor Y, subunit C4), has been shown to increase levels of leaf/seed protein without affecting the growth and yield of agronomic species. Here, we demonstrate that overexpression of AtQQS and NF‐YC4 in Arabidopsis and soybean enhances resistance/reduces susceptibility to viruses, bacteria, fungi, aphids and soybean cyst nematodes. A series of Arabidopsis mutants in starch metabolism were used to explore the relationships between QQS expression, carbon and nitrogen partitioning, and defense. The enhanced basal defenses mediated by QQS were independent of changes in protein/carbohydrate composition of the plants. We demonstrate that either AtQQS or NF‐YC4 overexpression in Arabidopsis and in soybean reduces susceptibility of these plants to pathogens/pests. Transgenic soybean lines overexpressing NF‐YC4 produce seeds with increased protein while maintaining healthy growth. Pull‐down studies reveal that QQS interacts with human NF‐YC, as well as with Arabidopsis NF‐YC4, and indicate two QQS binding sites near the NF‐YC‐histone‐binding domain. A new model for QQS interaction with NF‐YC is speculated. Our findings illustrate the potential of QQS and NF‐YC4 to increase protein and improve defensive traits in crops, overcoming the normal growth‐defense trade‐offs.  相似文献

Ribosomes are essential components of all cells. A large body of knowledge has been accumulated regarding ribosome synthesis and assembly; however, the pathways of normal ribosome turnover, especially rRNA decay, are not known. Some information on ribosome recycling derives from studies on starved yeast cells that use a specialized type of autophagy, called ribophagy, to differentially target ribosomes for degradation. We found that Arabidopsis RNS2, a conserved ribonuclease of the RNase T2 family, is necessary for normal decay of rRNA. Mutants lacking RNS2 activity have longer-lived rRNA, accumulate RNA in the vacuole and show constitutive macroautophagy. Thus, it is clear that normal rRNA decay is necessary to maintain cellular homeostasis. These phenotypes and the subcellular localization of RNS2 in the endoplasmic reticulum and the vacuole suggest that RNS2 participates in a ribophagy-like mechanism that targets ribosomes for recycling under normal growth conditions.  相似文献

Diatom survivorship in ballast water during trans-Pacific crossings   总被引:1,自引:0,他引:1  
Ship ballast water is believed to be responsible for global dispersal of alien biota; mid-ocean ballast water exchange is most commonly used to mitigate this process. Diatoms are among the most abundant biotic-component in ballast water, yet their invasive biology is poorly understood. To test effectiveness of MOE we examined diatom species composition and cell density in two sets of samples. First, we examined samples collected daily during one 24 days long trans-Pacific crossing in tanks with and without ballast exchanged. Second, we used samples from 23 trans oceanic vessels arriving at Vancouver harbour where diatoms were collected on arrival. Up to 86,429 live diatom cells/l were found in the tanks, ~50% of the samples share up to eight species consistently present. Cell densities and species richness declined over time and with replacement of coastal ballast water by mid-oceanic water. In both data sets diatoms survive in the tanks for as long as 33 days despite ballast exchange.  相似文献

Here, we examined whether diving parameters and complexity in the temporal organization of diving behavior of little penguins (Eudyptula minor) are influenced by sea surface temperature (SST), water stratification, and wind speed—three oceanographic features influencing prey abundance and distribution in the water column.
  3. Using fractal time series analysis, we found that foraging complexity, expressed as the degree of long‐range correlations or memory in the dive series, was associated with SST and water stratification throughout the breeding season, but not with wind speed. Little penguins foraging in warmer/more‐stratified waters exhibited greater determinism (memory) in foraging sequences, likely as a response to prey aggregations near the thermocline. They also showed higher foraging efficiency, performed more dives and dove to shallower depths than those foraging in colder/less‐stratified waters.
  4. Reductions in the long‐term memory of dive sequences, or in other words increases in behavioral stochasticity, may suggest different strategies concerning the exploration–exploitation trade‐off under contrasting environmental conditions.

Ribosomes account for a majority of the cell''s RNA and much of its protein and represent a significant investment of cellular resources. The turnover and degradation of ribosomes has been proposed to play a role in homeostasis and during stress conditions. Mechanisms for the turnover of rRNA and ribosomal proteins have not been fully elucidated. We show here that the RNS2 ribonuclease and autophagy participate in RNA turnover in Arabidopsis thaliana under normal growth conditions. An increase in autophagosome formation was seen in an rns2–2 mutant, and this increase was dependent on the core autophagy genes ATG9 and ATG5. Autophagosomes and autophagic bodies in rns2–2 mutants contain RNA and ribosomes, suggesting that autophagy is activated as an attempt to compensate for loss of rRNA degradation. Total RNA accumulates in rns2–2, atg9–4, atg5–1, rns2–2 atg9–4, and rns2–2 atg5–1 mutants, suggesting a parallel role for autophagy and RNS2 in RNA turnover. rRNA accumulates in the vacuole in rns2–2 mutants. Vacuolar accumulation of rRNA was blocked by disrupting autophagy via an rns2–2 atg5–1 double mutant but not by an rns2–2 atg9–4 double mutant, indicating that ATG5 and ATG9 function differently in this process. Our results suggest that autophagy and RNS2 are both involved in homeostatic degradation of rRNA in the vacuole.  相似文献

Soybean aphid, Aphis glycines Matsumura (Hemiptera: Aphididae), is a serious pest of soybean, Glycine max (L.) Merr., in the North Central United States. Current management recommendations rely on the application of insecticides based on an economic threshold (ET) of 250 aphids per plant. Natural enemies are important in slowing the increase of aphid populations and can prevent them from reaching levels that can cause economic losses. However, biological control of A. glycines is inconsistent and can be affected negatively by the intensity of agricultural activity. We measured the impact of a natural-enemy-free environment on the capacity of the current ET to limit yield loss. In 2008 and 2009, caged microplots were assigned to one of three treatments: plants kept aphid-free (referred to as the control), plants that experienced a population of 250 aphids per plant (integrated pest management [IPM]), and plants that experienced unlimited aphid population growth (unlimited). The population growth rate of aphids in the unlimited treatment for the 10 d after the application of insecticides to the IPM treatment was calculated using linear regression. The linear equation was solved to determine the mean number of days between the ET and the EIL for an aphid population in absence of predators. The number of days was determined to be 6.97 +/- 1.11 d. The 2-yr average yield for the IPM treatment was 99.93% of the control treatment. Our study suggests the current soybean aphid ET of 250 aphids per plant can effectively protect yield even if the impact of natural enemies is reduced.  相似文献

The purpose of this study was to examine the association between cardiorespiratory fitness, ectopic triglyceride accumulation, and insulin sensitivity among youth with and without type 2 diabetes. Subjects included 137 youth ages 13-18 years including 27 with type 2 diabetes, 97 overweight normoglycemic controls, and 13 healthy weight normoglycemic controls. The primary outcome measure was cardiorespiratory fitness defined as peak oxygen uptake indexed to fat free mass. Secondary outcomes included liver and muscle triglyceride content determined by (1)H-magnetic resonance spectroscopy and insulin sensitivity determined by frequently sampled intravenous glucose tolerance test. Despite similar measures of adiposity, peak oxygen uptake was 11% lower (38.9 ± 7.9 vs. 43.9 ± 6.1 ml/kgFFM/min, P = 0.002) and hepatic triglyceride content was nearly threefold higher (14.4 vs. 5.7%, P = 0.001) in youth with type 2 diabetes relative to overweight controls. In all 137 youth, cardiorespiratory fitness was negatively associated with hepatic triglyceride content (r = -0.22, P = 0.02) and positively associated with insulin sensitivity (r = 0.29, P = 0.002) independent of total body and visceral fat mass. Hepatic triglyceride content was also negatively associated with insulin sensitivity (r = -0.35, P < 0.001), independent of adiposity, sex, age, and peak oxygen uptake. This study demonstrated that low cardiorespiratory fitness and elevated hepatic triglyceride content are features of type 2 diabetes in youth. Furthermore, cardiorespiratory fitness and hepatic triglyceride are associated with insulin sensitivity in youth. Taken together, these data suggest that cardiorespiratory fitness and hepatic steatosis are potential clinical biomarkers for type 2 diabetes among youth.  相似文献

Primates are among the most threatened of animal taxa, and it is therefore increasingly important to examine ways to monitor their health in the wild. We here investigate trends in nutrition and health in Japanese macaques (Macaca fuscata yakui) under natural conditions using a common tool from human and veterinary health practices: urinalysis. Between October 2007 and August 2009, we collected 103 urine samples ad libitum from 29 identified individuals of a group of macaques on Yakushima Island. We used multireagent dipstick analysis to examine variation in 10 urine chemistry parameters across subjects in combination with systematic monitoring of clinical signs of disease (via focal sampling). Positive tests tended to be transient and weak, i.e., analytes detected in trace amounts, and were not associated with clinical signs of disease. In addition, most urine samples we collected were highly concentrated (urine specific gravity ≥1.030), which could have increased the likelihood of detecting clinically insignificant amounts of analytes that can occur in urine. However, we found that ketone bodies, which are not normally found in urine, were more prevalent during winter than summer, which may indicate that Japanese macaques inhabiting the richest habitat available to the species nonetheless experience an energy deficit during periods of low food quality and high thermoregulatory costs. We conclude that dipstick urinalysis can be used to examine general trends in nutrition and health, but that results must be interpreted with care because positive tests may not reflect clinically significant urological conditions in many cases.  相似文献
