E-Cadherin-mediated formation of adherens junctions (AJs) is essential for the morphogenesis of epithelial cells. However, the mechanisms underlying E-cadherin clustering and AJ maturation are not fully understood. Here we report that type Iγ phosphatidylinositol-4-phosphate 5-kinase (PIPKIγ) associates with the exocyst via a direct interaction with Exo70, the exocyst subunit that guides the polarized targeting of exocyst to the plasma membrane. By means of this interaction, PIPKIγ mediates the association between E-cadherin and Exo70 and determines the targeting of Exo70 to AJs. Further investigation revealed that Exo70 is necessary for clustering of E-cadherin on the plasma membrane and extension of nascent E-cadherin adhesions, which are critical for the maturation of cohesive AJs. In addition, we observed phosphatidylinositol-4,5-bisphosphate (PI4,5P(2)) accumulation at E-cadherin clusters during the assembly of E-cadherin adhesions. PIPKIγ-generated PI4,5P(2) is required for recruiting Exo70 to newly formed E-cadherin junctions and facilitates the assembly and maturation of AJs. These results support a model in which PIPKIγ and PIPKIγ-generated PI4,5P(2) pools at nascent E-cadherin contacts cue Exo70 targeting and orient the tethering of exocyst-associated E-cadherin. This could be an important mechanism that regulates E-cadherin clustering and AJ maturation, which is essential for the establishment of solid, polarized epithelial structures.  相似文献

China is rich of germplasm resources of common wild rice (Oryza rufipogon Griff.) and Asian cultivated rice (O. sativa L.) which consists of two subspecies, indica and japonica. Previous studies have shown that China is one of the domestication centers of O. sativa. However, the geographic origin and the domestication times of O. sativa in China are still under debate. To settle these disputes, six chloroplast loci and four mitochondrial loci were selected to examine the relationships between 50 accessions of Asian cultivated rice and 119 accessions of common wild rice from China based on DNA sequence analysis in the present study. The results indicated that Southern China is the genetic diversity center of O. rufipogon and it might be the primary domestication region of O. sativa. Molecular dating suggested that the two subspecies had diverged 0.1 million years ago, much earlier than the beginning of rice domestication. Genetic differentiations and phylogeography analyses indicated that indica was domesticated from tropical O. rufipogon while japonica was domesticated from O. rufipogon which located in higher latitude. These results provided molecular evidences for the hypotheses of (i) Southern China is the origin center of O. sativa in China and (ii) the two subspecies of O. sativa were domesticated multiple times.  相似文献

利用内蒙古锡林浩特国家气候观象台1994~2009年牧草生长季逐月实测资料,对CENTURY模型进行检验,模拟内蒙古典型草原1953~2010年间地上净初级生产力(ANPP)动态,并与26个气象因子进行相关性分析。模型检验结果显示,模拟值与观测值之间的相关系数为R2=0.66,斜率b=0.95,误差平方根值为50.51 g·m-2,平均绝对百分比误差为44.19%。结果表明:(1)CENTURY模型能比较准确地模拟这类草原的季节动态和年际变化;在过去的58年中,内蒙古典型草原温度增加,降水减少,ANPP下降;ANPP变化趋势与降水量相似。(2)用实际气象观测资料模拟获得的ANPP随气温和降水的变化呈现出明显的变化规律,生长季内地上生物量对降水和温度的季节性分布也非常敏感;相关分析进一步表明,ANPP对生长季内降水量和极端高温非常敏感,而与年极端最低气温、平均地面温度、日照时数、平均风速和最大积雪深度无显著相关关系;过去58年研究区ANPP下降是降水减少、温度升高以及干旱事件频发共同作用的结果。(3)根据预测,在SRES B2情景下,未来50~100年内蒙古典型草原生长季平均最高气温和最低气温都将呈升高趋势,2080s分别升高4.01℃、4.35℃,每10年增加速率分别为0.35℃和0.38℃;降水量略呈增加,2020s、2050s和2080s研究区生长季将分别增加3.17%、5.13%和7.03%,每10年增加速率为0.09 mm;ANPP呈下降趋势年际间波动较大,2020s、2050s和2080s研究区将分别下降5.76%、7.52%和11.42%,每10年下降速率为0.76 g·m-2。  相似文献

A single Rad52-related protein is evident by blast analysis of the Ustilago maydis genome database. Mutants created by disruption of the structural gene exhibited few discernible defects in resistance to UV, ionizing radiation, chemical alkylating or cross-linking agents. No deficiency was noted in spontaneous mutator activity, allelic recombination or meiosis. GFP-Rad51 foci were formed in rad52 cells following DNA damage, but were initially less intense than normal suggesting a possible role for Rad52 in formation of the Rad51 nucleoprotein filament. A search for interacting genes that confer a synthetic fitness phenotype with rad52 after DNA damage by UV irradiation identified the genes for Mph1, Ercc1 and the Rad51 paralogue Rec2. Testing known mutants in recombinational repair revealed an additional interaction with the BRCA2 orthologue Brh2. Suppression of the rec2 mutant's UV sensitivity by overexpressing Brh2 was found to be dependent on Rad52. The results suggest that Rad52 serves in an overlapping, compensatory role with both Rec2 and Brh2 to promote and maintain formation of the Rad51 nucleoprotein filament.  相似文献

Human fibrinogen-related protein-1/liver fibrinogen-related protein-1 (HFREP-1/LFIRE-1), a liver-specificprotein, is a member of fibrinogen superfamily that exerts various biological activities. However, the func-tion of HFREP-1/LFIRE-1 in liver remains unknown. Here we isolated its mouse ortholog gene-mousefibrinogen-related protein-1 (mfrep-1), which encoded 314 amino acids, exhibiting 80.4% similarity toHFREP-1/LFIRE-1. Northern blot analysis revealed that 1.2-kb mfrep-1 mRNA was detected selectivelyin mouse liver. To explore the function of MFREP-1, we examined the levels of mfrep-1 mRNA duringregeneration after 70% partial hepatectomy (PHx) in mice. mfrep-1 mRNA increased in the regeneratingliver and reached the first shoulder peak at 2-4 h after PHx. Cycloheximide pretreatment could suppress theinduction of mfrep-1, indicating the up-regulation of this gene need de novo protein synthesis. Its mRNAcontinued to elevate at 6 h thereafter and reached the second peak at 24 h. The enhanced expression ofmfrep-1 maintained high until 72 h and then declined slowly to the basal level. Immunohistochemistryassessment confirmed the up-regulated expression of MFREP-1 protein in parenchymal cells during liverregeneration. These data suggested that MFREP-1 might play an important role in liver regeneration andbe involved in the regulation of cell growth.  相似文献

从3种不同土壤中分离和纯化得到10个石油降解细菌菌株,分离菌株均为好氧菌、革兰氏阴性菌、不形成芽孢的杆菌,10个菌株均能利用中等链长的烷烃、柴油和原油作为碳源,而不能以单环芳烃和多环芳烃为碳源。根据其生理生化性状和16SrDNA序列分析结果表明,分离菌株EVA5,EVA6,EVA7,EVA8和EVA9为假单胞菌(Pseudomonas spp.),EVA10、EVA11、EVA12、EVA13和EVA14为不动杆菌(Acinetobacter spp.),均属于Proteobac-teria的γ亚群。  相似文献

研究了以克氏螯虾壳为原料制备氨基葡萄糖盐酸盐 (GAH)的工艺条件 :酸水解液为 10mol·L-1HCl,85~ 92℃水解 5~ 6h ;甲壳素∶酸水解液为 3∶5。GAH产率为甲壳素的 5 2 %~ 5 5 %。GAH纯度 :99%~ 10 0 .5 %。  相似文献
