Enriched bottle experiments were conducted in situ during winter (January and February) and summer (July and August) 2001 to examine the effects of nutrient enrichments (+ N, + P and + NP) on phytoplankton in Bizerte Lagoon, Tunisia. Chlorophyll a (Chl a), ranging from 3.05 μg L−1 in winter to 4.52 μg L−1 in summer, was dominated by the small size-faction (<5 μm) during both seasons. However, the contribution of the large size-fraction (5-200 μm) to Chl a increased from winter (26%) to summer (37%). Similarly, the carbon biomass of the 5-200 μm algae increased during the July/August period that was characterised by the high proliferation of several diatom taxa. In winter, N was the limiting element for phytoplankton growth. Its addition alone (+ N) or with P (+ NP) increased both the <5 μm and 5-200 μm Chl a concentrations. There was no change in the phytoplankton size structure, with the small cells dominating the final algal biomass in all treatments after 5 days. In summer, N and P limited the phytoplankton, but small and large algae exhibited diverse responses to different nutrient enrichments: addition of P increased the Chl a only in the 5-200 μm fraction, the + N treatment enhanced both size classes, and the NP fertilisation mostly stimulated the biomass of large cells. Consequently, the N and P addition in summer was followed by a significant change in the phytoplankton size structure, since both size-fractions contributed equally to the final Chl a biomass. Within the 5-200 μm algal community, various taxa had diverse responses to the nutrient supply during both seasons, leading to a change in the final community composition. The autotrophic flagellates appeared to grow well under N-deficient conditions. In contrast, diatom growth and biomass were mostly stimulated by the N enrichment while dinoflagellates exhibited the highest increase in their growth and biomass with P fertilisation. Our results suggest that the increasing anthropogenic supply of nutrients in the lagoon may influence algal dynamics as well as productivity in different ways depending on the nutrient composition. 相似文献

An indispensable role for oligodendrocytes in the protection of axon function and promotion of neuronal survival is strongly supported by the finding of progressive neuron/axon degeneration in human neurological diseases that affect oligodendrocytes. Imaging and pathological studies of the CNS have shown the presence of neuroaxonal injury in progressive multifocal leukoencephalopathy (PML), a demyelinating disease of the CNS, resulting from destruction of oligodendrocytes upon productive replication of the pathogenic neurotropic polyomavirus JC. Here, we examined the extracellular factors involved in communication between oligodendrocytes and neurons. Culturing cortical neurons with conditioned medium (CM) from rat CG4 oligodendrocytic cells that express the JCV agnoprotein showed that CXCL5/LIX, which is a chemokine closely related to the human CXCL5/ENA78 and CXCL6/GCP-2 chemokines, is essential for neuronal cell survival. We found that in CM from agnoprotein-producing CG-4 cells level of CXC5/LIX is decreased compared to control cells. We also demonstrated that a reduced expression of CXCL5/LIX by CG4 GFP-Agno cells triggered a cascade of signaling events in cortical neurons. Analysis of mitogen-activated protein kinases (MAPK) and glycogen synthase kinase (GSK3) pathways showed that they are involved in mechanisms of neuronal apoptosis in response to the depletion of CXCL5/LIX signaling. These data suggest that agnoprotein-induced dysregulation of chemokine production by oligodendrocytes may contribute to neuronal/axonal injury in the pathogenesis of PML lesions. 相似文献

Diabetes mellitus (DM) is commonly associated with metabolic and cardiac dysfunctions. The aim of this study was to examine the effect of ghrelin on metabolic and cardiac dysfunctions in a type-2 diabetes mellitus (T2DM) rat model. For this, 48 male adult Sprague-Dawley rats were divided equally into 4 groups: Group I, fed normal chow, served as normal control group; Groups II-IV, were fed a high-fat diet for 2?weeks followed by injection of streptozotocin (STZ) (35?mg/kg body mass) to create a model of T2DM; Group II, were not treated; Group III, were treated with the vehicle (saline); Group IV, were treated with ghrelin (40?μg/kg body mass) twice daily for 10 days. The untreated diabetic rats showed a significant increase in serum fasting blood glucose, insulin homeostasis model assessment (HOMA) index, triglycerides (TGs), low-density lipoprotein cholesterol (LDL-C), total serum cholesterol (TC), and body mass, with a decrease in high-density lipoprotein cholesterol (HDL-C) (p?< 0.05). Hearts isolated from diabetic rats showed a significant increase in myocardial fat content, a significant decrease in GLUT4, and an increase in acyl-CoA oxidase enzyme mRNA (p?< 0.05). Ghrelin administration for 10?days caused a significant improvement in lipid profile, HOMA index, and body mass, and significantly corrected the myocardial mass, significantly reduced the fat content of the myocardium, significantly increased GLUT4, and decreased acyl CoA oxidase mRNA (p?< 0.05). Thus, ghrelin improves both the metabolic functions and the disturbed energy metabolism in the cardiac muscle of obese diabetic rats. 相似文献

The purpose of this study was to develop a new therapeutic approach for atorvastatin (ATV) adopting nanostructured polymeric micelles for its controlled delivery to the cancer cells. Amphiphilic block copolymers of stearyl chitosan (SC) and sulfated stearyl chitosan (S-SC) that could self assemble to form polymeric micelles with different degree of substitution (DS) were synthesized and characterized. The synthesized chitosan derivatives were able to self assemble and form micelles encapsulating ATV with critical micellar concentrations ranging from 6.9 to 21μg/ml, drug-loading ranging from 40% to 84.1% and encapsulation efficiency ranging from 10.4% to 35%. ATV caused a significant decrease in particle size and zeta potential of both SC and S-SC micelles. Micelles encapsulating ATV exhibited a sustained release and more cytotoxic activity against MCF 7 and HCT 116 cell lines than ATV alone. The 50% cellular growth inhibition (IC50%) of the drug decreased from 10.4 to 3.7 in case of MCF 7 and from 9.4 to 3.4 in case of HCT 116 after its loading in micelles. These results indicate that SC ATV polymeric micelles can be considered as a promising system for site specific controlled delivery of ATV to tumor cells. 相似文献

Conducting polymers have generated a great deal of interest because of their physical and chemical properties as well as their potential application in industry particularly in packaging applications. However one of short comings of most conducting polymer is that they are often formed as intractable films that are difficult to process. To overcome this problem we have incorporated conducting polymer, namely polyaniline into sheets of paper in order to create new composite material which combine the universal properties of paper product with the chemical and electrically conducting properties of the conducting polymer. Paper conducting polymer composite have been prepared by polymerizing aniline directly onto the paper sheet using ammonium peroxydisulfate (APS) as an oxidant at different temperatures. The prepared composite was characterized by FT-IR and SEM. The thermo-oxidative degradation was studied by thermo gravimetric analysis (TGA); electrical conductivities measurements of the composites were significantly increased over those of the precursor paper. 相似文献

The silver-nanoparticle (AgNP) containing paper was successfully prepared. The AgNP is deposited by the in situ reduction of silver nitrate on the acrylamide grafted bagasse paper sheets in the presence of citrate molecules as stabilizing agent. In the present paper, grafting of acrylamide onto bagasse paper sheets using potassium persulfate was carried out under the influence of microwave radiations (MWR). The modified paper sheets were characterized by Fourier transform infrared spectroscopy (FTIR), UV-spectroscopy, and scanning electron microscopy (SEM). Antimicrobial activities of the prepared paper sheets were also investigated against G+ve bacterium Staphylococcus aureus, G-ve bacterium Pseudomomas aeruginosa, and yeast Candida albicans, which are model microorganisms for testing bactericidal properties. The photo-induced transformation of monuron (3-(4-chlorophenyl)-1,1 dimethylurea) was investigated in an aqueous solution containing nitrates and nitrites at 310 nm and 365 nm, respectively. In both NO(3)(-) and NO(2)(-) conditions, the degradation of monuron followed pseudo-first order kinetics. The intermediate products were identified by GC-MS, and the nitration, hydroxylation and coupling reactions were determined. In addition, the oxidation of the N-terminus group, the substitution of chlorine by ˙OH and the nitration by ˙NO(2) radical onto the phenyl ring were observed. The photo-induced transformation of monuron was studied under variable conditions of pH, inducer concentration, substrate concentration, humic acids, oxygen content and salts used as hydroxyl radical scavengers. The photodegradation rates were strongly influenced by all the above parameters. The degradation of monuron was also studied in the dark and in the presence of NO(2)(-) as well as in an aqueous solution with the addition of hydrogen peroxide. 相似文献
