Large, rapidly evolving intergenic spacers in the mitochondrial DNA of the salamander family Ambystomatidae (Amphibia: Caudata) 总被引:3,自引:2,他引:3
We report the presence, in the mitochondrial DNA (mtDNA) of all of the
sexual species of the salamander family Ambystomatidae, of a shared 240- bp
intergenic spacer between tRNAThr and tRNAPro. We place the intergenic
spacer in context by presenting the sequence of 1,746 bp of mtDNA from
Ambystoma tigrinum tigrinum, describe the nucleotide composition of the
intergenic spacer in all of the species of Ambystomatidae, and compare it
to other coding and noncoding regions of Ambystoma and several other
vertebrate mtDNAs. The nucleotide substitution rate of the intergenic
spacer is approximately three times faster than the substitution rate of
the control region, as shown by comparisons among six Ambystoma
macrodactylum sequences and eight members of the Ambystoma tigrinum
complex. We also found additional inserts within the intergenic spacers of
five species that varied from 87-444 bp in length. The presence of the
intergenic spacer in all sexual species of Ambystomatidae suggests that it
arose at least 20 MYA and has been a stable component of the ambystomatid
mtDNA ever since. As such, it represents one of the few examples of a large
and persistent intergenic spacer in the mtDNA of any vertebrate clade.

Mitochondrial DNA evolution in the genus Equus 总被引:7,自引:0,他引:7
Employing mitochondrial DNA (mtDNA) restriction-endonuclease maps as the
basis of comparison, we have investigated the evolutionary affinities of
the seven species generally recognized as the genus Equus. Individual
species' cleavage maps contained an average of 60 cleavage sites for 16
enzymes, of which 29 were invariant for all species. Based on an average
divergence rate of 2%/Myr, the variation between species supports a
divergence of extant lineages from a common ancestor approximately 3.9 Myr
before the present. Comparisons of cleavage maps between Equus przewalskii
(Mongolian wild horse) and E. caballus (domestic horse) yielded estimates
of nucleotide sequence divergence ranging from 0.27% to 0.41%. This range
was due to intraspecific variation, which was noted only for E. caballus.
For pairwise comparisons within this family, estimates of sequence
divergence ranged from 0% (E. hemionus onager vs. E. h. kulan) to 7.8% (E.
przewalskii vs. E. h. onager). Trees constructed according to the parsimony
principle, on the basis of 31 phylogenetically informative restriction
sites, indicate that the three extant zebra species represent a
monophyletic group with E. grevyi and E. burchelli antiquorum diverging
most recently. The phylogenetic relationships of E. africanus and E.
hemionus remain enigmatic on the basis of the mtDNA analysis, although a
recent divergence is unsupported.

Growing evidence suggests that activation of mitogen-activated protein kinase (MAPK) signal transduction mediates changes in muscle gene expression in response to exercise. Nevertheless, little is known about upstream or downstream regulation of MAPK in response to muscle contraction. Here we show that ex vivo muscle contraction stimulates extracellular signal-regulated kinase 1 and 2 (ERK1/2), and p38(MAPK) phosphorylation. Phosphorylation of ERK1/2 or p38(MAPK) was unaffected by protein kinase C inhibition (GF109203X), suggesting that protein kinase C is not involved in mediating contraction-induced MAPK signaling. Contraction-stimulated phosphorylation of ERK1/2 and p38(MAPK) was completely inhibited by pretreatment with PD98059 (MAPK kinase inhibitor) and SB203580 (p38(MAPK) inhibitor), respectively. Muscle contraction also activated MAPK downstream targets p90 ribosomal S6 kinase (p90(Rsk)), MAPK-activated protein kinase 2 (MAPKAP-K2), and mitogen- and stress-activated protein kinase 1 (MSK1). Use of PD98059 or SB203580 revealed that stimulation of p90(Rsk) and MAPKAP-K2 most closely reflects ERK and p38(MAPK) stimulation, respectively. Stimulation of MSK1 in contracting skeletal muscle required the activation of both ERK and p38(MAPK). These data demonstrate that muscle contraction, separate from systemic influence, activates MAPK signaling. Protein evolution is governed by processes that alter primary sequence but also the length of proteins. Protein length may change in different ways, but insertions, deletions and duplications are the most common. An optimal protein size is a trade‐off between sequence extension, which may change protein stability or lead to acquisition of a new function, and shrinkage that decreases metabolic cost of protein synthesis. Despite the general tendency for length conservation across orthologous proteins, the propensity to accept insertions and deletions is heterogeneous along the sequence. For example, protein regions rich in repetitive peptide motifs are well known to extensively vary their length across species. Here, we analyze length conservation of coiled‐coils, domains formed by an ubiquitous, repetitive peptide motif present in all domains of life, that frequently plays a structural role in the cell. We observed that, despite the repetitive nature, the length of coiled‐coil domains is generally highly conserved throughout the tree of life, even when the remaining parts of the protein change, including globular domains. Length conservation is independent of primary amino acid sequence variation, and represents a conservation of domain physical size. Impact of Flooding on the Species Richness, Density and Composition of Amazonian Litter-Nesting Ants 总被引:1,自引:0,他引:1
Litter-nesting ants are diverse and abundant in tropical forests, but the factors structuring their communities are poorly known. Here we present results of the first study to examine the impact of natural variation in flooding on a highly diverse (21 genera, 77 species) litter-nesting ant community in a primary Amazonian forest. Fifty-six 3 × 3 m plots experiencing strong variation in flooding and twenty-eight 3 × 3 m terra firme plots were exhaustively searched for litter-nesting ants to determine patterns of density, species richness and species composition. In each plot, flooding, litter depth, twig availability, canopy cover, plant density, percent soil nitrogen, carbon, and phosphorus were measured. Degree of flooding, measured as flood frequency and flood interval, had the strongest impact on ant density in flooded forest. Flooding caused a linear decrease in ant abundance, potentially due to a reduction of suitable nesting sites. However, its influence on species richness varied: low-disturbance habitat had species richness equal to terra firme forest after adjusting for differences in density. The composition of ant genera and species varied among flood categories; some groups known to contain specialist predators were particularly intolerant to flooding. Hypoponera STD10 appeared to be well-adapted to highly flooded habitat. Although flooding did not appear to increase species richness or abundance at the habitat scale, low-flooding habitat contained a mixture of species found in the significantly distinct ant communities of terra firme and highly flooded habitat.

Although Alzheimer's disease pathologically affects the brain, familial Alzheimer's disease associated mutations of beta-amyloid precursor protein and presenilin are ubiquitously expressed and therefore aberrant intracellular signals, separate from but similar to, the brain may be expected. Here, we report selective down regulation of the serine/threonine kinase, Akt/PKB, concurrent with elevated endogenous GSK3beta kinase activity in familial Alzheimer's disease beta-amyloid precursor protein expressing human embryonic kidney (HEK) and familial Alzheimer's disease presenilin lymphoblast cells. Further, familial Alzheimer's disease presenilin in the human lymphoblast was associated with beta-catenin destabilization. Moreover, limited immunohistochemistry analysis reveals Akt/PKB in a subset of neurofibrillary tangles where GSK3beta and tau have been reported to co-localize, suggesting a possible Akt/GSK3beta and tau interaction in vivo. Our data suggest that familial Alzheimer's disease mutants of beta-amyloid precursor protein and presenilin signal, at least in part, through the Akt/GSKbeta pathway and that Akt/GSK3beta-mediated signalling may contribute to the underlying Alzheimer's disease pathogenesis induced by familial Alzheimer's disease mutants. 相似文献

The human nuclear gene for the catalytic subunit of mitochondrial DNA polymerase (POLG) contains within its coding region a CAG microsatellite encoding a polyglutamine repeat. Previous studies demonstrated an association between length variation at this repeat and male infertility, suggesting a mechanism whereby the prevalent (CAG)10 allele, which occurs at a frequency of >80% in different populations, could be maintained by selection. Sequence analysis of the POLG CAG microsatellite region of more than 1000 human chromosomes reveals that virtually all allelic variation at the locus is accounted for by length variation of the CAG repeat. Analysis of POLG from African great apes shows that a prevalent length allele is present in each species, although its exact length is species-specific. In common chimpanzee (Pan troglodytes) a number of different sequence variants contribute to the prevalent length allele, strongly supporting the idea that the length of the POLG microsatellite region, rather than its exact nucleotide or amino acid sequence, is what is maintained. Analysis of POLG in other primates indicates that the repeat has expanded from a shorter, glutamine-rich sequence, present in the common ancestor of Old and New World monkeys. 相似文献

Phylogenetic relationships within mammalian order Carnivora indicated by sequences of two nuclear DNA genes 总被引:8,自引:0,他引:8
Phylogenetic relationships among 37 living species of order Carnivora spanning a relatively broad range of divergence times and taxonomic levels were examined using nuclear sequence data from exon 1 of the IRBP gene (approximately 1.3 kb) and first intron of the TTR gene (approximately 1 kb). These data were used to analyze carnivoran phylogeny at the family and generic level as well as the interspecific relationships within recently derived Felidae. Phylogenetic results using a combined IRBP+TTR dataset strongly supported within the superfamily Califormia, the red panda as the closest lineage to procyonid-mustelid (i.e., Musteloidea) clade followed by pinnipeds (Otariidae and Phocidae), Ursidae (including the giant panda), and Canidae. Four feliform families, namely the monophyletic Herpestidae, Hyaenidae, and Felidae, as well as the paraphyletic Viverridae were consistently recovered convincingly. The utilities of these two gene segments for the phylogenetic analyses were extensively explored and both were found to be fairly informative for higher-group associations within the order Carnivora, but not for those of low level divergence at the species level. Therefore, there is a need to find additional genetic markers with more rapid mutation rates that would be diagnostic at deciphering relatively recent relationships within the Carnivora. 相似文献
