Changes in the Microbiology of Vacuum-packaged Beef   总被引:9,自引:9,他引:0  
The development of the microbial flora on meat stored in vacuum-bags at 0–2° for up to 9 weeks was studied. Although the proportion of lactic acid bacteria increased relative to the aerobic spoilage organisms, the numbers of the latter continued to increase throughout storage. The initial contamination of the meat before vacuum-packaging was important; meat with a very low initial number had lower numbers of bacteria throughout storage for up to 9 weeks and steaks cut from such meat which had been stored always had 1–2 days' additional aerobic shelf life at 4°. Spoilage of these steaks was due either to slime formation and off-odour associated with high counts of presumptive Pseudomonas spp., or by discoloration and souring (lactic acid bacteria). Extract release volume and pH measurements performed on the vacuum-packaged primal joints were only of value in determining the onset of aerobic spoilage when large numbers of Gram negative organisms were present, whereas the titrimetric method of spoilage evaluation of the vacuum-packaged meat showed a correlation with spoilage due to lactic organisms.  相似文献

UNTIL now, there has been no totally satisfactory hypothesis proposed for the hybridization of lactate dehydrogenase in vivo, This is because hybridization in vitro, the only experimental approach, has usually been accomplished by chaotropic agents: freeze-thaw1, guanidine2, 3, urea3, or acid denaturation4. Whatever means the cell uses, it follows5–8 that it is susceptible to environmental influences. We now describe a gentle hybridization between heart and muscle types which can occur in more physiological conditions and which mimics many observations in vivo.  相似文献

The major isoenzymes of -mannosidase (EC 3.2.1.24 [EC] ) and ß-galactosidase(ECf 3.2.1.23 [EC] ) have been separated from cotyledons of gardenpea, Pisum sativum L. (Vicieae), chick pea, Cicer arietinumL. (Cicereae), and cowpea, Vigna unguiculata (L.) Walp. (Phaseoleae).Some of their properties have been determined, including pHoptima, Km values for p-nitrophenyl glycosidc substrates, andthe effects of several inhibitors. Swainsonine, an indolizidinealkaloid, was the most effective inhibitor of mannosidase 1,with I30 values of 5.6 x 10–8 M (cowpea), 1x 10–7M (chick pea) and 2.9 x 19–7 M (pea). The most effectiveinhibitor of ß-galactosidase 2 from all sources wasD-galactonic acid-1,4-lactonwe (-lactone), with Ki values rangingbetween 3.0 and 3.9x 10–3 M. An inhibitor of the E. coliß-galactosidose, p-aminophenyl thio-ß-D-galactopyranoside,did not inhibit any of the legume ß-galctosidases;rather it enhanced the activites of the enzymes from chick peaand cowpea cotyledons. Etiolated hull and seed tissues frompea pods developing in darkness contained similar acid glycosidaseactivities to normal green tissues, thus the chloroplast isan unlikely location for ß-galactosidase 2. The majorß-galactosidasesdetected with an indigogenic substrate (5-bromo-4-chloro-3-indoxyl-ß-D-galactopyranoside)following gel electrophoresis of extracts from pea hull, seedcoats and cotyledons appeared to be different from ß-galactosidase2. The response of leaf tissue to white, blue, red, and far-redlight has been examined. Leaves on plants grown in darknessshow increased cell number, cell volume, and area when exposedto long periods (up to 48 h) of low-intensity red, blue, orfar-red radiation. This is believed to be a photomorphogenicresponse which does not involve photosynthesis. Leaves fromplants exposed to white light during germination do not usuallyrespond to red, blue, or far-red light. An exception to thiswas found for leaf discs which showed a larger increase in areathan the dark controls following exposure to far-red light for24 h. Leaf tissue from light-grown plants responds to high-intensitywhite light, probably through photosynthesis. Discs cut fromdark-grown plants and cultured in white light grow equally wellin air and CO2-free conditions. Application of the photosyntheticinhibitor DCMU reduces growth and chlorophyll formation, however. It is concluded that light, perhaps acting through the phytochromemechanism, has initially a number of morphogenic effects includinginitiation of development of the photosynthetic apparatus. Theresponses to photomorphogenically active radiation do not persistand light effects through photosynthesis are rapidly initiatedand dominate the later stages of leaf growth.  相似文献

1. Forty-three strains of moulds, selected from ten species,were examined to ascertain their capabilities as producers offat, when grown in four different solutions of nutrient inorganicsalts together with sucrose. The three highest yields of feltwere given by Penicillium javanicum van Beyma, P. Soppi Zaleski,and Aspergillus nidulans Eidam, in that order. The fat contenton felt weight was maximal at 34·8 per cent, with P.Soppi, at 28·4 per cent. with Fusarium lini(1), and at25·8 per cent. with A. nidulans; on sugar utilized itwas maximal at 11·4 per cent. with P. Soppi, at 7·9per cent, with A. nidulans, and at 5·6 per cent. withF. lini(1). 2. Other moulds considered worthy of further trial were P. spinulosum,P. aurantiobrunneum, F. graminearum, A. flavus, F. oxysporum,A. clavatus, and P. oxalicum.  相似文献

A new genus and species of osteoglossomorph fish is described from the Eocene Mahenge site in north central Tanzania. The new species is sympatric with the previously described osteoglossomorph † Singida jacksonoides , but is easily distinguished from it by the presence in the new species of teeth on the jaws and entopterygoid. The new species shares characters with the Heterotidinae, such as the two arms of the preopercle being of similar length and the posterior end of the maxilla lying on the dentary, and other characters with the Osteoglossinae (including † Phareodus ), for example the enlarged pectoral fin ray and the shape of the opercular bone. New specimens of † S. jacksonoides also have been recovered from the site, providing more information on the osteology of this fish. The relationships of these two Mahenge osteoglossomorphs are assessed, with both fishes placed within the Osteoglossidae, but of uncertain position in the family.
