Historical relationships were inferred for the oriental lizards of the genus Takydromus Daudin 1802 (Lacertidae) on the basis of DNA sequences. Of the 17 species currently recognized for the genus, 13 species represented by 42 specimens from 29 localities were examined. Maximum-likelihood and maximum-parsimony analyses of data for 829 aligned sites from parts of the mitochondrial 12S and 16S rRNA genes yielded relationships that, while showing no substantial discrepancy with each other, were strikingly different from a currently prevailing phylogenetic hypothesis from a parsimony analysis of morphological characters. Based on the results of these molecular analyses, supplemented by results of the morphological analysis that offered robust evidence for positions of two additional species ( T. khasiensis and T. sylvaticus ), the following interrelationships were hypothesized as the most preferred phylogeny: ( kuehnei ( sexlineatus khasiensis ))( tachydromoides (( smaragdinus ( sauteri ( dorsalis sylvaticus ))) ( amurensis ((( formosanus wolteri ) hsuehshanensis )( toyamai ( septentrionalis stejnegeri )))))). These interrelationships indicate: (1) invalidity of Platyplacopus Boulenger 1917 , which was recently resurrected as a subgenus of Takydromus on the basis of the morphological analysis; (2) homoplasy in states of some morphological characters, such as green dorsal coloration, that were considered as synapomorphs of certain nodes in the morphological analysis; and (3) involvement of the genus in a series of vicariances in both the continental and insular parts of eastern Eurasia. A new cell-extraction medium was developed, under conditionssuch that physical injury was minimized, for the simultaneousisolation of plant nuclei, mitochondria, and chloroplasts inmorphological forms closely resembling those in living cells.Comparisons were made of organelles isolated in buffered NaCl,sucrose, and specially developed formulations with the synthetichigh-molecular-weight polymers Ficoll (polymerized form of sucrose,MW 400,000) and Dextran 40 (MW 26,000). The finally developedmedium contained: sucrose (osmoticum); Tris-acetate (buffer);Ficoll, Dextran 40, and bovine serum albumin (structure preservatives);glutathione, cysteine, ascorbic acid, or mercaptoethanol (reducingagents); and magnesium acetate and manganese acetate (activitypreservatives). The properties of the SFDP medium are such thatnuclei, chloroplasts, and mitochondria resemble their formsin living cells; mitochondria exhibit dynamic pleomorphic changesin form similar to those occurring in living cells and haircells immersed in the medium can be cut open while cytoplasmicstreaming continues during and after the incision. 相似文献

The complex structure of the crown of Cornus kousa, generallyfive-forked in vegetative branching and two-forked in reproductivebranching, is analysed quantitatively and described by two basicbranching principles: decussate phyllotaxy and the resettingrule for planes of branching. Most Cornus species have opposite,decussate phyllotaxis. The leaf pair (with axillary buds) definesthe branching plane of a node. Because of regular phyllotaxis,the fundamental branching pattern is that every branching planealong an axis is perpendicular to the preceding one. However,the first node of a lateral horizontal shoot always has a horizontalbranching plane; we term this the resetting rule. We observedthat resetting occurs when the first nodes initiated in thevertical plane are repositioned by a twisting of their firstinternodes. All later nodes alternate directions, i.e. showusual decussate alternation. Foliage leaf nodes usually producethree-forked branchings. When vegetative winter buds are formed,a foliar node and adjacent scale leaf node produce a five-forkedbranching. When reproductive winter buds with a terminal inflorescenceare formed, the last foliar node and two adjacent scale leafnodes can produce a variety of branchings but usually producean equal two-forked branching. To understand better the architecturein C. kousa, we contrast it with C. capitata which does notproduce buds with scale leaves and whose vegetative nodes areclearly separated. The rhythm of adult eclosion in the Indian meal moth Plodia interpunctella Hübner (Lepidoptera: Pyralidae) is investigated under various photoperiods and temperatures aiming to determine the nature of the temperature compensation and the free‐running period. Insects that are committed to a nondiapause larval development show diel rhythms of adult eclosion at 30, 25 and 20 °C. At 30 °C, the eclosion peak (i.e. the mean time of eclosion) occurs approximately 20 h after lights off under an LD 4 : 20 h photocycle, and at approximately 15 h under an LD 20 : 4 h photocycle. At 25 °C, the peak of eclosion occurs approximately 19 h after lights off under an LD 2 : 20 h photocycle and at approximately 16 h under an LD 20 : 4 h photocycle. At 20 °C, the eclosion peak is significantly advanced under long days of >12 h (i.e. approximately 20 h after lights off under an LD 4 : 20 h photocycle and approximately 9 h under an LD 20 : 4 h photocycle), indicating an effect of both lights‐off and lights‐on signals on the timing of the adult eclosion. To determine the involvement of a self‐sustained oscillator, the rhythm of adult eclosion is examined under darkness at different temperatures (30 to 21 °C). The mean free‐running periods are 22.4, 22.8, 22.0 and 22.5 h at 30, 24, 23 and 22 °C, respectively, indicating that the eclosion rhythm is temperature‐compensated. However, this rhythm does not free‐run under constant darkness at 21 °C. Because a clear diel rhythm is observed under 24‐h photocycles at 20 °C, the oscillator might be damped out within 24 h at the lower temperature. 相似文献
