The quolls are the largest native predators remaining on mainland Australia. We describe the cross‐species testing of all available microsatellite loci for representative across the entire distribution of quolls, with the additional characterization of five new microsatellite loci from the Chuditch. All primers produced clear and polymorphic amplification patterns containing between nine and 17 alleles and with high levels of diagnostic variability. These highly polymorphic primers make them useful additional tools in planning conservation strategies across related conspecifics, many of which are under threat.  相似文献

Abstract Orb web spiders face a dilemma: forage in open habitats and risk predation or forage in closed habitats to minimize risk but at reduced foraging profitability. We tested whether Argiope keyserlingi opts for safer habitats at the expense of foraging success by (i) determining habitat selection indices in open and closed habitats; (ii) marking and releasing individual juvenile, subadult and adults over two 4‐week periods to determine if life‐history stage influences habitat selection; and (iii) determining the biotic and abiotic environmental parameters that relate to A. keyserlingi abundance. We found that A. keyserlingi selected closed habitats. Sedge and anthropogenic structures were selected and trees were avoided. Juveniles were never found in open habitats, most likely because of high postdispersal mortality. Subadults and adults may shift from closed to open habitats while juveniles never shifted habitat. Foliage density, plant height, potential prey abundance, and mantid and bird abundance were correlated with A. keyserlingi abundance, with only bird abundance explaining habitat selection. We measured web capture area, spiral distance (distance between spiral threads) and the number of decoration arms (0, 1, 2, 3 or 4) in the field and did laboratory experiments to test the influence of (i) space and vegetation; (ii) prey abundance; and (iii) web damage, on web architecture. Argiope keyserlingi webs exhibited geometric plasticity by having larger prey capture areas and spiral distances in open habitats. Decoration design did not differ between habitats however. Variation in space availability, air temperature, prey abundance and web damage explained the variations in web architecture. Potential prey size and diversity differed between habitats but prey abundance did not. As large prey may be important for spider survivorship, foraging success appears to be compromised by occupying closed habitats.  相似文献

Abstract Resource selection is a multi-staged process of behavioral responses to various resource cues or stimuli. Previous research suggests some aspects of resource selection may be inherent (i.e., genetic predisposition) or based on early experience and that individuals respond to certain resource cues but not to others. In other words, resource selection may be based on a template that specifies which cues to use in the resource-selection process and the appropriate response to those cues. We used resource utilization functions (RUFs) to examine the resource-selection template of translocated three-toed box turtles (Terrapene carolina triunguis; hereafter turtles) and made comparisons to resident turtles. Translocated turtles, previously residents of a predominantly forested landscape with low edge-density, used forest openings, forest edges, and southwest-facing slopes before and after translocation to a fragmented site containing resident turtles. In contrast, resident turtles used forested areas and northeast-facing slopes within a predominantly open landscape with high edge-density. Our comparison of resource selection by translocated and resident turtles revealed population-specific resource selection and consistency in selection following translocation, which reinforces the idea of a resource-selection template and suggests that in the short-term box turtles may not adapt their predisposed behavior to local conditions. Thus, translocated animals may evaluate and respond to resource cues as if they were at the original site. Lack of site fidelity may result from individuals seeking additional resources to match their resource-selection template. Successful translocation of turtles may require an assessment of resource selection prior to translocation and development of management strategies that mitigate turtle response to translocation.  相似文献

Abstract

Neurosurgical image-guidance has historically relied on the registration of the patient and preoperative imaging series with surgical instruments in the operating room (OR) coordinate space. Recent studies measuring intraoperative tissue motion have suggested that deformation-induced misregistration from surgical loading is a serious concern with such systems. In an effort to improve registration fidelity during surgery, we are pursuing an approach which uses a predictive computational model in conjunction with data available in the OR to update the high resolution preoperative image series. In previous work. we have developed an in vivo experimental system in the porcine brain which has been used to investigate a homogeneous finite element rendering of consolidation theory as a tissue deformation model. In this paper, our computational approach has been extended to include heterogeneous tissue property distributions determined from an image-to-grid segmentation scheme. Results produced under two different loading conditions show that heterogeneity in the stiffness properties and interstitial pressure gradients varied over a range of physiologically reasonable values account for 1-3% and 5-8% of the predicted tissue motion, respectively, while homogeneous linear elasticity is responsible for 60-70% of the surgically-induced motion that has been recoverable with our model-based @approach.  相似文献

Founder effects during colonization of a novel environment are expected to change the genetic composition of populations, leading to differentiation between the colonizer population and its source population. Another expected outcome is differentiation among populations derived from repeated independent colonizations starting from the same source. We have previously detected significant founder effects affecting rate of laboratory adaptation among Drosophila subobscura laboratory populations derived from the wild. We also showed that during the first generations in the laboratory, considerable genetic differentiation occurs between foundations. The present study deepens that analysis, taking into account the natural sampling hierarchy of six foundations, derived from different locations, different years and from two samples in one of the years. We show that striking stochastic effects occur in the first two generations of laboratory culture, effects that produce immediate differentiation between foundations, independent of the source of origin and despite similarity among all founders. This divergence is probably due to powerful genetic sampling effects during the first few generations of culture in the novel laboratory environment, as a result of a significant drop in N e. Changes in demography as well as high variance in reproductive success in the novel environment may contribute to the low values of N e. This study shows that estimates of genetic differentiation between natural populations may be accurate when based on the initial samples collected in the wild, though considerable genetic differentiation may occur in the very first generations of evolution in a new, confined environment. Rapid and significant evolutionary changes can thus occur during the early generations of a founding event, both in the wild and under domestication, effects of interest for both scientific and conservation purposes.  相似文献

Abstract Wildlife biologists use knowledge about wildlife-habitat relationships to create habitat models to predict species occurrence across a landscape. Researchers attribute limitations in predictive ability of a habitat model to data deficiencies, missing parameters, error introduced by specifications of the statistical model, and natural variation. Few wildlife biologists, however, have incorporated intra- and interspecific interactions (e.g., conspecific attraction, competition, predator-prey relationships) to increase predictive accuracy of habitat models. Based on our literature review and preliminary data analysis, conspecific attraction can be a primary factor influencing habitat selection in wildlife. Conspecific attraction can lead to clustered distributions of wildlife within available habitat, reducing the predictive ability of habitat models based on vegetative and geographic parameters alone. We suggest wildlife biologists consider incorporating a parameter in habitat models for the clustered distribution of individuals within available habitat and investigate the mechanisms leading to clustered distributions of species, especially conspecific attraction.  相似文献

Summary

Long-term maintenance of lobster, Homarus americanus and crayfish, Pacifasticus leniusculus primary cell cultures of testicular and hematopoietic tissues, for 11 and 3 months, respectively, succeeded in a modified Medium 199 supplemented with 10% fetal bovine serum (pH 7.5, 200°C). In addition, NaC1 was used to adjust the lobster culture medium to 1000 mOsm and the crayfish medium to 400 mOsm. Proline concentration was also elevated. Testes were dissociated with 200 U/ml type II collagenase 2–3 days prior to culture.

Lobster hemocytes reacted to 10?7 M 20-hydroxyecdysone (20-HE) by reducing contact inhibition and increasing invasive behavior one week after hormonal exposure. The presence of 10?7 M 20-HE caused mesodermal cell death and spermatogonial proliferation in lobster testicular cell cultures within one week. Crayfish testicular mesodermal cells formed vacuoles 5 days after exposure to 10?8 M 20-HE. These results are discussed in relation to the cellular events that occur in vivo during premolt.  相似文献

1. Early‐season flooding and ice entrapment at sub‐zero temperatures pose significant challenges to any polar terrestrial invertebrate. 2. The chironomid midge, Eretmoptera murphyi, is native to the sub‐Antarctic island of South Georgia and has been introduced to the maritime Antarctic (Signy Island). While the majority of its 2‐year life cycle is spent as a terrestrial larva, it is found in habitats potentially exposed to prolonged flooding. 3. The current study explored the tolerance of the larvae to extended submergence, demonstrating survival for at least 28 days, underlain by their ability to respire (oxy‐regulate) whilst submerged. To date, this ability is not known to be shared by any other terrestrial midge. Larvae also demonstrated notable anoxia tolerance whilst encased in ice, surviving for up to 28 days. 4. These data indicate a capacity to survive ecologically relevant periods of submergence and/or ice entrapment, such as may be experienced in their natural habitats.  相似文献
