Two DNA fragments, AP-1 and AP-2, encoding amino acid sequences closely related to Ser/Thr protein phosphatases were amplified from Arabidopsis thaliana genomic DNA. Fragment AP-1 was used to screen. A. thaliana cDNA libraries and several positive clones were isolated. Clones EP8a and EP14a were sequenced and found to encode almost identical proteins (97% identity). Both proteins are 306 amino acids in length and are very similar (79–80% identity) to the mammalian isotypes of the catalytic subunit of protein phosphatase 2A. Therefore, they have been designated PP2A-1 and PP2A-2. A third cDNA clone, EP7, was isolated and sequenced. The polypeptide encoded (308 amino acids, lacking the initial Met codon) is 80% identical with human phosphatases 2A and was named PP2A-3. The PP2A-3 protein is extremely similar (95% identity) to the predicted protein from a cDNA clone previously found in Brassica napus. Southern blot analysis of genomic DNA using AP-1 and AP-2 probes, as well as probes derived from clones EP7, EP8a and EP14a strongly indicates that at least 6 genes closely related to type 2A phosphatases are present in the genome of A. thaliana. Northern blot analysis using the same set of probes demonstrates that, at the seedling stage, the mRNA levels for PP2A-1, PP2A-3 and the gene containing the AP-1 sequence are much higher than those of PP2A-2 and AP-2. These results demonstrate that a multiplicity of type 2A phosphatases might be differentially expressed in higher plants. 相似文献

The growth of Staph. aureus was inhibited by dehydrated garlic at levels of 1.5% (w/v) and over. Enterotoxins A, B and C1 were only detectable in broth containing < 1% of garlic while enterotoxin D was produced at a level of 2%. Summary The effect of phenylacetic acid (PAA) and several analogs on the activity of isopenicillin N synthase (IPNS) and acyl-CoA: 6-APA acyltransferase (AT) fromPenicillium chrysogenum Wis 54-1255 has been tested. Whereas the substitution on the ring of a hydrogen atom by hydroxy-, methyl- or methoxy- groups did not cause any effect, the presence of halogens (Cl or Br) at positions 3 and/or 4 of PAA strongly inhibited these two enzymes. An Escherichia coli strain bearing the dnaQ49 mutation, which results in a defective s subunit of DNA polymerase III, and carrying the lexA71 mutation, which causes derepression of the SOS regulon, is totally unable to maintain high-copy-number plasmids containing the umuDC operon. The strain is also unable to maintain the pAN4 plasmid containing a partial deletion of the umuD gene but retaining the wild-type umuC gene. These results suggest that a high cellular level of UmuC is exceptionally harmful to the defective DNA polymerase III of the dnaQ49 mutant. We have used this finding as a basis for selection of new plasmid umuC mutants. The properties of two such mutants, bearing the umuC61 or umuC95 mutation, are described in detail. In the umuC122:: Tn 5 strain harbouring the mutant plasmids, UV-induced mutagenesis is severely decreased compared to that observed with the parental umuDC + plasmid. Interestingly, while the frequency of UV-induced GC → AT transitions is greatly reduced, the frequency of AT → TA transversions is not affected. Both mutant plasmids bear frameshift mutations within the same run of seven A residues present in umuC +; in umuC61 the run is shortened to six A whereas in umuC95 is lengthened to eight A. We have found in both umuC61 and umuC95 that translation is partially restored to the proper reading frame. We propose that under conditions of limiting amounts of UmuC, the protein preferentially facilitates processing of only some kinds of UV-induced lesions. 相似文献

Bulkiness, longevity and solidity of the body in woody plants enable the successive development of accessory shoots and adventitious roots in (1) both proximal and distal positions on organs, (2) both the above-ground and below-ground space, (3) both the aerial and soil environments. In monocotyledons, woody rhizomes play an essential role in the basic growth habit and architectural models. In dicotyledonous and a few gymnospermous trees, attached and successively disconnected ramets play a multilateral role in the pertinent life strategies. The majority of sprouts, coppice shoots and root suckers behave as opportunistic organs (a) serving as means of vegetative reproduction, (b) securing colonization of unoccupied ground, (c) increasing competitive power of the species within the community, (d) increasing survival rate of the stressed/disturbed genet in marginal habitats, (e) forming replacement for ageing or damaged organs, and (f) enabling reiteration of the genet's entire architectural model. 相似文献

La Salada de Chiprana Lake, located in the Ebro River basin, northeastern Spain, is the only permanent and deep water hypersaline ecosystem in all of western Europe. With a total surface of 31 ha and a maximum depth of 5.6 m, it has several basins bounded by elongated sandstone-bodies or ribbons which are paleochannels of Miocene age. Its salinity varied from 30 to 73 g 1–1 during the 1989 hydrological cycle and the most abundant ions were magnesium and sulphate. Depth-time distributions of major physico-chemical variables demonstrated that the lake was stratified in two distinctive layers during most of the year. The chemocline disappeared only in October, with the complete overturn of the water column. In the deep water, three conditions occurred which allowed development of green sulphur bacteria populations: (1) oxygen depletion, (2) presence of hydrogen sulphide and (3) presence of light.
