Crystal proteins from Bacillus thuringiensis serovar. medellin   总被引:1,自引:0,他引:1  
Colombian strains 163-131 and 24-726 of the Bacillus thuringiensis serovar. medellin (Btmed), serotype H-30, are very toxic to mosquito larvae. Strain 24-726 was serologically and biochemically characterized. It is almost identical to the reference-strain 163-131. The parasporal inclusion of Btmed strain 163-131 was analysed by electron microscopy. The crystal protein matrix was very similar to that observed in B. thuringiensis serovar. israelensis (Bti). Aedes aegypti, Anopheles albimanus and Culex quinquefasciatus larvae were exposed to 500× the half-lethal concentration (LC50) of Btmed strains, Bti strain 1884 and B. thuringiensis serovar. morrisoni (Btm) strain PG-14. Mortality of Aedes aegypti occurred within approx. 60 min with the four strains, whereas C. quinquefasciatus mortality was three times slower with Btmed than with strains 1884 and PG14. The onset mortality of Anopheles albimanus starts when other species are already dead. The thermolabilities of the mosquitocidal activities of the crystal proteins were tested by incubation of cultures of 20 min at various temperatures. Btmed lost all mosquitocidal activity at 73°C, and 1884 and PG-14 at 79°C. Sodium dodecyl sulphate-polyacrylamide gel electrophoresis analysis of the crystals purified from strain 163-131 shows polypeptides at 100 kDa, multiple bands at 80, 75, 70, 67, and 65 kDa, and two doubles at 40–41 and 28–30 kDa. Immunodetection with antibodies raised against Bti toxins shows cross-reaction between the 30-kDa and to a lesser extent the 28-kDa polypeptides of Btmed crystals and Cyt A of Bti. Summary This study highlights data about the production of a recombinant protein (glyceraldehyde-3-phosphate dehydrogenase) byE. coli HB 101 (GAPDH) during batch and fed-batch fermentations in a complex medium. From a small number of experiments, this strain has been characterized in terms of protein production performance and glucose and acetate influences on growth and recombinant protein production. The present results show that this strain is suitable for recombinant protein production, in fed-batch culture 55 g L–1 of biomass and 6 g L–1 of GAPDH are obtained. However this strain, and especially GAPDH overproduction is sensitive to glucose availability. During fermentations, maximum yields of GAPDH production have been obtained in batch experiments for glucose concentration of 10 g L–1, and in fed-batch experiments for glucose availability of 10 g h–1 (initial volume 1.5 L). The growth of the strain and GAPDH overproduction are also inhibited by acetate. Moreover acetate has been noted as an activator of its own formation.  相似文献

Short photoperiods followed by low temperatures induced cold hardiness in Acer negundo, Viburnum plicatum tomentosum, and Weigela florida. Hardiness was also obtained under long days and natural fall temperatures if the leaves were removed, either manually or by low temperature. Similarly, removal of leaves from plants exposed to long days at 5° brought about an accelerated rate of hardening. These observations suggested the presence of a hardiness inhibitor in the leaves which was counteracted by short days or removal of the leaves.  相似文献

The effect of methyltestosterone given with feed during one and two months on the state of gonads, the level of thyroid, and sex steroid hormones in blood and on rheoreaction is studied in juvenile rainbow trout Parasalmo mykiss (=Oncorhynchus mykiss) raised under unfavorable conditions (tropics, Vietnam). In comparison with the control, in the experimental group, the part of sterile specimens was higher (83 vs. 35%), the part of males was lower (17 vs. 40%), while females and hermaphrodites were absent. At prolonged starvation (12 days), the experimental specimens demonstrated lower increase of the level of thyroid and sex steroid hormones in blood than that in the control specimens. Methyltestosterone modified the behavior of rainbow trout in water flow: delivery of the hormone in food during 1 month increased the part of fish moving against the flow and suppressed movement both upstream and downstream during 2 months. Thus, methyltestosterone negatively influences the physiological condition and adaptation capacity of rainbow trout.  相似文献

Chrysanthemum stunt viroid (CSVd), the smallest plant pathogen known to infect chrysanthemums, is a single-stranded circular RNA viroid that induces stunting that results in an overall height reduction of 30–50 % in mature plants. A catalytic single-chain variable antibody, 3D8 scFv, which exhibits intrinsic DNase and RNase activities, was expressed in chrysanthemums to generate transgenic plant resistance to CSVd infection. Moreover, a codon-optimized version of the 3D8 scFv gene for chrysanthemums was also transformed into plants; these codon-optimized transgenic chrysanthemums expressed twice as much 3D8 scFv and displayed 60 % more resistance to CSVd infection, compared with transgenic chrysanthemums harboring the original 3D8 scFv gene. CSVd challenge experiments with codon-optimized and original 3D8 scFv-transgenic chrysanthemums showed that CSVd in newly produced leaves of both codon-optimized and original 3D8 scFv-transgenic plants was not detected by RT-PCR. This is the first report describing the development of a CSVd-resistant chrysanthemum harboring a catalytic single-chain antibody, 3D8 scFv, which has intrinsic RNase activity.  相似文献

This is the first study on the ultrastructure of the zoospores of Ichthyodinium chabelardi, a parasitoid of the fish egg and early larval stages. The zoospores were characterized by the cell structure specific for dinoflagellates; particularly, cells contained large trichocysts and the “dinokaryon”-type nucleus. An unusual large electron-transparent zone was the only significant difference from the “classical” cell structure in Dinoflagellata. We did not find cell structures for the penetration to the host cell (microtubular basket, conoid, or secretory organelles such as rhoptries). The data on the fine structure of the zoospores of I. chabelardi agree with the results of molecular phylogeny; this allows us argue that excluding this species from Dinoflagellata and assigning it to Protalveolata was a mistake.  相似文献

Following the first report of Opisthorchis viverrini infection in a domestic duck in Phu My District of Binh Dinh Province, Central Vietnam, many other cases were observed in the province. We determined the infection rate and intensity of O. viverrini infection in ducks in 4 districts of the province. A total of 178 ducks were randomly selected from 34 farms for examination of flukes in the liver and gall bladder. An infection rate of 34.3% (range 20.7-40.4% among districts) was found; the intensity of infection was 13.8 worms per infected duck (range 1-100). These findings show the role of ducks as a host for O. viverrini, duck genotype, which is sympatric with the human O. viverrini genotype in this province. It also stresses the need for investigations on the zoonotic potential and the life cycle of this parasite.  相似文献
