Magnesium has important structural, catalytic and signaling roles in cells, yet few tools exist to image this metal ion in real time and at subcellular resolution. Here we report the first genetically encoded sensor for Mg2+, MagFRET-1. This sensor is based on the high-affinity Mg2+ binding domain of human centrin 3 (HsCen3), which undergoes a transition from a molten-globular apo form to a compactly-folded Mg2+-bound state. Fusion of Cerulean and Citrine fluorescent domains to the ends of HsCen3, yielded MagFRET-1, which combines a physiologically relevant Mg2+ affinity (K
d = 148 µM) with a 50% increase in emission ratio upon Mg2+ binding due to a change in FRET efficiency between Cerulean and Citrine. Mutations in the metal binding sites yielded MagFRET variants whose Mg2+ affinities were attenuated 2- to 100-fold relative to MagFRET-1, thus covering a broad range of Mg2+ concentrations. In situ experiments in HEK293 cells showed that MagFRET-1 can be targeted to the cytosol and the nucleus. Clear responses to changes in extracellular Mg2+ concentration were observed for MagFRET-1-expressing HEK293 cells when they were permeabilized with digitonin, whereas similar changes were not observed for intact cells. Although MagFRET-1 is also sensitive to Ca2+, this affinity is sufficiently attenuated (K
d of 10 µM) to make the sensor insensitive to known Ca2+ stimuli in HEK293 cells. While the potential and limitations of the MagFRET sensors for intracellular Mg2+ imaging need to be further established, we expect that these genetically encoded and ratiometric fluorescent Mg2+ sensors could prove very useful in understanding intracellular Mg2+ homeostasis and signaling. 相似文献

Peach palm (Bactris gasipaes) is a multi-purpose palm tree native to tropical Latin America, which is predominantly cultivated by smallholders in agroforestry systems. The fruits are rich in starch and contribute importantly to food security and the cash income of farmers who cultivate them. Complex value chains have emerged that link producers to consumers, but irregular product quality and market chain inequalities undermine the economic well-being of producers and retailers. Peach palm is genetically diverse, but screening for traits of commercial and nutritional interest is required to enhance the use of its genetic resources. Alliances between public organizations and private enterprises are needed to realize the potential for processing novel products from peach palm, especially in the pharmaceutical and cosmetic sectors. The diverse challenges that emerge at different stages of production, processing and marketing require participatory research that directly involves stakeholders from the beginning. 相似文献

Most landscape genetic studies assess the impact of landscape elements on species' dispersal and gene flow. Many of these studies perform their analysis on all possible population pairs in a study area and do not explicitly consider the effects of spatial scale and population network topology on their results. Here, we examined the effects of spatial scale and population network topology on the outcome of a landscape genetic analysis. Additionally, we tested whether the relevant spatial scale of landscape genetic analysis could be defined by population network topology or by isolation‐by‐distance (IBD) patterns. A data set of the wetland grasshopper Stethophyma grossum, collected in a fragmented agricultural landscape, was used to analyse population network topology, IBD patterns and dispersal habitats, using least‐cost transect analysis. Landscape genetic analyses neglecting spatial scale and population network topology resulted in models with low fits, with which a most likely dispersal habitat could not be identified. In contrast, analyses considering spatial scale and population network topology resulted in high model fits by restricting landscape genetic analysis to smaller scales (0–3 km) and neighbouring populations, as represented by a Gabriel graph. These models also successfully identified a likely dispersal habitat of S. grossum. The above results suggest that spatial scale and potentially population network topology should be more explicitly considered in future landscape genetic analyses. 相似文献

Herbivore‐induced plant volatiles are often involved in direct and indirect plant defence against herbivores. Linalool is a common floral scent and found to be released from leaves by many plants after herbivore attack. In this study, a linalool/nerolidol synthase, FaNES1, was overexpressed in the plastids of chrysanthemum plants (Chrysanthemum morifolium). The volatiles of FaNES1 chrysanthemum leaves were strongly dominated by linalool, but they also emitted small amount of the C11‐homoterpene, (3E)‐4,8‐dimethyl‐1,3,7‐nonatriene, a derivative of nerolidol. Four nonvolatile linalool glycosides in methanolic extracts were found to be significantly increased in the leaves of FaNES1 plants compared to wild‐type plants. They were putatively identified by LC‐MS‐MS as two linalool–malonyl–hexoses, a linalool–pentose–hexose and a glycoside of hydroxy–linalool. A leaf‐disc dual‐choice assay with western flower thrips (WFT, Frankliniella occidentalis) showed, initially during the first 15 min of WFT release, that FaNES1 plants were significantly preferred. This gradually reversed into significant preference for the control, however, at 20–28 h after WFT release. The initial preference was shown to be based on the linalool odour of FaNES1 plants by olfactory dual‐choice assays using paper discs emitting pure linalool at similar rates as leaf discs. The reversal of preference into deterrence could be explained by the initial nonvolatile composition of the FaNES1 plants, as methanolic extracts were less preferred by WFT. Considering the common occurrence of linalool and its glycosides in plant tissues, it suggests that plants may balance attractive fragrance with ‘poor taste’ using the same precursor compound. 相似文献

Food webs are increasingly evaluated at the landscape scale, accounting for spatial interactions involving different nutrient and energy channels. Also, while long viewed as static, food webs are increasingly seen as dynamic entities that assemble during vegetation succession. The next necessary step is, therefore, to link nutrient flows between ecosystems to local food web assembly processes. In this study, we used a 100-year salt marsh succession in which we investigated the long-term changes in food web organization, especially focusing on the balance between internal versus external nutrient sources. We found that during food web assembly, the importance of internal (terrestrial) nutrient cycling increases at the expense of external (marine) inputs. This change from external to internal nutrient cycling is associated with strong shifts in the basis of energy channels within the food web. In early succession, detritivores are mostly fuelled by marine inputs whereas in later succession they thrive on locally produced plant litter, with consequences for their carnivores. We conclude that this 100 years of food web assembly proceeds by gradual decoupling of terrestrial nutrient cycling from the marine environment, and by associated rearrangements in the herbivore and detritivore energy channels. High litter mass is hypothesized to produce an invader-directed invasion by changing ecosystem properties such as nutrient cycling rates and light availability. An invasive plant species that stimulates litter accumulation may induce a positive feedback when it benefits from high litter conditions. Phalaris arundinacea is an invasive wetland grass that may induce positive litter feedback, as it produces abundant litter that varies in quality due to a wide range of foliar C:N content. In this study we investigated the range of growth responses within native and invasive genotypes of Phalaris that varied in initial foliar C:N levels (high C:N content was present in the invasive genotypes) when grown under varying litter mass. Overwintering with high litter reduced establishing tiller survivorship and the presence of litter delayed tiller emergence by 2 weeks. Overall, genotypes exhibited high trait plasticity in response to litter. Our results indicate that high litter mass can stimulate Phalaris growth, specifically for the genotypes with high initial C:N foliar tissue. Additionally, genotypes with initially high C:N ratios exhibited plastic responses consistent with a Master-of-some strategy indicating that their performance under high litter may depend upon the nutrient conditions under which they are grown. This study provides evidence for conditions that may lead to a positive feedback in Phalaris’ introduced range. Future studies should investigate how changing litter quantity alters nutrient cycling and competitor growth. 相似文献
