Folding of proteins entering the secretory pathway in mammalian cells frequently requires the insertion of disulfide bonds. Disulfide insertion can result in covalent linkages found in the native structure as well as those that are not, so‐called non‐native disulfides. The pathways for disulfide formation are well characterized, but our understanding of how non‐native disulfides are reduced so that the correct or native disulfides can form is poor. Here, we use a novel assay to demonstrate that the reduction in non‐native disulfides requires NADPH as the ultimate electron donor, and a robust cytosolic thioredoxin system, driven by thioredoxin reductase 1 (TrxR1 or TXNRD1). Inhibition of this reductive pathway prevents the correct folding and secretion of proteins that are known to form non‐native disulfides during their folding. Hence, we have shown for the first time that mammalian cells have a pathway for transferring reducing equivalents from the cytosol to the ER, which is required to ensure correct disulfide formation in proteins entering the secretory pathway. 相似文献

Genes essential for the production of a linear, bacterial (1-->3)-beta-
glucan, curdlan, have been cloned for the first time from Agrobacterium sp.
ATCC31749. The genes occurred in two, nonoverlapping, genomic fragments
that complemented different sets of curdlan( crd )-deficient
transposon-insertion mutations. These were detected as colonies that failed
to stain with aniline blue, a (1-->3)-beta-glucan specific dye. One
fragment carried a biosynthetic gene cluster (locus I) containing the
putative curdlan synthase gene, crdS, and at least two other crd genes. The
second fragment may contain only a single crd gene (locus II).
Determination of the DNA sequence adjacent to several locus I mutations
revealed homology to known sequences only in the cases of crdS mutations.
Complete sequencing of the 1623 bp crdS gene revealed highest similarities
between the predicted CrdS protein (540 amino acids) and glycosyl
transferases with repetitive action patterns. These include bacterial
cellulose synthases (and their homologs), which form
(1-->4)-beta-glucans. No similarity was detected with putative
(1-->3)- beta-glucan synthases from yeasts and filamentous fungi.
Whatever the determinants of the linkage specificity of these beta-glucan
synthases might be, these results raise the possibility that
(1-->3)-beta-glucans and (1-->4)-beta-glucans are formed by related
catalytic polypeptides.

The bracts of wild banana species are pigmented by glycosides(probably 3-diglucosides) of four combinations of anthocyanidins:cyanidin-pelargonidin; cyanidin-delphinidin; delphinidin-petunidin-cyanidin-malvidin-peonidin;mal-vidin-peonidin. Stands of groundnut were grown in computer-controlled glasshouseson water stored in an undisturbed soil profile. The maximumsaturation vapour pressure deficit (D) of the air was either1.0, 2.0, 2.5, or 3.0 kPa, and the mean air temperature was27 °C. Transpiration (E), determined from the soil water balance, wasstrongly dependent on D, because D influenced both the fractionof incident solar radiation intercepted by foliage (f) and thetranspiration rate per unit f (E/f). When D exceeded 2 kPa,canopy expansion was restricted and f reduced during early growth,but differences in f diminished as the canopies closed. E/fincreased with D, implying that any restriction of transpirationthrough stomatal closure at large D was outweighted by a steeperhumidity gradient from leaf to air. In all treatments E/f decreased as the soil profile dried. Saturationdeficit per se had little influence on the proportional reductionin E/f with time, even though soil water deficit was considerablygreater at large D. Combining data from different ethnic populations in a study can increase efficacy of methods designed to identify expression quantitative trait loci (eQTL) compared to analyzing each population independently. In such studies, however, the genetic diversity of minor allele frequencies among populations has rarely been taken into account. Population dynamics of the White Stork Ciconia ciconia were studied in Charente-Maritime, France from 1978 to 1996, during which time the number of breeding pairs increased from one to 44. Modal age at first return and first breeding were 2.4 and 3.4 years, respectively. White Storks produced an average of 3.2 fledglings per nest. The average number of fledglings per nest decreased with increasing nest density, probably because of an increase in the number of interactions between breeding birds. Nests surrounded by marshes had slightly greater productivity than nests on peripheral sites. Individual White Storks followed an annual breeding cycle and attempted to breed in 97% of seasons once mature. Nest-site and mate fidelity were high (88 and 83%, respectively). Immigration rate was nearly twice that of emigration during the last few years of the study and recruitment was close to 30%, although underestimated. We modelled survival and recapture probabilities using capture-mark-recapture methods. Adult survival was found to be dependent on age, but not sex. Survival of younger birds varied greatly over the years, whereas survival of older birds was relatively constant and averaged 78%. Survival rates of young birds wintering in the Sahel zone were positively linked to the amount of rainfall in their wintering area. The proximate reason for the population increase was probably immigration of birds from other European countries, probably encouraged by a high adult survival rate. Ringing recoveries indicate that some birds winter in Spain and the high adult survival rate may reflect a change in migratory pattern in recent years.
