以在宿主菌株BL21(DE3)中成功表达的重组金黄色葡萄球菌a-溶血素蛋白为研究对象, 分析比较通过凝胶过滤层析(Gel filtration chromatography)和镍柱亲和层析纯化试剂盒(Ni-NTA spin columns)纯化所得到的重组蛋白的蛋白含量和生物特性方面的差异。SDS-PAGE分析检测纯化产物, Bradford法测定蛋白含量, 兔红细胞测定半数溶血效价。结果显示这2种方法得到的纯化产物在53 kD处均呈现单一清晰带, 达电泳级纯度。与此同时, 凝胶过滤对目的蛋白的纯化率为14.04%, 蛋白含量为0.337 mg/mL, 溶血活性为1519 HU/mg; 镍柱亲和层析的纯化率为17.5%, 蛋白含量为0.35 mg/mL, 溶血活性为1463 HU/mg。由此可见, 凝胶过滤得到的纯化产物在蛋白含量和蛋白活性方面丝毫不亚于镍柱亲和层析纯化试剂盒。  相似文献

甘珀酸干预对大鼠脑缺血再灌注损伤的影响   总被引:1,自引:0,他引:1  
目的观察缝隙连接阻断剂甘珀酸对局灶性脑缺血/再灌注损伤的影响。方法采用大鼠大脑中动脉阻塞再灌流模型(MCAO),将动物随机分为脑缺血60min再灌注(MCAO)组,脑缺血再灌注加甘珀酸干预(MCAO+CBX)组和假手术组(sham)。采用尼氏染色显示脑梗死灶并计算梗死灶体积;应用免疫荧光与TUNEL染色法分别观察脑缺血后3d与7d不同时间点缺血边缘区胶质纤维酸性蛋白(GFAP)的表达和细胞凋亡情况。结果(1)缺血后3d、7d MCAO+CBX组大鼠梗死体积小于MCAO组,3d、7d MCAO+CBX组大鼠梗死体积较MCAO组分别缩小5%和4.6%;(2)缺血后3d、7d于缺血边缘区可见大量TUNEL阳性染色细胞,且MCAO组大鼠缺血边缘区细胞凋亡数目明显多于MCAO+CBX大鼠(P〈0.001);(3)缺血后3d和7d组缺血边缘区GFAP表达明显增强,3d的MCAO组与MCAO+CBX组大鼠缺血边缘区GFAP的表达均较假手术组强(P〈0.05),7d的MCAO+CBX组大鼠缺血边缘区GFAP的表达较假手术组强(P〈0.001),但明显弱于MCAO组大鼠(P〈0.01);结论缝隙连接阻断剂甘珀酸可减少大鼠大脑中动脉阻塞后脑梗死体积,其机制可能与阻断缝隙连接后缺血边缘区神经元凋亡降低有关,星型胶质细胞的反应性变化参与了该过程。  相似文献

Cationic cotton was prepared by a designed two-bath pad-bake process with 3-chloro-2-hydroxypropyltrimethylammonium chloride as cationizing reagent to realize recycle utilization of the reagent and continuous processing of cationization. Experiments showed that 8.0% (o.w.bath) of the reagent, 1:1 of molar ratio of sodium hydroxide to the reagent, 60 °C and 6 min of baking temperature and time were selected for cationization and the obtained cationic cotton was suitable for application in salt-free reactive dyeing. The structures of both the untreated and cationic fibers were investigated by X-ray diffraction and scanning electronic microscopy. Higher dye utilization and color yields could be realized on the cationic cotton than that on the untreated one in the conventional dyeing. Levelness dyeing and good fastness properties of the dyes on the cationic fabrics were obtained. Besides, colorimetric properties and mechanical strength of the dyed fabrics were both evaluated to show applicability of this preparation process of cationic cotton.  相似文献

TRAF2 has an important function in mediating the TNF-R signaling pathway toward activation of NF-κB and JNKs. Here we reveal a novel function of TRAF2 in the epidermal growth factor (EGF) signaling pathway. Knockdown of TRAF2 blocked EGF-induced AP-1 activity and anchorage- independent cell transformation. Notably, we showed that EGF induces ribosomal S6 kinase 2 (RSK2) ubiquitination, and knocking down TRAF2 suppresses ubiquitination of RSK2 induced by EGF. We also found that TRAF2 affects RSK2 activity through RSK2 ubiquitination. RSK2 plays a critical role in AP-1 activity mediated through CREB and c-Fos, which regulates anchorage-independent cell transformation. In addition, TRAF2 is overexpressed in colon cancer and required for colon cancer development, suggesting that TRAF2 might be a potential molecular target for cancer prevention and treatment.  相似文献

Interleukin-22 (IL-22), an IL-10 family cytokine, mediates the crosstalk between leukocytes and epithelial cells. Previous studies reported that IL-22 expresses in mouse brain, and the rat PC12 cells are responsive to IL-22 stimulation. However, the biological roles of IL-22 in neuronal cells remain largely unknown. We show here that IL-22 activates Stat3, p38 mitogen-activated protein kinases (MAPK), and Akt pathways and inhibits Erk/MAPK pathway in na?ve PC12 cells. We further demonstrate that IL-22 protects na?ve PC12 cells from serum starvation-induced cell death via the Jak1/Stat3 and Akt pathways. We also show that IL-22 has no effects on na?ve PC12 cell proliferation and cannot protect na?ve PC12 cells from 1-methyl-4-phenylpyridinium (MPP+)-induced cytotoxicity. However, IL-22 exerts a dose-dependent protective effect on MPP+-induced neurodegeneration in nerve growth factor-differentiated PC12 cells. Overall, our data suggest that IL-22 might play a role in neurological processes. To our knowledge, this is the first report showing that IL-22 confers a neuroprotective function, which may provide a new therapeutic option for treatment of neurodegenerative diseases.  相似文献

Floral spurs are regarded as features affecting pollinator behaviour. Anacamptis pyramidalis is regarded as a deceitful, non-rewarding orchid species. In the form fumeauxiana, additional spurs occur on the lateral sepals. In this study we analyse micromorphological and ultrastructural floral features and suggest the mechanism of deception in A. pyramidalis and A. pyramidalis f. fumeauxiana. In f. fumeauxiana, the adaxial surface of the lip, the lip calli, the tips of the lateral sepals, the abaxial and adaxial epidermises of the lip spur, and the lateral sepal spur have a secretory function. Numerous stomata were observed on the abaxial surfaces of spurs and sepals. The characteristic features of the ultrastructure of osmophore cells were noted: dense cytoplasm with numerous profiles of ER, mitochondria, plastids with plastoglobuli and tubular structures, a large nucleus, lipid droplets, and vesicles fusing with the plasmalemma. The similarity of the floral morphology and anatomy, the flowering period, and pollinators of A. pyramidalis, A. pyramidalis f. fumeauxiana and Gymnadenia conopsea suggest a possible food-deceptive mechanism—imitation of nectar presence produced in the spurs of Gymnadenia.  相似文献
