Spontaneous bleeding of sugar-rich sap from cambial-deep incisions in the bark of trunks was demonstrated for Eucalyptus globulus and other eucalypts across a range of localities and seasonal conditions in south-west Australia. High levels of sucrose and raffinose (up to 31% w/v total sugars) were present in the exudates, and upward and downward gradients in exudate sugar concentrations were recorded between samples obtained at different heights up trunks of E. globulus. The data indicated a phloem origin for the exudates, with source:sink pressure gradients driving translocation. Concentration ratios of sugars to amino acids were consistently lower in exudate from upper (distal) than basal regions of trunks, suggesting preferential partitioning of nitrogen upwards towards the trunk apex. A comparison of phloem and xylem sap composition from one plantation over a season showed nitrate in xylem but not phloem and substantial amounts of sodium, and high concentrations of chloride and sulphate relative to phosphate in xylem and phloem. Phloem sap sampled across a range of 29 contrasting plantations of E. globulus at peak stress (autumn) showed great inter-site variability in concentrations of amino acids, sulphur, sodium and certain trace elements and in C:N and Na:K ratios of sap. Carbon isotope ratios (δ13C) were strongly correlated with sugar concentrations of the sap samples from these and other plantations. Use of sap compositional attributes of phloem and δ13C values of translocated carbon is suggested for assessing the current nutritional condition and water status of E. globulus plantings. Summary The distribution and time course of development of transfer cells in the hypocotyl region of lettuce (Lactuca sativa L.) and groundsel (Senecio vulgaris L.) are examined by light microscopy of serial sections through a sequence of ages of hypocotyls. Investments of xylem transfer cells occur in departing traces to the cotyledons and, later, in the traces to foliage leaves; phloem transfer cells are widely distributed but particularly prominent in those bands of protophloem in the plumule vasculature which lie alongside xylem of the cotyledonary traces. Both classes of transfer cell are well endowed with wall ingrowths before differentiation of xylem and perforation of stomata occurs in the plumule. Autoradiographic evidence is obtained of a transport pathway from cotyledonary trace xylem elements to xylem transfer cell to plumule, and analyses of xylem sap collected from above or below the zones of transfer cells in the hypocotyl show that certain materials can be removed from the xylem sap by transfer cells as it moves towards the cotyledons. From these findings it is concluded that the seedling transfer cells play an important role in nutrition of the young plumule, particularly before the latter has become adequately connected with the vascular systems of cotyledons and root.Experiments on the experimental modification of transfer cell development in the hypocotyl suggest that both photosynthetic fixation of carbon dioxide and a transpirational loss of water by a cotyledon must take place before the presumptive xylem transfer cells in its traces can develop normal sets of wall ingrowths.Discussion is extended to the general role of transfer cells in the nodal regions of stems. Possible functions envisaged are, the general nutrition of young tissues of the apical region, the abstraction of assimilates for local storage, the transfer of assimilates to axillary buds released from apical dominance, and the interchange of assimilates between adjacent vascular traces running through the node.  相似文献

Asticcacaulis biprosthecum sp. n. is a gram-negative aerobic heterotroph which undergoes a dimorphic life cycle typical of bacteria of the caulobacter group. Prosthecae of these cells are shown to be structurally homologous with prosthecae of two strains ofCaulobacter crescentus. Summary Bleeding from phloem of cut distal tips of attached fruits was demonstrated in the genera Spartium, Genista, Lupinus and Jacksonia. Bleeding occurred over a 2–25 min period enabling 0.5–10 l of sap to be collected from a fruit. A detailed study of Lupinus albus L. showed that exudation rate declined exponentially after cutting, but without any change with time in solute levels in exudate. Bleeding resumed at its initial rate and solute concentration on recutting the fruit tip.Phloem exudates had a high pH (7.8-8.0), a sucrose content of 100–210 mg ml-1 but only traces of monosaccharides. Surrounding pod tissues contained only 15–35 mg ml-1 of sugars (tissue water basis) more than two thirds of this monosaccharide. Amino compounds were present in phloem exudates at 8–28 mg ml-1, asparagine and glutamine predominating but a wide spectrum of other amino acids being also present. No significant differences in levels of organic solutes were observed in phloem exudates collected from tips of attached versus detached fruits, from phloem exudates collected from fruit tips versus pedicels, or from basal versus distal ends of a detached fruit.Potassium was the major cation (1.5–2.2 mg ml-1) of the phloem exudate, Ca2+ was at a much lower level than either Mg2+ or Na+. Trace element levels in phloem exudates appeared to be influenced by availability to the plant from the rooting medium. Nitrate was absent though detectable in non-vascular tissues of the shoot. 14C- labelled assimilates were detected in exudates of L. albus one hour after feeding a source leaf 14CO2; sucrose, organic acids and certain amino compounds achieved high specific labelling. 14CO2 feeding studies coupled with the phloem bleeding technique revealed highly specific source-sink relationships between foliar organs and fruits of the primary inflorescence.  相似文献

以河北山前平原区秸秆还田条件下小麦-玉米轮作体系为研究对象,设置农民习惯、高产高效、再高产和再高产高效4个模式,通过定位试验探讨各栽培模式对3个轮作周期作物产量、土壤硝态氮累积量及氮平衡的影响.结果表明: 小麦、玉米产量均以再高产模式最高,高产高效和再高产高效模式次之,均显著高于农民习惯模式;小麦季和玉米季氮肥利用效率(PFP)均以高产高效模式最高,显著高于其他模式;0~400 cm土体硝态氮累积量在 768.4~1133.3 kg·hm-2之间,其中80%~85%累积在根下90~400 cm土层;4种模式的土壤硝态氮均有明显向下淋移现象,120~150 cm和270~330 cm处均出现了累积峰,以270~330 cm土层硝态氮累积量最大;高产高效模式的土壤硝态氮含量整体水平均低于其他模式,浓度基本维持在30 mg·kg-1以下,在一定程度上能有效缓解环境压力;冬小麦季0~90 cm土体氮素盈余量均小于夏玉米季,并以高产高效模式的氮素表观损失量最低,显著低于其他模式.综合考虑产量、氮肥利用效率、硝态氮累积和氮平衡,以高产高效模式表现最优,但还有一定的提升空间.  相似文献

Principal component analysis (PCA) was used to analyse the behaviour of a chromatographic separation as its scale increased. Three 4.6 mm diameter columns identical in every respect except for column length (25, 15 and 5 cm), were used to generate the data from a test system based on the reversed-phase HPLC separation of crude erythromycin on a polystyrene matrix (PLRP 1000) having a particle diameter of 8 mu;m and a pore diameter of 100 nm. The species were separated with an isocratic solvent composed of 45/55 acetonitrile/water at about pH 7. An experimental design technique was used to investigate the effects of four process variables (load volume, load concentration, temperature and pH of buffer) on the chromatogram shapes. Following appropriate pre-processing of the chromatographic data, subsets of critical chromatograms were selected which sufficiently characterised the entire data set. From this subset, the corresponding runs were performed on the different sized columns and principal component models were generated for each. At 5 and 15 cm a single principal component was sufficient to characterise all the variance in the chromatograms which the range of process variables introduced, but at 25 cm two principal components were required, particularly to characterise the chromatograms with small loads. Excellent correlations were observed between the first principal components at the three scales. The possibility of predicting the separations on the 25 cm column from an analysis of the separations observed at 5 cm was investigated. The study revealed that good predictions could be made at high loads (>92%) , but the model was not effective at low loads because of the need to incorporate a second principal component which was not defined by the range of variables applied to the 5 cm column.  相似文献

本文根据1987年在内蒙古东北部低山丘陵区和平原区草原的调查资料,应用频数分布比较法研究了达乌尔黄鼠的空间分布型。结果表明:在抽样单位(样方)大小为1公项(1OO米×100米)时,4个样地内达乌尔黄鼠交尾期和幼鼠分居期的空间分布型均属poisson分布(随机分布)。环境与密度制约因素的影响不明显。并经回归分析得到:种群密度(m)与零样方频率(Po)和有鼠样方频率(Px )的曲线关系分别为m=4.5451-0.96721nPo和m=4.5451-0.96721n(1-Px)。经X2检验与poisson分布理论曲线基本一致(x2《x2 0.05,P>0.05)。为达乌尔黄鼠种群密度估计提供了理论依据.  相似文献

动物线粒体DNA控制区是线粒体基因组复制与基因表达的最主要的调控区.采用杂交和测序的方法对草鱼线粒体DNA控制区进行定位、克隆并测定了控制区及其旁侧的tRNAPhe、rRNAPro和rRNAThr三个基因的序列,与多种脊椎动物的相应序列进行了比较,并进行了结构分析.草鱼线粒体控制区全长927bp,含有与酵母和爪蟾线粒体启动子相似的序列,其CSBⅠ、CSBⅡ和CSBⅢ序列与其他几种动物的CSB比较相当保守,TAS与其回文基序可形成稳定的茎环结构,成为H-链复制的终止信号.草鱼线粒体tRNAPhe、tRNAPro和tRNAThr可折叠成三叶草形二级结构,其基因具有许多不同于细胞质tRNA基因的结构特点,可能反映了线粒体tRNA与线粒体核糖体具有不寻常的作用方式  相似文献
