A large inbred Italian kindred with 10 members homozygous for the rare Rh gene complex-D-is described. The propositus has immune antibody to a high-incidence Rh antigen: her baby required exchange transfusion. None of the other 9-D-/-D- individuals has made antibody even though 2 were women with children. This kindred supports previous observations of the excess of consanguinity among the parents of -D-homozygotes. 40.9% (instead of the 25% expected) of the offspring of -D-heterozygous parents are -D-homozygotes. 相似文献

Chlamydia trachomatis is a Gram-negative eubacterium with a dimorphic developmental cycle and obligate intracellular growth in the eucaryotic host. The Dam transmethylase of Escherichia coli methylates at the N6 position of adenine in the sequence 5'-GATC-3' and the Dcm transmethylase adds methyl groups to the C5 position of the internal cytosines in the sequences 5'-CCWGG-3'. T cell function is a critical determinant of immune responses as well as susceptibility to allergic diseases. Activated T cells can differentiate into effectors whose cytokine profile is limited to type 1 (IFN-gamma-dominant) or type 2 (IL-4-, IL-5-dominant) patterns. To investigate mechanisms that connect extracellular stimuli with the regulation of effector T cell function, we have measured immune responses of transgenic mice whose NF-kappa B/Rel signaling pathway is inhibited in T cells. Surprisingly, these mice developed type 2 T cell-dependent responses (IgE and eosinophil recruitment) in a model of allergic pulmonary inflammation. In contrast, type 1 T cell responses were severely impaired, as evidenced by markedly diminished delayed-type hypersensitivity responses, IFN-gamma production, and Ag-specific IgG2a levels. Taken together, these data indicate that inhibition of NF-kappa B can lead to preferential impairment of type 1 as compared with type 2 T cell-dependent responses. 相似文献

The 297bp gene coding for the HIV-1 protease was chemically synthesized and expressed in E. coli. Single amino acid substitutions (Arg 87 - greater than Lys; Arg 87 - greater than Glu) were introduced in the C-terminally located conserved region GlyArgAsn of the protease gene in the wild-type clone. The products of the mutant and the wild-type clones were expressed at approximately similar levels at 30 minutes of induction but the mutant protease proteins accumulated as a function of time of induction unlike the wild-type protease which declined after 60 minutes. The mutants were completely devoid of proteolytic activity as determined in assays employing as substrates a synthetic nonapeptide and a gag related recombinant polyprotein. 相似文献

The distribution of albumin variants amongst several Mexican Indian and non-Indian (Mestizo) groups was studied. Of the former, a total of 1606 individuals belonging to 11 different tribes were examined with an overall frequency of 1.5% of Albumin Mexico fairly uniformly distributed in all four main linguistic groups. The 2548 Mestizos studied belong in six groups, two from Mexico City and four from elsewhere in the country. The first of the Mexico City groups consisted of 1313 individuals randomly chosen from the outpatient clinic of the Instituto Nacional de la Nutrición, while the rest included healthy individuals. The overall frequency was 1.9%, also fairly evenly distributed, with no difference between the hospital population and the others. No anomalous albumin, other than Albumin Mexico was encountered in the whole study. It is concluded that the similarity between Indians and Mestizos is due to the high Indian component of the latter and that the presence of albumin Mexico is a good anthropological marker for this region of the world. 相似文献

Little is known about the role of the hindbrain during development of spinal network activity. We set out to identify the activity patterns of reticulospinal (RS) neurons of the hindbrain in fictively swimming (paralyzed) zebrafish larvae. Simultaneous recordings of RS neurons and spinal motoneurons revealed that these were coactive during spontaneous fictive swim episodes. We characterized four types of RS activity patterns during fictive swimming: (i) a spontaneous pattern of discharges resembling evoked high-frequency spiking during startle responses to touch stimuli, (ii) a rhythmic pattern of excitatory postsynaptic potentials (EPSPs) whose frequency was similar to the motoneuron EPSP frequency during swim episodes, (iii) an arrhythmic pattern consisting of tonic firing throughout swim episodes, and (iv) RS cell activity uncorrelated with motoneuron activity. Despite lesions to the rostral spinal cord that prevented ascending spinal axons from entering the hindbrain (normally starting at approximately 20 h), RS neurons continued to display the aforementioned activity patterns at day 3. However, removal of the caudal portion of the hindbrain prior to the descent of RS axons left the spinal cord network unable to generate the rhythmic oscillations normally elicited by application of N-methyl-d-aspartate (NMDA), but in approximately 40% of cases chronic incubation in NMDA maintained rhythmic activity. We conclude that there is an autonomous embryonic hindbrain network that is necessary for proper development of the spinal central pattern generator, and that the hindbrain network can partially develop independently of ascending input. 相似文献

Ongoing climate change can alter conditions for plant growth, in turn affecting ecological and social systems. While there have been considerable advances in understanding the physical aspects of climate change, comprehensive analyses integrating climate, biological, and social sciences are less common. Here we use climate projections under alternative mitigation scenarios to show how changes in environmental variables that limit plant growth could impact ecosystems and people. We show that although the global mean number of days above freezing will increase by up to 7% by 2100 under “business as usual” (representative concentration pathway [RCP] 8.5), suitable growing days will actually decrease globally by up to 11% when other climatic variables that limit plant growth are considered (i.e., temperature, water availability, and solar radiation). Areas in Russia, China, and Canada are projected to gain suitable plant growing days, but the rest of the world will experience losses. Notably, tropical areas could lose up to 200 suitable plant growing days per year. These changes will impact most of the world’s terrestrial ecosystems, potentially triggering climate feedbacks. Human populations will also be affected, with up to ~2,100 million of the poorest people in the world (~30% of the world’s population) highly vulnerable to changes in the supply of plant-related goods and services. These impacts will be spatially variable, indicating regions where adaptations will be necessary. Changes in suitable plant growing days are projected to be less severe under strong and moderate mitigation scenarios (i.e., RCP 2.6 and RCP 4.5), underscoring the importance of reducing emissions to avoid such disproportionate impacts on ecosystems and people. 相似文献

We evaluated the effects of X-ray irradiation on larvae of Ceratitis capitata (Wiedemann) (Diptera: Tephritidae), with the aim of finding a treatment that prevented adult fly emergence, yet did not adversely affect larval quality as rearing hosts for the parasitoid Diachasmimorpha longicaudata (Ashmead) (Hymenoptera: Ichneumonidae). Two experiments were carried out. In the first experiment, small numbers of larvae were held in Petri dishes with and without small amounts of rearing media and then irradiated with doses of X-rays ranging from 0 to 8333.6 R (equivalent to 80 Gy). In the second experiment, higher numbers of larvae were held in a manner resembling mass-rearing conditions, and were then irradiated with X-rays ranging from 0 to 10,417 R (equivalent to 100 Gy). In both experiments, the only factor that significantly affected fly emergence was irradiation of larvae. Fly emergence decreased markedly as the irradiation dose increased, and complete suppression of fly emergence was achieved at 6250.2 R (equivalent to 60 Gy) when larvae were irradiated in small batches with or without rearing media. Irradiation also affected the fertility of those flies that did emerge following treatment. In the second experiment, we found the parasitoids reared from irradiated larvae produced a higher parasitism rate and a higher number of female offspring than did parasitoids reared from control (non-irradiated) larvae. Mean fecundity of F1 parasitoids reared from irradiated larvae were affected positively by irradiation only at the 8333.6 R (80 Gy) dose. Our results show that X-ray irradiation can be used to inhibit fruit fly adult emergence and that irradiated larvae are at least as good a rearing substrate as non-irradiated larvae. Future studies should focus on the adjustment of our findings to a mass-rearing scale. 相似文献

Agricultural intensification is linked to reduced species richness and may limit the effectiveness of predators in agricultural systems. We studied the abundance, diversity, and species composition of wood-nesting ants and frequency of parasitism of poneromorph ants in coffee agroeco systems and a forest fragment in Chiapas, Mexico. In three farms differing in shade management and in a nearby forest fragment, we surveyed ants nesting in rotten wood. We collected pupae of all poneromorph ants encountered, and incubated pupae for 15 d to recover emerging ant parasites. If no parasites emerged, we dissected pupae to examine for parasitism. Overall, we found 63 ant morphospecies, 29 genera, and 7 subfamilies from 520 colonies. There were no significant differences in ant richness or abundance between the different sites. However, there were significant differences in the species composition of ants sampled in the four different sites. The parasitism rates of ants differed according to site; in the forest 77.7% of species were parasitized, and this number declined with increasing intensification in traditional polyculture (40%),commercial polyculture (25%), and shade monoculture (16.6%). For three of four poneromorph species found in >1 habitat, parasitism rates were higher in the more vegetatively complex sites. The result that both ant species composition and ant parasitism differed among by site indicates that coffee management intensification affects wood-nesting ant communities. Further, coffee intensification may significantly alter interactions between ants and their parasites, with possible implications for biological control in coffee agroecosystems. 相似文献
