Previous calculations using continuum electrostatic calculations showed that a fully hydrated monovalent cation is electrostatically stabilized at the center of the cavity of the KcsA potassium channel. Further analysis demonstrated that this cavity stabilization was controlled by a balance between the unfavorable reaction field due to the finite size of the cavity and the favorable electrostatic field arising from the pore helices. In the present study, continuum electrostatic calculations are used to investigate how the stability of an ion in the intracellular vestibular cavity common to known potassium channels is affected as the inner channel gate opens and the cavity becomes larger and contiguous with the intracellular solution. The X-ray structure of the calcium-activated potassium channel MthK, which was crystallized in the open state, is used to construct models of the KcsA channel in the open state. It is found that, as the channel opens, the barrier at the helix bundle crossing decreases to approximately 0 kcal/mol, but that the ion in the cavity is also significantly destabilized. The results are compared and contrasted with additional calculations performed on the KvAP (voltage-activated) and KirBac1.1 (inward rectifier) channels, as well as models of the pore domain of Shaker in the open and closed state. In conclusion, electrostatic factors give rise to energetic constraints on ion permeation that have important functional consequences on the various K+ channels, and partly explain the presence or absence of charged residues near the inner vestibular entry.  相似文献

Background

Since loss of height may indicate vertebral fracture, the accuracy of the information on height is relevant for clinical practice. We undertook this study to compare reported and measured loss of height among post-menopausal women in a primary care setting. We also analyzed the determinants of this height loss.

Methods

In an observational study conducted between December 2007 and May 2008, we asked 1779 randomly selected general practitioners to recruit the first five female patients who were more than 60 years of age, regardless of the reason for the consultation. Using a questionnaire, physicians collected data on demographic and clinical variables, history of osteoporosis and current anti-osteoporotic treatment. We used three assessments of height: tallest height in early adulthood recalled by the patient, estimated current height reported by the patient at the visit and current measured height. We defined loss of height as the difference between the patient’s tallest recalled height and her current measured height.

Results

A total of 8610 patients were included in the analysis; the mean age was 70.9 (standard deviation [SD] 7.2) years. The mean loss of height was 4.5 cm. The mean current reported height was 2.1 (SD 2.5) cm lower than the tallest recalled height and 2.4 (SD 2.6) cm lower than the measured current height. The best predictors of a loss of height of 3 cm or more were age (odds ratio [OR] 1.09, 95% confidence interval [CI] 1.08–1.10), previous vertebral fracture (OR 1.49, 95% CI 1.16–1.91), previous nonvertebral fracture (OR 1.26, 95% CI 1.06–1.51), thoracic kyphosis (OR 2.07, 95% CI 1.69–2.55), scoliosis (OR 1.35, 95% CI 1.12–1.63), back pain (OR 1.22, 95% CI 1.07–1.39) and osteoporosis (OR 1.39, 95% CI 1.20–1.60).

Interpretation

Our study showed that the patients’ estimated current height was not correct, with a mean difference of −2.5 cm from the current measured height. The mean height loss was 4.5 cm. Previous vertebral fracture and thoracic kyphosis were strong determinants of the height loss.Loss of height is common with advancing age.1,2 Causes include changes in curvature of the spine, narrowing of intervertebral discs and vertebral fractures. Height loss is associated with back pain and thoracic hyperkyphosis.3,4 Two-thirds of adults have back pain at any time. Controversies exist about the need for radiographs of the spine: Does the benefit of detecting treatable disorders of the spine such as vertebral fracture outweigh the harm of unnecessary radiographs? Loss of height is usually recorded as one of the clinical signs to help identify postmenopausal women with vertebral fractures.5 The use of this parameter to decide whether radiography is needed depends on the threshold for height loss associated with a strong risk of vertebral fracture. The thresholds useful in clinical practice to detect prevalent vertebral fracture range from 3 cm to 6 cm,69 with the risk of prevalent fracture increasing with the magnitude of the height loss. Thus, the accuracy of the information on height is relevant for clinical practice.We conducted this study to compare reported and measured loss of height in a large population of women more than 60 years old in a primary care setting and to analyze the determinants of this height loss.  相似文献

Extensification modify the C and N cycles in grassland ecosystems, but it is not clear whether reduced exploitation increases or decreases soil nitrogen availability, and whether these changes result from a direct effect of the treatment or from an indirect treatment effect through a change in plant species composition. A formerly intensively exploited grassland was submitted to the following treatments: (i) control with one mowing and four grazing periods per year (4G+M), (ii) cessation of mowing (4G), (iii) cessation of mowing and suppression of three grazing periods (1G). During the 13th year of the experiment, the species composition and key indicators of the C and N cycles were measured, using 20 samples per treatment. Lolium perenne and Trifolium repens disappeared in favor of tall caespitose grasses in 4G, and of rhizomatous species in 1G. The species composition and the nitrate concentration of the soil solution suggested an increase in nutrient availability under reduced exploitation, whereas the nitrification and denitrification potentials decreased. More particulate organic matter accumulated in proportion to the below-ground phytomass, whereas the C:N ratio remained constant. Testing treatment effect at similar species composition and plant community effect within the same treatment showed that: (1) the increase in POM residence time was mainly due to the changes in species composition, (2) the decrease in nitrification activity resulted mainly from a direct effect of the treatment, and (3) a compensation between a direct positive and an indirect negative effect of the treatment resulted in no change in extractable N. All results suggested that soil N availability was not decreased, although litter degradability decreased.

Zusammenfassung

Ein Unterlassen der Heuernte und eine Reduktion der Beweidungsintensität ändern die C- und N-Kreisläufe in Grünland-Ökosystemen. Bislang ist unklar, ob eine extensive Nutzung die Nährstoffverfügbarkeit erhöht oder senkt, und ob diese Änderung der Nährstoffverfügbarkeit in direkter Verbindung mit der verringerten Nutzung steht, oder indirekt ein Resultat der sich ändernden botanischen Zusammensetzung ist. In der vorliegenden Untersuchung wurden die botanische Zusammensetzung sowie die C- und N-Kreisläufe im Boden eines naturnahen, ehemals intensiv beweideten Grünlands nach 12 Jahren unterschiedlicher Nutzungsintensitäten – eine Heuernte und vier Beweidungen jährlich (4G+M), viermalige (4G) und einmalige (1G) jährliche Beweidung – verglichen. Pro Weide wurden je 20 Punkte beprobt. Bei verringerter Nutzung waren die Nitrifikation/Denitrifikation geringer als bei intensiver Nutzung. Die botanische Zusammensetzung sowie die Nitrat-Konzentration im Bodenwasser wurden mehr eutroph. So wurden Lolium perenne und Trifolium repens durch horstbildende Grässer (4G) bzw. rhizombildende Arten (1G) abgelöst. Bei extensiver Nutzung war das Verhältnis von organischer Bodenmasse (particle organic matter, POM) zu unterirdischen pflanzlicher Biomasse höher, hatte jedoch keinen Einfluss auf das C/N-Verhältnis. Ein Vergleich von Weiden gleicher botanischer Zusammensetzung (Nutzungs-Effekt) und von Weiden gleicher Nutzung aber unterschiedlicher botanischer Zusetzung (plant-community-Effekt) zeigte: (i) eine Zunahme der POM-Verweildauer in Folge einer veränderten botanischen Zusammensetzung, (ii) eine Abnahme der Nitrifikation in Folge der verringerten Nutzung und (iii) Kompensation der verringerten Nutzung durch eine Änderung der botanischen Zusammensetzung, womit der leicht verfügbare N unverändert blieb. Eine abnehmende Streu-Qualität scheint keinen Einfluss auf das N-Nachlieferungsvermögen zu haben. Eine hohe N-Verfügbarkeit in Verbindung mit geringer Streu Zersetzung scheinen verantwortlich für eine funktionelle Änderung des Ökosystems und seiner Biodiversität.  相似文献

A series of 6-aryl-2,4-dioxo-5-hexenoic acids, were synthesized and tested against HIV-1 in cell-based assays and against recombinant HIV-1 integrase (rIN) in enzyme assays. Compound 8a showed potent antiretroviral activity (EC(50)=1.5 microM) and significant inhibition against rIN (strand transfer: IC(50)=7.9 microM; 3'-processing: IC(50)=7.0 microM). A preliminary molecular modeling study was carried out to compare the spatial conformation of 8a with those of L-731988 (4) and 5CITEP (7) in the IN core.  相似文献

We have used pulsed field gel electrophoresis for further physical mapping studies in the q27 region of the human X chromosome. We show that the DXS 102 locus and the F9 gene are separated by only 300 kb despite a genetic distance of 1.4 cM; this linkage orients our large-scale map and shows that the mcf.2 transforming sequence is telomeric to F9. A BssHII complete-digest jumping library was used to jump toward the DXS 105 locus; a 130-kb jump was achieved and the corresponding "linking clone" was obtained.  相似文献

Limited proteolysis experiments were performed to study conformation changes induced by ligand binding on in vitro produced wild-type and I747T mutant glucocorticoid receptors. Dexamethasone-induced conformational changes were characterized by two resistant proteolysis fragments of 30 and 27 kDa. Although dexamethasone binding affinity was only slightly altered by the I747T substitution (Roux, S., Térouanne, B., Balaguer, P., Loffreda-Jausons, N., Pons, M., Chambon, P., Gronemeyer, H., and Nicolas, J.-C. (1996) Mol. Endocrinol. 10, 1214-1226), higher dexamethasone concentrations were required to obtain the same proteolysis pattern. This difference was less marked when proteolysis experiments were conducted at 0 degrees C, indicating that a step of the conformational change after ligand binding was affected by the mutation. In contrast, RU486 binding to the wild-type receptor induced a different conformational change that was not affected by the mutation. Analysis of proteolysis fragments obtained in the presence of dexamethasone or RU486 indicated that the RU486-induced conformational change affected the C-terminal part of the ligand binding domain differently. These data suggest that the ligand-induced conformational change occurs via a multistep process. In the first step, characterized by compaction of the ligand binding domain, the mutation has no effect. The second step, which stabilizes the activated conformation and does not occur at 4 degrees C, seems to be a key element in the activation process that can be altered by the mutation. This step could involve modification of the helix H12 position, explaining why the conformation induced by RU486 is not affected by the mutation.  相似文献

An unprecedented epidemic of chikungunya virus (CHIKV) infection recently started in countries of the Indian Ocean area, causing an acute and painful syndrome with strong fever, asthenia, skin rash, polyarthritis, and lethal cases of encephalitis. The basis for chikungunya disease and the tropism of CHIKV remain unknown. Here, we describe the replication characteristics of recent clinical CHIKV strains. Human epithelial and endothelial cells, primary fibroblasts and, to a lesser extent, monocyte-derived macrophages, were susceptible to infection and allowed viral production. In contrast, CHIKV did not replicate in lymphoid and monocytoid cell lines, primary lymphocytes and monocytes, or monocyte-derived dendritic cells. CHIKV replication was cytopathic and associated with an induction of apoptosis in infected cells. Chloroquine, bafilomycin-A1, and short hairpin RNAs against dynamin-2 inhibited viral production, indicating that viral entry occurs through pH-dependent endocytosis. CHIKV was highly sensitive to the antiviral activity of type I and II interferons. These results provide a general insight into the interaction between CHIKV and its mammalian host.  相似文献

The endemic New Zealand ground wētā (Hemiandrus sp. ‘promontorius’) has a Naturally Uncommon conservation status. This is because of the paucity of information on its density and distribution. Here, the biology, density and distribution of a population of this wētā found in and around vineyards in the Awatere Valley, Marlborough was studied. Wētā density was assessed in vineyards, paddocks and shrublands in this valley. Soil moisture, penetration resistance, pH and organic matter were recorded at locations with and without wētā. Wētā density in vineyards was significantly higher than in either paddocks or shrub habitats. In vineyards, the density of this insect was significantly higher under-vines than in the inter-rows. Higher numbers of this wētā were found in moist soils that required lower force to burrow. Females laid an average of 55 eggs between March and April, which hatched in September. These findings highlight the intersection between agriculture and conservation.  相似文献
