Chemosensory similarities among oils: does viscosity play a role?   总被引:1,自引:1,他引:0

It is widely thought that viscosity plays an important rolein the perception of fats. Rats were conditioned to avoid oilsuspensions by treating them with lithium chloride after theyhad ingested the oil suspension. Control rats received the samelithium chloride injection after they drank vehicle. Three,chemically different, oils were examined, triglyceride oil,silicone oil and mineral oil. Rats trained to avoid a 4% aqueoussuspension of one of these oils, reliably avoided suspensionscontaining any of the other oils, although the rats could discriminatebetween the oils. The viscosity of the oil suspension was slightlygreater than the viscosity of the vehicle alone. However, ratstrained to avoid an oil suspension, did not avoid a fluid havinga viscosity similar to that of the oil suspension. In orderto assess the possibility that rats could sense the viscosityof the oil separately from that of the vehicle, rats were testedfor their aversion to oils having viscosities much higher andmuch lower than that of the oil they were trained to avoid.Rats trained to avoid triolein having a viscosity of 67 cp,reliably avoided silicone oils having viscosities of 5 and 203cp. However, rats trained to avoid oil did not avoid an oil-freefluid having a viscosity of 22–29 cp. A final experimentexamined whether the use of viscous or non-viscous vehiclesinfluenced the conditioned aversion. No significant effect ofvehicle viscosity appeared. Thus, chemically-diverse oils areperceived, by rats, to have some perceptible attribute in common.It is proposed that this common physical attribute is boundarylubrication rather than viscosity.  相似文献

Intact sexually mature New Zealand White male rabbits, raised under natural lighting and temperature conditions, were isolated and housed in air-conditioned quarters, in a 12-h light:12-h dark cycle. Push-pull cannulae were implanted towards the tuberal region of the hypothalamus, and animals were perfused with modified Krebs'-Ringer phosphate medium for an average period of 4 h. Most rabbits were repetitively perfused over an average period of 3.7 months. Perfusions were grouped into seasonal periods of about 40 or 80 days through the entire 1-year cycle: (A) 23 November to 31 December (winter solstice period: N = 6), (B) 1 January to 23 March (winter: N = 10), (C) 24 March to 13 June (spring: N = 9), (D) 14 June to 23 July (summer solstice period: N = 23), (E) 24 July to 13 October (late summer-early fall: N = 7) and (F) 14 October to 22 November (fall: N = 4). Maximal and minimal values of mean release, mean amplitude and mean frequency from every animal in each block were obtained. In the summer solstice group (D), maximal mean (+/- s.e.) LHRH release levels were significantly greater (14.42 +/- 6.62 pg/10 min) than for all groups (A, 0.76 +/- 0.27; B, 1.59 +/- 0.39; C, 1.34 +/- 0.22; E, 1.33 +/- 0.33; and F, 1.18 +/- 0.11) while during the winter solstice period (A), minimal mean LHRH release levels (0.48 +/- 0.04 pg/10 min) were significantly lower than in all other groups (B, 1.43 +/- 0.41; C, 0.96 +/- 0.11; D, 7.25 +/- 4.12; E, 1.18 +/- 0.37; and F, 1.18 +/- 0.11). Maximal values were highest during the summer solstice period while the minimal values were lowest during the winter solstice period. The amplitude and frequency of the LHRH pulses showed changes similar to those observed with the mean LHRH release. For 1 year, an estimated left testis weight was measured in 4-7 rabbits every 2-5 weeks. The percentage estimated testis weight peaked in early August and reached minimal levels during the winter months. These data demonstrate that the rabbit LHRH neural apparatus is very sensitive to seasonal influences although animals remained in a fixed photoperiod during the entire duration of the experiment.  相似文献

An endemic area of sporotrichosis is described in the Lake of Ayarza District, South Guatemala, where 53 patients have been observed within 3 years. In 45.3% of the cases, the infection appeared after handling fish. The disease was more frequently observed in man (83%) and in patients less than 30 years of age. The most frequent clinical type was the ascending lymphocutaneous sporotrichosis of the limbs. Some cases of ulcerative or verrucous lesions were seen. Almost all the patients cured rapidly either by potassium iodide (46 patients) or spontaneously, or after application of local heat (4 patients). Intradermal tests were performed in healthy population in the endemic area and, for comparison, in Guatemala City. Whole yeast cell antigens of Sporothrix schenckii and Ceratocystis stenoceras were used in these tests. Skin tests to both antigens were more frequently positive in the endemic area; the highest frequency was obtained with the antigen of C. stenoceras. A serological study was performed in 26 patients. The fluorescent antibody staining technique was more sensitive than yeast cell and latex particles agglutinations. C. stenoceras was isolated from bark of some trees, especially Eucalyptus, in the environment, but not S. schenckii.  相似文献

The pea aphid, Acyrthosiphon pisum Harris, is able to colonize various habitats and form genetically distinct biotypes worldwide. In Chile, few genotypes are dominating in space and time and are qualified as “superclones”. Here, we compared in the laboratory different biological features of the most common clones of A. pisum collected from pea (Ap1) and alfalfa (Ap2) fields, in order to gain some insight on their differential ability to colonize broad bean plants. We also studied their probing behavior, profiled their proteome and characterized their community of endosymbionts. We found that the Ap1 genotype performed better on Vicia faba and realized more salivation events in phloem than Ap2. In addition, Ap1 presented a higher prevalence of the endosymbiont Serratia symbiotica. Among the total of 40 proteins that were differentially expressed, 14 and 26 were upregulated in Ap1 and Ap2 genotypes, respectively. The symbionin from Buchnera aphidicola was found to be upregulated in Ap1. A field experiment showed that both genotypes were able to colonize wild legumes, with Ap1 reproducing better on Vicia nigricans than Ap2. However, Ap2 exhibited higher reproduction in the other three wild legumes, suggesting higher invasiveness capacity on wild plants. Variation in the ability to colonize, feeding behavior and the putative involvement of differentially regulated proteins between Ap1 and Ap2 are discussed in relation to their respective endosymbiotic composition, nutritional lifestyle and consequences on their “superclone” status.  相似文献

The amino acid substitution or post-translational modification of a cytosolic protein can cause unpredictable changes to its electrophoretic mobility during SDS-PAGE. This type of "gel shifting" has perplexed biochemists and biologists for decades. We identify a mechanism for "gel shifting" that predominates among a set of ALS (amyotrophic lateral sclerosis) mutant hSOD1 (superoxide dismutase) proteins, post-translationally modified hSOD1 proteins, and homologous SOD1 proteins from different organisms. By first comparing how 39 amino acid substitutions throughout hSOD1 affected SDS-PAGE migration, we found that substitutions that caused gel shifting occurred within a single polyacidic domain (residues ~80-101), and were nonisoelectric. Substitutions that decreased the net negative charge of domain 80-101 increased migration; only one substitution increased net negative charge and slowed migration. Capillary electrophoresis, circular dichroism, and size exclusion chromatography demonstrated that amino acid substitutions increase migration during SDS-PAGE by promoting the binding of three to four additional SDS molecules, without significantly altering the secondary structure or Stokes radius of hSOD1-SDS complexes. The high negative charge of domain 80-101 is required for SOD1 gel shifting: neutralizing the polyacidic domain (via chimeric mouse-human SOD1 fusion proteins) inhibited amino acid substitutions from causing gel shifting. These results demonstrate that the pattern of gel shifting for mutant cytosolic proteins can be used to: (i) identify domains in the primary structure that control interactions between denatured cytosolic proteins and SDS and (ii) identify a predominant chemical mechanism for the interaction (e.g., hydrophobic vs. electrostatic).  相似文献

The genetic background of the European Mesolithic and the extent of population replacement during the Neolithic [1-10] is poorly understood, both due to the scarcity of human remains from that period [11-18] and the inherent methodological difficulties of ancient DNA research. However, advances in sequencing technologies are both increasing data yields and providing supporting evidence for data authenticity, such as nucleotide misincorporation patterns [19-22]. We use these methods to characterize both the mitochondrial DNA genome and generate shotgun genomic data from two exceptionally well-preserved 7,000-year-old Mesolithic individuals from La Bra?a-Arintero site in León (Northwestern Spain) [23]. The mitochondria of both individuals are assigned to U5b2c1, a haplotype common among the small number of other previously studied Mesolithic individuals from Northern and Central Europe. This suggests a remarkable genetic uniformity and little phylogeographic structure over a large geographic area of the pre-Neolithic populations. Using Approximate Bayesian Computation, a model of genetic continuity from Mesolithic to Neolithic populations is poorly supported. Furthermore, analyses of 1.34% and 0.53% of their nuclear genomes, containing about 50,000 and 20,000 ancestry informative SNPs, respectively, show that these two Mesolithic individuals are not related to current populations from either the Iberian Peninsula or Southern Europe.  相似文献

The centromere is a repeat-rich structure essential for chromosome segregation; with the long-term aim of understanding centromere structure and function, we set out to identify cotton centromere sequences. To isolate centromere-associated sequences from cotton, (Gossypium hirsutum) we surveyed tandem and dispersed repetitive DNA in the genus. Centromere-associated elements in other plants include tandem repeats and, in some cases, centromere-specific retroelements. Examination of cotton genomic survey sequences for tandem repeats yielded sequences that did not localize to the centromere. However, among the repetitive sequences we also identified a gypsy-like LTR retrotransposon (Centromere Retroelement Gossypium, CRG) that localizes to the centromere region of all chromosomes in domestic upland cotton, Gossypium hirsutum, the major commercially grown cotton. The location of the functional centromere was confirmed by immunostaining with antiserum to the centromere-specific histone CENH3, which co-localizes with CRG hybridization on metaphase mitotic chromosomes. G. hirsutum is an allotetraploid composed of A and D genomes and CRG is also present in the centromere regions of other AD cotton species. Furthermore, FISH and genomic dot blot hybridization revealed that CRG is found in D-genome diploid cotton species, but not in A-genome diploid species, indicating that this retroelement may have invaded the A-genome centromeres during allopolyploid formation and amplified during evolutionary history. CRG is also found in other diploid Gossypium species, including B and E2 genome species, but not in the C, E1, F, and G genome species tested. Isolation of this centromere-specific retrotransposon from Gossypium provides a probe for further understanding of centromere structure, and a tool for future engineering of centromere mini-chromosomes in this important crop species.  相似文献

目的:探讨等离子注入技术(Plasma immersion ion implantation,PⅢ)对膨体聚四氟乙烯(ePTFE)膜改性后表面性能以及对细菌生存状态的影响.方法:采用氧等离子注入技术(PⅢ)对ePTFE膜表面处理,获得改性ePTFE膜,利用扫描电镜(SEM)观察改性ePTFE膜和未处理膜的表面形貌,体外培养变形链球菌,观察改性前后各组膜作体外细菌生存状态变化.结果:与未处理ePTFE膜相比,改性ePTFE膜表面形貌、亲水性发生改变.荧光染色后观察长脉冲组表面的细菌最少;短脉冲组膜表面粘附的细菌密集,形成生物膜;空白组可见少量死菌和活菌粘附于ePTFE膜上.结论:长脉冲PⅢ改性ePTFE膜后有良好的抗细菌粘附能力.  相似文献
